Dynamic Change of Serum NGF Levels Induced by Oxaliplatin-Based Chemotherapy and Its Clinical Meanings

Abstract:Objective:To observe the dynamic change of circulating nerve growth factor(NGF)levels during the course of oxali- platin(L-OHP)-based chemotherapy and discuss the relationship between the serum level of NGF and neurotoxicity associated with L-OHP.Methods:The L-OHP-associated neurotoxieity was evaluated on 37 patients.The grade of neurotoxicity was determined ac- cording to the Bertheault-Cvitkovic criteria at different time points. The level of serum NGF was monitered by ELISA method before and after treatment.Correlation between the serum level of NGF and the severity of neurotoxicity was analyzed. Results: There was a de- creasing trend of NGF at 24 to 48 h just after the first cycle of L-OHP chemotherapy compared with the baseline value.A significant de- crease appeared 3-4 weeks later.The proportion of patients whose NGF level decreased more than 33.24%(the median value of de- crease)markedly increased accompanied with the increasing cycles of chemotherapy.Spearman correlation analysis showed a highly sig- nificant correlation between the decreasing extent of NGF and the severity of clinical neurotoxicity(r=0.522,P0.001).The decrea- sing extent of NGF did not significantly correlate with the final severity of neurotoxicity after multiple cycles of chemotherapy (P0.05).Conclusion:Our results indicate that NGF level decreased during the course of L-OHP chemotherapy.The decreasing extent of NGF correlated with the severity of neurotoxiciy.The decreases in the serum level of NGF may predict the short-term neurotoxiciy to a certain extent,but could not predict the long-term neurotoxicity of L-OHP.
