Abstract:To understand the impacts of different processing methods on the composition and effects of the herb Saussurea laniceps (SL), the present study report the first comparison of the chemical constituents of aqueous and ethanolic SL extracts using chromatographic analysis, and to compare their pharmacological effects in a mouse anti-inflammatory, anti-nociceptive model and an in vitro anti-oxidant test. Chemical comparison demonstrated that the types of chemicals in the two extracts were identical, but the contents of the main constituents in the aqueous extract were lower than those of the ethanolic extract. A transesterification of dicaffeoylquinic acids took place in the aqueous extract during boiling. As for pharmacological effects, oral administration of aqueous and ethanolic SL extracts significantly inhibited croton oil-induced mice ear edema, and significantly inhibited acetic acid-induced mice writhings, respectively. In the DPPH anti-oxidant activity test, the IC50 values were calculated as 409.6 mg/L and 523.4 mg/L for the ethanolic and aqueous extracts, respectively. The inhibitory effects of the ethanolic extract were more potent than those of the aqueous extract in all pharmacological tests, although there was no significant difference. This study suggests that the two preparations should be distinguished when used.

Abstract:Thirty-six naturally occurring compounds, including four C10-acetylenic glycosides and a lignan, were isolated from the whole plants of Saussurea cordifolia. Their structures were elucidated by means of spectroscopic and chemical methods to be 4,6-decadiyne-1-O-β-d-apiofuranosyl-(1→6)-β-d-glucopyranoside (1), 4,6-decadiyne-1-O-α-l-rhamnopyranosyl-(1→6)-β-d-glucopyranoside (2), (8E)-decaene-4, 6-diyn-1-O-α-l-rhamnopyranosyl-(1→6)-β-d-glucopyranoside (3), (8Z)-decaene-4,6-diyn-1-O-β-d-apiofuranosyl-(1→6)-β-d-glucopyranoside (4), and (2R,3S,4S)-4-(4-hydroxy-3-methoxybenzyl)-2-(5-hydroxy-3-methoxyphenyl)-3-(hydroxymethyl)-tetrahydrofuran-3-ol (5).
Comparison of the Anti-Inflammatory Active Constituents and Hepatotoxic Pyrrolizidine Alkaloids in Two Senecio Plants and Their Preparations by LC–UV and LC–MS

Abstract:Two Senecio plants, Senecio cannabifolius Less. and its variety S. cannabifolius Less. var. integrifolius (Kiodz.) Kidam., were both used as the raw material of Feining granule, a traditional Chinese medicine product for treating respiratory diseases. In this study, the chemical profiles of these two plants were investigated and compared by liquid chromatography-mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR). A total number of 83 constituents, including 55 organic acids, 11 flavonoids, 4 alkaloids, 3 terpenes and 10 other types of compounds, were characterized. The results indicated that the levels of most flavonoids were higher in S. cannabifolius than in S. cannabifolius var. integrifolius, however, the levels of hepatotoxic pyrrolizidine alkaloids (PAs) were higher in S. cannabifolius var. integrifolius than in S. cannabifolius. Fifteen constituents were evaluated on lipopolysaccharides (LPS) induced RAW 264.7 cells, and eleven of them showed inhibition effect against nitric oxide (NO) production. Finally, the levels of ten major constituents (including seven anti-inflammatory active ones) and two PAs in Feining granule from two Senecio plants were determined and compared by the LC-UV and LC-MS methods, respectively. It was found that one organic acid (homogentisic acid) and two PAs (seneciphylline and senecionine) had higher contents in the preparation of S. cannabifolius var. integrifolius than in that of S. cannabifolius, however, the situations were inverse for the levels of four organic acids and flavonoids (chlorogenic acid, hyperoside, isoquercitrin, and isochlorogenic acid B). Based on the above results, S. cannabifolius might be a better raw material for Feining granule than S. cannabifolius var. integrifolius, because it contained more anti-inflammatory constituents and less hepatotoxic PAs than the latter. However, more pharmacological evaluations should be carried out to support the selection. The results in this study were helpful for the quality control of Feining granule. (C) 2015 Elsevier B.V. Abstract:Objective To study the chemical constituents of Acanthopanax senticosus.Methods A.senticosus was extracted with 75% EtOH and separated with petroleum ether and EtOAc,successively.The EtOAc-soluble fraction was isolated by column chromatography and RP-preparative HPLC.Their structures were identified by spectroscopic method(MS,1H-NMR,and 13C-NMR).Results Nineteen compounds were isolated from the EtOAc fraction of A.senticosus and identified as quercetin(1),quercitrin(2),kaempferol(3),hyperoside(4),rutin(5),acacetin(6),daidzin(7),3′-methoxydaidzin(8),puerarin(9),3′-methoxypuerarin(10),4′-methoxypuerarin(11),syringaldehyde(12),syringic acid(13),glucosyringic acid(14),isofraxidin(15),2,3-di(3′,4′-methyl-endioxy-benzyl)-2-buten-4-olide(16),l-sesamin(17),methylpluviatolide(18),and 4′-hydroxy-2′-methoxycinnamaldehyde(19).Conclusion Compounds 6—11,14,16,18,and 19 are isolated from the plants in Acanthopanax(Deche et Planch.) Miq.for the first time.

Abstract:Objective To study the antibacterial constituents of Senecio cannabifolius.Methods The antibacterial effect of all the extracts was tested in vitro with Staphylococcus aureus,Bacillus subtilis(Gram-positive bacilli),and Escherichia coli(Gram-negative bacillus).Chemical constituents were isolated by chromatography.Physicochemical characters and spectroscopic analyses were employed for their structural identification.Results Five compounds were isolated and their structures were elucidated by chemical and spectral methods as follows: methyl-5-hydroxy-2-pyridinecarboxylate(E-13),6-hydroxy-7,7a-dihydro-2(6H)-benzofuranone(E-14),2-(1,4-dihydroxycyclohexanyl)-acetic acid(E-16),3-hydroxycyclohexanecarboxylic acid(E-17),and 4-hydroxy benzaldehyde(B-14).Conclusion All of them are isolated from the plants of Senecio L.for the first time.

Abstract:Casearia sylvestris (Salicaceae), popularly known as "guaçatonga", is a plant widely used in folk medicine to treat various diseases, including cancer. The present work deals with the chemical composition as well as the cytotoxic evaluation of its essential oil, its main constituent and derivatives. Thus, the crude essential oil from leaves of C. sylvestris was obtained using a Clevenger type apparatus and analyzed by GC/MS. This analysis afforded the identification of 23 substances, 13 of which corresponded to 98.73% of the total oil composition, with sesquiterpene a-zingiberene accounting for 50% of the oil. The essential oil was evaluated for cytotoxic activity against several tumor cell lines, giving IC50 values ranging from 12 to 153 mg/mL. Pure a-zingiberene, isolated from essential oil, was also evaluated against the tumor cell lines showing activity for HeLa, U-87, Siha and HL60 cell lines, but with IC50 values higher than those determined for the crude essential oil. Aiming to evaluate the effect of the double bonds of a-zingiberene on the cytotoxic activity, partially hydrogenated a-zingiberene (PHZ) and fully hydrogenated a-zingiberene (THZ) derivatives were obtained. For the partially hydrogenated derivative only cytotoxic activity to the B16F10-Nex2 cell line (IC50 65 mg/mL) was detected, while totally hydrogenated derivative showed cytotoxic activity for almost all cell lines, with B16F10-Nex2 and MCF-7 as exceptions and with IC50 values ranging from 34 to 65 mg/mL. These results indicate that cytotoxic activity is related with the state of oxidation of compound.

Abstract:Phytochemical investigation of the whole plants of Lycopodiella cernua resulted in the isolation and identification of three new compounds (1-3), namely lycocernuaside E (1), lycernuic ketone F (2), and lycernuic B (3) and 12 known ones (4-15). Their chemical structures were established based on 1 D/2D NMR spectroscopic and HR-ESI-MS data analyses. Compounds 5, 12, and 13 displayed NO inhibitory effects in LPS-stimulated BV2 cells, with IC50 values of 21.2 ± 1.1, 28.5 ± 1.4, and 21.9 ± 1.1 µM, respectively. In addition, cytotoxic activity of the isolated compounds against MCF7 (breast carcinoma), HepG2 (hepatocarcinoma), and SK-Mel2 (melanoma) cancer cell lines were also reported.

Abstract:Objective: To investigate the chemical constituents of dried whole plants of Artemisia annua. Method: The chemical constituents were isolated by repeated silica gel chromatography, medium pressure column chromatography, and semi-preparative HPLC, and their structures were elucidated by spectroscopic analyses and comparison of NMR data with those reported in literature. Result: 15 compounds were isolated and identified to be 5-O-[(E)-Caffeoyl] quinic acid(l), 1,3-di-O-caffeoylquinic acid(2), 4 5-di-O-caffeoylquinic acid(3), 3, 5-di-O-caffeoylquinic acid (4), 3, 4-di-O-caffeoylquinic acid (5), methyl-3,4-di-O-caffeoylquinic acid(6), methyl-3,5-di-O-caffeoylquinic acid(7), 3,6'-O-diferuloylsucrose(8), 5'-β-D-glucopyranosyloxyjasmonic acid(9), Scopoletin(10), scoparone (11), 4-O-β-D-glucopyranosyl-2-hydroxyl-6-methoxyacetophenone (12), chrysosplenol D (13), casticin (14), chrysosplenetin(15). Conclusion: Compounds 2, 6, 8 and 9 are obtained from the Artemisia genus for the first time. Compounds 7 and 15 are obtained from this plant for the first time.
Identification of the chemical components of Saussurea involucrata by high-resolution mass spectrometry and the mass spectral trees similarity filter technique

Abstract:Rationale: Saussurea involucrata is a rare traditional Chinese medicine (TCM) that displays anti-fatigue, anti-inflammatory and anti-tumor effects. In this paper, the different chemical components of Saussurea involucrata were characterized and identified over a wide dynamic range by high-performance liquid chromatography coupled with high-resolution hybrid mass spectrometry (HPLC/HRMS/MS(n)) and the mass spectral trees similarity filter (MTSF) technique. Methods: The aerial parts of Saussurea involucrata were extracted with 75% ethanol. The partial extract was separated on a chromatography column to concentrate the low-concentration compounds. Mass data were acquired using full-scan mass analysis (resolving power 50,000) with data-dependent incorporation of dynamic exclusion analysis. The identified compounds were used as templates to construct a database of mass spectral trees. Data for the unknown compounds were matched with those templates and matching candidate structures were obtained. Results: The detected compounds were characterized based on matching to candidate structures by the MTSF technique and were further identified by their accurate mass weight, multiple-stage analysis and fragmentation patterns and through comparison with literature data. A total of 38 compounds were identified including 19 flavones, 11 phenylpropanoids and 8 sphingolipids. Among them, 7 flavonoids, 8 phenylpropanoids and 8 sphingolipids were identified for the first time in Saussurea involucrata. Conclusions: HPLC/HRMS/MS(n) combined with MTSF was successfully used to discover and identify the chemical compounds in Saussurea involucrata. The results indicated that this combined technique was extremely useful for the rapid detection and identification of the chemical components in TCMs.

Their structures were elucidated by IR, MS, 1HNMR and 13CNMR analysis. RESTS: Six constituents were obtained, their structures were elucidated as grandifloric acid (I), 16β, 17-dihydroxy-kaurane (II), ferulic acid (II), heptacosanol (IV), β-sitosterol (V) and succinic acid (VI). CONCLUSION: Compounds (III), (IV) and (VI) were separated from S. glabrescens for first time.

Abstract:This paper aimed to study the chemical constituents from Clitocybe clavipes. Silica gel, ODS, Sephadex LH-20, and semi-p reparative HPLC were employed to separate the ethanol extract of C. clavipes. Six compounds were identified by ~1H-NMR, ~(13)CNMR,and ESI-MS as clavilactone L(1), clavilactone A(2), clavilactone B(3), clavilactone E(4), clavilactone H(5), and clav ilactone I(6). Among them, compound 1 was a new meroterpenoid with a 10-membered carbocycle connected to a hydroquinone. Theantitumor activities of compounds 1-6 were determined by the methyl thiazolyl tetrazolium(MTT) ass ay. The results showed that compounds 1-6 exerted inhibitory effects on the proliferation of human gastric cancer cells(MGC-803),human non-small cell lung cancer cells(A549), and cervical cancer cells(HeLa). Compound 1 exhibited significant inhibitory activity against MGC-803 cells, with the half maximal inhibitory concentration(IC_(50)) of 11. 76 μmol·L~(-1).
Compositions and Biological Activities of Essential Oils of Kadsura Longepedunculata and Schisandra Sphenanthera.

Abstract:The chemical compositions, antimicrobial activities, antioxidant activities and cytotoxicities of the essential oils isolated from the root of Kadsura longepedunculata Finet et Gagnep (KLREO) and the fruit of Schisandra sphenanthera Rehd. et Wills. (SSFEO) were investigated.The analyses of gas chromatography-mass spectrometry (GC-MS) showed that cadinane type compounds and their derivatives were rich in both oils (54.2% and 39.7%, respectively) and delta-cadinene was the major component of both oils (13.8% and 25.6%, respectively). The antimicrobial activities of both oils were evaluated against five microorganisms with the disc diffusion and the broth micro-dilution method. Results showed that Gram-positive bacteria were more sensitive to both oils than Gram-negative bacteria and the yeast. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the oil of KLREO were lower than those of SSFEO, indicating that the former possessed slightly stronger antibacterial capability than the latter. The reducing power and lipid peroxidation assays were employed to study the potential antioxidant activities of both oils. Both oils remarkably decreased the content of malondialdehyde (MDA) in rat liver homogenate in a dose dependent manner. The antioxidant activities of KLREO appeared to be more potent than that of SSFEO. The oils of KLREO and SSFEO exhibited concentration-dependent cytotoxicities and were proved to be toxic to HepG2 cells with IC(50) of 147 and 189 mug/ml, respectively.

Abstract:Chemical exploration of Artemisia myriantha Wall. ex Bess (Asteraceae) aerial parts afforded one new seco-guaianolide (1) and one new germacranolide (2). Structure elucidation of the new compounds was unambiguously performed based on HRESIMS, 1D and 2D NMR spectroscopic analyses. The two new compounds exhibited in vitro strong cytotoxic activities against human cancer cell lines HCT-8 with IC50 values of 0.76 and 0.28 μM, respectively.

Abstract:Abstract Saussurea costus ( S. costus ) is a medicinal plant from the Asteraceae family that is widely used in traditional medicine in Saudi Arabia. This study examines S. costus root extract for its chemical composition and its antioxidant, anti-cancer, and antibacterial properties. The results of the study on the methanol root extract of S. costus reveal a rich chemical composition, as identified by GC-MS/FID analysis. The extract also showed high levels of total phenolic content (188.2 ± 2.1 mg GAE/g DM) and total flavonoid content (129 ± 2.6 mg QE/g DM). In antioxidant tests, the extract exhibited strong activity, with the half-maximal inhibitory concentration (IC 50 ) values of 137.15 μg/mL for ABTS and 175.5 μg/mL for DPPH as compared to positive control’s IC 50 values of 45.5 ± 0.3 μg/mL for ABTS and 55.3 ± 0.1 μg/mL for DPPH. The cytotoxic assessment against MCF-7 and A549 cell lines showed notable effects, particularly at higher concentrations. Additionally, the extract induced apoptosis in these cell lines, evidenced by changes in gene expression. Antibacterial tests revealed significant activity against various strains, with MIC values ranging from 7.81 to 125 μg/mL. The study underscores the importance of plant extracts in modern healthcare and suggests future research directions, including clinical applications and compound identification.

Their structures were elucidated as 2α, 3β, 19α, 23, 24-pentahydroxyurs-12-en-28-oic acid-28-O-β-D-glucopyranoside (1), 2α, 3α, 19α, 24-tetrahydroxyurs-12-en-28-oic acid-28-O-β-D-glucopyranoside (2), 2α, 3α, 24-trihydroxyurs-12-en-28-oic acid (3), 2α, 3α, 24-trihydroxyolean-12-en-28-oic acid (4), 2α, 3α, 23, 24-tetrahydroxyurs -12-en-28-oic acid (5), 2α, 3β, 23, 24-tetrah-ydroxyurs-12-en-28-oic acid (6), 2α, 3β, 23-trihydroxy-12-en-28-oic acid (7), 2α, 3β, 23-trihydroxyurs-12, 20(30)-dien-28-oic acid (8), and 2α, 3α, 23-trihydroxyurs-12, 20(30)-dien-28-oic acid (9). Compounds 1 and 2 were isolated from the Actinidia genus for the first time. Compounds 2, 3, and 4 showed cytotoxic activity against human SKVO3 and TPC-1 cancer cell lines with IC₅₀ values ranging from 10.99 to 16.41 μmol•L⁻¹, compounds 3 and 4 have cytotoxic activity against human HeLa cancer cell line with IC₅₀ values of 15.53 and 13.07 μmol•L⁻¹, respectively.

Chemical constituents and coagulation activity of Syringa oblata Lindl flowers

Abstract:The leaves and bark of Syringa oblata Lindl are used as folk medicine which has heat-clearing, detoxifying, dampness-removing and jaundice-relieving effects. There are many studies about leaves of S. oblata because of its abundant resource, however, less reports about the components of S. oblata flowers. The previous studies on S. oblate flowers were mainly focused on the volatile components and its traditional pharmacological activity. Thus, this study aimed to investigate the nonvolatile chemical constituents and the coagulation activity of S. oblate flowers. The chemical constituents of S. oblate flowers were isolated with various column chromatographies and coagulation activity of the major constituents was investigated by assaying the activated partial thromboplastin time (APTT), prothrombin time (PT), thrombin time (TT) and fibrinogen (FIB) on plasma of rabbit in vitro. Fifteen known compounds (namely compound 1-15) were isolated from S. oblata flowers. Compound 6, 10, 11 and 14 were isolated from Syringa genus for the first time. Compound 1, 2, 4, 5, 8 and 9 were isolated from the plant for the first time. The results of coagulation activity showed that water part of S. oblate flowers, lauric acid and kaempferol-rutinose significantly shorten PT (P < 0.001), TT (P < 0.001) and APTT (P < 0.001) compared with blank group, thus revealed that water extract of S. oblate flowers, lauric acid and kaempferol-rutinose possessed the procoagulant activity, but the effects were not better than that of Yunnan Baiyao as positive control.
Chemical Constituents from the Roots of Lindera Glauca and Their Antitumor Activity on Four Different Cancer Cell Lines

Abstract:Objective:To study the chemical constitutes from the roots of Lindera glauca and the alkaloids influence on proliferation of HT-29,SGC-7901,SMMC-7721 and A549 cell lines.Methods:The constituents were isolated by column chromatography such as RP-18,Sephadex LH-20 and silica gel,and their structures were elucidated by spectroscopic data analysis and compared with literature data. The antitumor activity was determined by MTT assay.Results:Ten compounds had been isolated and identified as(-)-magnocurarine( 1),N-methyl-laurotetanine( 2),laurotetanine( 3),( +)-boldine( 4),(-)-norisoboldine( 5),( +)-norisocorydine( 6),pmethane-3,8-trans-diol( 7),p-methane-3,8-cis-diol( 8),eudesm-4( 15)-ene-7,11-diol( 9) and 4β,6β-dihydroxy-1α,5β( H)-guai-9-ene( 10). Compounds 2 ~ 4 showed significant inhibitory activities against HT-29,SGC-7901,SMMC-7721 and A549 cells.Conclusion:Compound 1,9 and 10 are isolated from this plant for the first time. The IC50 value of compound 2 against HT-29 and SGC-7901 cell lines is even lower than VP-16.
