Effects of Carvedilol on Atrial Gap Junctional Remodeling after Chronic Left Ventricular Myocardial Infarction

Abstract:Objective: To investigate the effects of carvedilol on atrial gap junctional remodeling in a rabbit model of chronic left ventricular myocardial infarction(MI).Methods: Thirty rabbits were randomly divided into three groups: control group(n=10,thoracotomy,left anterior descending artery was not ligated) and MI(n=10,thoracotomy + MI,left anterior descending artery was ligated) group,and Carvedilol group(n=10).The rabbits in the carvedilol group received oral carvedilol [5 mg/(kg·d)] for eight weeks.The rabbits in the other two groups received a matching saline.Eight weeks after operation,distribution of atrial connexin 40(Cx40) and its protein level were studied by immunohistochemistry and Western blot.Results: In MI group,atrial Cx40 level of left atrial appendage decreased significantly than that in control group(P0.01).The distribution Cx40 was changed.The sideto-side junctions increased,while the end-to-end junctions decreased.Atrial Cx40 in MI group was highly heterogeneous.In the Carvedilol group,the atrial Cx40 level increased significantly than that in MI group(P0.05).Conclusion: Chronic left ventricular myocardial infarction can results in atrial Cx40 remodeling.Carvedilol can attenuate the Cx40 remodeling effectively.
