Abstract:Objective To prepare monoclonal antibody (mAb) against M protein of dengue virus type 2 (DV2),which could be applied to the development of gold colloidal rapid diagnostic kit for the specific detection of dengue virus infection. Methods The prm cDNA was amplified with PCR from the plasmid containing the genomic-length cDNA of DV2 strain New Guinea C,and was inserted to the multi-cloning sites of plasmid pET-32a(+) vector and expressed with induction of IPTG. The recombinant protein was purified with Ni-NTA,while the immunogenicity of the recombinant protein was identified by Western blot assay with rabbit antiserum against dengue virus. Balb/c mice were immunized with the recombinant M protein. Splenocytes of the immunized mice were collected and fused with the mouse myeloma cell line Sp2/0 cells. The hybridoma cells secreting anti-M protein mAbs were cloned by limited dilution method. The affinity of the obtained mAbs was determined by indirect ELISA. The specificity of mAbs was tested by ELISA and Western blot analysis. Results From over 86 positive hybridomas secreting anti-M protein mAbs,a pair of hybridomas were screened out,designated as Ⅲ1A6 and Ⅲ3D2. Chromosome analysis revealed that the obtained hybridomas had the universal characteristics of the monoclonal hybridoma cells. The relative affinity constants of Ⅲ1A6 and Ⅲ3D2 mAb were both determined as 105. Conclusion A pair of high titer specific mAbs against DV2 M protein have been successfully prepared and primarily identified,which may be useful in the development of a rapid and convenient diagnostic kit for detection of dengue viral infection.
