Study Heterogenous Expression and Activity Determination of Human Cytochrome P450 2B6

Abstract:OBJECTIVE To obtain an active recombinant human CYP2B6 in order to elucidate the role of this enzyme in drug metabolism.METHODS The Bac-to-Bac baculovirus expression system was used to express recombinant CYP2B6 with His-tag on the C-terminal.The His-tagged recombinant CYP2B6 expressed in Spodoptera frugiperda(Sf9) cells was detected using anti-His antibody.The enzyme activity was determined using bupropion as a probe substrate which was determinated by HPLC.RESULTS The expression of CYP2B6 was detected by Western blot analysis.The Km and Vmax with bupropion as substrate were(138±42)μmol·L-1 and(676 ± 21)pmol · min-1· mg-1 protein,respectively.CONCLUSION Active recombinant human CYP2B6 obtained using baculovirus expression system will facilitate further examination of the role of this enzyme in drug metabolism.
