Abstract:The application of immobilized lipase in the enzymatic production of biodiesel has shown numerous advantages. In this study, surface of Polyacrylonitrile (PAN) hollow membrane was first modified using nitrile-click chemistry in order to fit for interaction with enzyme proteins. Then sodium alginate (SA) was introduced and the membrane was post-treated by CaCl2. When the prepared PAN-PEI-SA-CaCl2 was used for lipase immobilization, the protein loading was 36.90 mg/g, and the enzyme activity reached up to 54.47 U/g, which was 2.5 times as much as that of Novozym® 435. As a result, the constructed immobilized lipase obtained a maximum biodiesel yield of 78.5%, which was 2.4 times that of the Novozym® 435 in transesterification reactions. Moreover, the biodiesel yield decreased by only 11% after the immobilized enzyme was continuously used for 20 times. This study exhibits that this technic has broad application prospects in the field of conversion of biomass resources.
Lipase nanogel for biodiesel production

Abstract:The production of biodiesel from plant oil can be efficiently catalyzed by lipase in a water-containing system without an organic solvent. However, this process is hindered by methanol induced deactivation of lipase. Lipase nanogel was prepared and characterized. The half-life time of native lipase was 50 min at 50°C while almost no visible loss of lipase activity was observed in the 500 min for the nanogel. After incubation in 40% methanol aqueous solution, lipase nanogel preserved about 80% of its hydrolytic activity while native lipase reserved only 10%. All these are of great interest for further exploration of the potential application of lipase nanogel for the production of biodiesel. Abstract:Lipase is the key enzyme for catalyzing triglyceride hydrolysis in vivo, and lipase inhibitors have been used in the management of obesity. We present the first report on the use of lipase-adsorbed halloysite nanotubes as an efficient medium for the selective enrichment of lipase inhibitors from natural products. A simple and rapid approach was proposed to fabricate lipase-adsorbed nanotubes through electrostatic interaction. Results showed that more than 85% lipase was adsorbed into nanotubes in 90 min, and approximately 80% of the catalytic activity was maintained compared with free lipase. The specificity and reproducibility of the proposed approach were validated by screening a known lipase inhibitor (i.e., orlistat) from a mixture that contains active and inactive compounds. Moreover, we applied this approach with high performance liquid chromatography-mass spectrometry technique to screen lipase inhibitors from the Magnoliae cortex extract, a medicinal plant used for treating obesity. Two novel biphenyl-type natural lipase inhibitors magnotriol A and magnaldehyde B were identified, and their IC50 values were determined as 213.03 and 96.96 mu M, respectively. The ligand-enzyme interactions of magnaldehyde B were further investigated by molecular docking. Our findings proved that enzyme-adsorbed nanotube could be used as a feasible and selective affinity medium for the rapid screening of enzyme inhibitors from complex mixtures. (C) 2015 Elsevier B.V. Enhanced Performance of Rhizopus Oryzae Lipase by Reasonable Immobilization on Magnetic Nanoparticles and Its Application in Synthesis 1,3-Diacyglycerol

Abstract:Nano-sized Fe 3 O 4 was synthesized by chemical co-precipitation and subsequently modified with 3-aminopropyltriethoxysilane (APTES) and glutaraldehyde to introduce aldehyde group on its surface. With the help of “interface activation” by adding sucrose esters-11 as surfactant, lipase from Rhizopus oryzae was successfully immobilized onto the carrier with great enhancement of activity. The hydrolysis activity of immobilized enzyme were 9.16 times and 31.6 times of free enzyme when p-nitrophenol butyrate and p-nitrophenol palmitate were used as substrates. The thermo-stability of immobilized enzyme was also enhanced compared to free enzyme. The immobilized enzyme was successfully applied in synthesis of 1,3-diacyglycerols (1,3-DAG). The specific esterification activity of immobilized enzyme was about 1.5 times of the free enzyme. The immobilized enzyme showed good region-selectivity towards 1,3-diacyglycerols and retained nearly 80% of its activity after reused for 60 times, revealing a good industrial application prospect.

Abstract:The present work showed that Candida rugosa lipase, which is inactive in anhydrous dimethyl sulfoxide (DMSO), has been granted its original catalytic activity and greatly enhanced stability when encapsulated into a polyacrylamide nanogel. The molecular simulation and structural analysis suggested that the polyacrylamide nanogel shielded the extraction of essential water and maintained the native configuration of encapsulated lipase in anhydrous DMSO at an elevated temperature. The electron and fluorescence microscopy showed that the lipase nanogel would be well dispersed in anhydrous DMSO where its native counterpart aggregated. The encapsulated lipase behaved as a stable catalyst for transesterification between dextran and vinyl decanoate in anhydrous DMSO at 60 degrees C for 240 h and yielded a dextran-based polymeric surfactant with regioselectivity toward the C-2 hydroxyl group in the glucopyranosyl unit of dextran. All these indicated a high potential of enzyme nanogel for nonaqueous biocatalysis.
Design and synthesis of lipase nanogel with interpenetrating polymer networks for enhanced catalysis: Molecular simulation and experimental validation

Abstract:A temperature-responsive lipase nanogel (denoted as CRL-IPN nanogel), in which lipase is encapsulated into an interpenetrating polymer matrix formed by polyacrylamide and poly(N-isopropylacrylamide) (PNIPAAm) has been designed and synthesized for an enhanced stability and activity in both aqueous and non-polar organic solvents. A three-step method, including acryloylation, polymerization with acrylamide and sequential polymerization with N-isopropylacrylamide, was established to fabricate enzyme nanogel with temperature-sensitive interpenetrating polymer network. It has been shown by an all-atom molecular dynamics simulation that above mentioned polymer matrix forms a more hydrophobic environment, as compared to that obtained with sole polyacrylamide, because of the penetration of N-isopropylacrylamide into the polymer acrylamide network via hydrogen bonding, which is further confirmed by the fluorescence spectrum. This favours the uptake of hydrophobic substrates and thus the overall rate of enzymatic catalysis. The enhanced stability and catalytic performance of this novel lipase nanogel in aqueous and non-polar organic solvent were demonstrated by using hydrolysis reaction of p-NPP in aqueous and esterification reaction of ibuprofen in isooctane. In aqueous solution, the residual activity of CRL-IPN nanogel maintains its 70% activity at 60°C after 4h, compared with that free lipase only has 30% at the same condition. In addition, the CRL-IPN nanogel can be reused for 10 cycles with no loss of its activity. In isooctane, CRL-IPN nanogel gave a 33% yield of esterification of ibuprofen, in comparison to 22% using free lipase and less than 5% using lipase encapsulated in a polyacrylamide matrix. The enhanced stability and activity make this CRL-IPN nanogel promising for enzymatic catalysis in non-polar solvents.
Activation and stabilization of a lipase nanogel using GMA for acryloylation

Abstract:A lipase nanogel has been prepared by aqueous in situ polymerization, with initial acryloylation to introduce vinyl groups onto the lipase surface for subsequent polymerization with acrylamide monomers. Activation of lipase was observed using glycidyl methacrylate (GMA) as the acryloylation agent, leading to an increase of activity yield from 78 +/- 4% (obtained using NAS for acryloylation) to 122 +/- 14%. The acryloylation ratio was also improved, from 8 +/- 5% to 20 +/- 8%, which favored the subsequent polymerization. In addition, the overall activity yield of the lipase nanogel was increased from 44 +/- 3% to 105 +/- 2%. The lipase nanogel prepared with GMA for acryloylation displayed a lower K-m and a higher k(cat) in comparison to its native counterpart, as well as the nanogel obtained using NAS for acryloylation. The half-life of the lipase nanogel at 60 degrees C was extended from 1.63 h (using NAS for acryloylation) to 5.61 h, while the half-life of the native lipase was 1.41 h. Molecular dynamics simulations and experiments further suggested that the activation effect was due in part to interactions between the lipase and the GMA, which reinforced the 'open' configuration of the lipase, thereby facilitating mass transport to and from the modified lipase in the free and encapsulated forms. The above mentioned activation effect of the enzyme nanogel illustrated the power of chemical modification by tailored design of the nanostructure of the enzyme catalyst, potentially expanding the applications of enzyme catalysis.

Abstract:Alternative to the traditionally independent production of lipase, chemical synthesis of nano-carriers, and then preparing nanoimmobilized enzymes, we exploit a yeast genetically programmed virus biomimetic lipase nanoreactor in a sustainable manner. The nanoreactor biogenesis process integrated lipase production, protein component (coat-protein subunit and scaffold protein) production, self-assembly of protein components, and the encapsulation of lipase into protein nanocages using a simple process. It included overexpression of nanocage components, coat-protein subunits, and fused lipase-scaffold proteins and subsequent spontaneous self-assembly and encapsulation based on the specific interaction between the coat-protein subunit and the scaffold protein fused in the target lipase enzyme. The genetically programmable lipase nanoreactor showed improved stability under various harsh conditions, and was validated in fatty acid methyl ester synthesis with 86% yield at a high concentration of waste cooking oil (200 mM), which demonstrates the robustness and feasibility of the lipase nanoreactor in biodiesel production.
Efficient Biocatalyst by Encapsulating Lipase into Nanoporous Gold

Abstract:Lipases are one of the most important biocatalysts for biotechnological applications. Immobilization is an efficient method to increase the stability and reusability of lipases. In this study, nanoporous gold (NPG), a new kind of nanoporous material with tunable porosity and excellent biocompatibility, was employed as an effective support for lipase immobilization. The pore size of NPG and adsorption time played key roles in the construction of lipase-NPG biocomposites. The morphology and composition of NPG before and after lipase loading are verified using a scanning electron microscope, equipped with an energy-dispersive X-ray spectrometer. The resulting lipase-NPG biocomposites exhibited excellent catalytic activity and remarkable reusability. The catalytic activity of the lipase-NPG biocomposite with a pore size of 35 nm had no decrease after ten recycles. Besides, the lipase-NPG biocomposite exhibited high catalytic activity in a broader pH range and higher temperature than that of free lipase. In addition, the leaching of lipase from NPG could be prevented by matching the protein’s diameter and pore size. Thus, the encapsulation of enzymes within NPG is quite useful for establishing new functions and will have wide applications for different chemical processes.

Abstract:A substrate or polyethyleneglycol (PEG) imprinted lipase nanogel displayed increased apparent activity in organic solvents by 2.5-4.7 folds, compared to native lipase. It enabled a one-step synthesis of chloramphenicol palmitate with a yield of similar to 99% and purity of similar to 99%, indicating that the imprinted lipase nanogel is an appealing catalyst in organic media.

Abstract:Enzyme-immobilized membrane bioreactors combine biocatalysis with separation, and thus have generated interest among applied researchers. Despite the advantages of enzyme-immobilized membrane bioreactors, some issues associated with the membrane and operation parameters still remain to be addressed in order to achieve scale-up of such systems. We report the fabrication of a biphasic lipase-immobilized nanofiber membrane bioreactor with high catalytic performance. A poly(acrylonitrile-co-acrylic acid) non-woven nanofiber membrane was prepared by electrospinning and lipase from Candida rugosa was covalently coupled to it via an activation process by N-(3-dimethylaminopropyl)-N'-ethyl-carbodiimide hydrochloride/N-hydroxy-succinimide. The influences of membrane diameters and operation variables on bioreactor efficiency were studied using the catalytic hydrolysis of olive oil as the model reaction. Furthermore, a pseudo first order model contributed to the evaluation of lipase performance in a more standardized way. A bioreactor activity of 1.85 x 10(4) U g(-1) was obtained under optimum operation conditions; also, the catalytic system exhibited good operational and recycling stability.

Abstract:The present work depicts the development of stable nanogels in an aqueous medium that were exploited for efficient surface-active lipase-catalyzed hydrolysis of water-insoluble substrates. Surfactant-coated gel nanoparticles (neutral NG1, anionic NG2, and cationic NG3) were prepared from peptide amphiphilic hydrogelator (G1, G2, and G3, respectively) at different hydrophilic and lipophilic balance (HLB). Chromobacterium viscosum (CV) lipase activity towards hydrolysis of water-insoluble substrates (p-nitrophyenyl-n-alkanoates (C4-C10)) in the presence of nanogels got remarkably improved by ~1.7-8.0 fold in comparison to that in aqueous buffer and other self-aggregates. An increase in hydrophobicity of the substrate led to a notable improvement in lipase activity in the hydrophilic domain (HLB>8.0) of nanogels. The micro-heterogeneous interface of small-sized (10-65 nm) nanogel was found to be an appropriate scaffold for immobilizing surface-active lipase to exhibit superior catalytic efficiency. Concurrently, the flexible conformation of lipase immobilized in nanogels was reflected in its secondary structure having the highest α-helix content from the circular dichroism spectra.
Nanofibrous Poly(acrylonitrile-Co-maleic Acid) Membranes Functionalized with Gelatin and Chitosan for Lipase Immobilization.

Abstract:Nanofibrous membranes with an average diameter of 100 and 180 nm were fabricated from poly(acrylonitrile-co-maleic acid) (PANCMA) by the electrospinning process. These nanofibrous membranes contain reactive groups which can be used to covalently immobilize biomacromolecules. Two natural macromolecules, chitosan and gelatin, were tethered on these nanofibrous membranes to fabricate dual-layer biomimetic supports for enzyme immobilization in the presence of 1-ethyl-3-(dimethyl-aminopropyl) carbodiimide hydrochloride (EDC)/N-hydroxyl succinimide (NHS). Lipase from Candida rugosa was then immobilized on these dual-layer biomimetic supports using glutaraldehyde (GA), and on the nascent PANCMA fibrous membrane using EDC/NHS as coupling agent, respectively. The properties of the immobilized lipases were assayed. It was found that there is an increase of the activity retention of the immobilized lipase on the chitosan-modified nanofibrous membrane (45.6+/-1.8%) and on the gelatin-modified one (49.7+/-1.8%), compared to that on the nascent one (37.6+/-1.8%). The kinetic parameters of the free and immobilized lipases, K(m) and V(max), were also assayed. In comparison with the immobilized lipase on the nascent nanofibrous membrane, there is an increase of the V(max) value for the immobilized lipases on the chitosan- and gelatin-modified nanofibrous membranes. Results also indicate that the pH and thermal stabilities of lipases increase upon immobilization. The residual activities of the immobilized lipases are 55% on the chitosan-modified nanofibrous membrane and 60% on the gelatin-modified one, after 10 uses.
Combined Strategy for Preparation of A Bioimprinted Geotrichum Sp Lipase Biocatalyst Effective in Non-Aqueous Media

Abstract:Geotrichum sp. lipase with enhanced activity and operational stability was prepared for use in non-aqueous media. A combined strategy comprising bioimprinting with dual imprint molecules and a co-solvent coupled to pH tuning, KCl salt activation, lecithin coating and immobilization on macroporous resin effectively enhanced the activity and operational stability of Geotrichum sp. lipase. The modified lipase exhibited 18.4-fold enhanced esterification activity towards methyl oleate synthesis, and retained 90% activity following repeated use in 10 cycles. The combined strategy exhibited a significant synergistic effect and was suitable for lipase modification, dramatically enhancing the enzyme activity and operational stability. This approach is applicable to the preparation of other enzyme biocatalysts, since the methods are effective for upgrading crude enzyme to a refined product with high activity and stability for use in non-aqueous media.

Abstract:Lipase B from Candida antarctica (CALB) plays a prominent role as a biocatalyst in several industries, especially for biphasic conversion of functional lipids. Herein, an amphiphilic Janus halloysite nanosheet (JHNS) was fabricated and employed simultaneously as a solid surfactant for stabilizing Pickering emulsion and as a carrier for immobilizing CALB, with the aim to realize highly efficient biphasic bioconversion. The obtained JHNS could stabilize Pickering emulsion for at least 7 days. Immobilization of CALB on JHNS improved the substrate affinity, catalytic efficiency, thermal stability, and alkaline tolerance of the enzyme. Moreover, JHNS-based immobilized CALB was exploited as a biocatalytic platform for the conversion of retinyl acetate, with almost twice increase in conversion efficiency. Taken together, the JHNS-based immobilized CALB paves the way for the design of efficient biphasic conversion system for the production of added-value lipids.
Enzyme-Nanoporous Gold Biocomposite: Excellent Biocatalyst with Improved Biocatalytic Performance and Stability

Abstract:Background: Applications involving biomolecules, such as enzymes, antibodies, and other proteins as well as whole cells, are often hampered by their unstable nature at extremely high temperature and in organic solvents.Methodology/Principal Findings: We constructed enzyme-NPG biocomposites by assembling various enzymes onto the surface of nanoporous gold (NPG), which showed much enhanced biocatalytic performance and stability. Various enzymes with different molecular sizes were successfully tethered onto NPG, and the loadings were 3.6, 3.1 and 0.8 mg g(-1) for lipase, catalase and horseradish peroxidase, respectively. The enzyme-NPG biocomposites exhibited remarkable catalytic activities which were fully comparable to those of free enzymes. They also presented enhanced stability, with 74, 78 and 53% of enzymatic activity retained after 20 successive batch reactions. Moreover, these novel biocomposites possessed significantly enhanced reaction durability under various thermal and in organic solvent systems. In a sample transesterification reaction, a high conversion rate was readily achieved by using the lipase-NPG biocomposite.Conclusion/Significance: These nano-biocomposite materials hold great potential in applications such as biosensing, molecular electronics, catalysis, and controlled delivery.
Robust biocatalytic single enzyme nanogels by versatile strategies

Abstract:Biocatalysis has been considered as an important and cost-effective technology for the manufacture of fine chemicals, synthesis of pharmaceuticals, biosensors, bioremediation, protein digestion in proteomic analysis, and biofuel cells. A facile in-situ aqueous polymerization procedure to fabricate single enzyme nanogels as robust nanometer scale biocatalysis was studied. Using versatile modification strategies, e.g., acryloylation of lysine group, introducing vinyl group to glutamic acid and asparagine, control of modification pH, active site protection by inhibitors, different single enzyme nanogels were successfully prepared. These nanogels showed greatly improved stability to temperature, organic solvent, and low pH solution without the compromise of catalytic capability. These nanogels were quite suitable to biocatalytic applications in harsh condition, which are constrained by small size or high mass-transfer rating reaction or exposed for extreme pH and temperature. Abstract:This study investigates the use of nanodiamonds (ND) as a promising carrier for enzyme immobilization and compares the effectiveness of immobilized and native enzymes. Three different enzyme types were tested, of which Rhizopus niveus lipase (RNL) exhibited the highest relative activity, up to 350 %. Under optimized conditions (1 h, pH 7.0, 40 °C), the immobilized ND-RNL showed a maximum specific activity of 0.765 U mg-1, significantly higher than native RNL (0.505 U mg-1). This study highlights a notable enhancement in immobilized lipase; furthermore, the enzyme can be recycled in the presence of a natural deep eutectic solvent (NADES), retaining 76 % of its initial activity. This aids in preserving the native conformation of the protein throughout the reusability process. A test on brine shrimp revealed that even at low concentrations, ND-RNL had minimal toxicity, indicating its low cytotoxicity. The in silico molecular dynamics simulations performed in this study offer valuable insights into the mechanism of interactions between RNL and ND, demonstrating that RNL immobilization onto NDs enhances its efficiency and stability. All told, these findings highlight the immense potential of ND-immobilized RNL as an excellent candidate for biological applications and showcase the promise of further research in this field.
