Hypoxia-responsive Elements Enhancing the Toxicity of HSVtk-mediated Gene-Directed Enzyme Prodrug Therapy in Lung Cancer Cells Induced by Hypoxia

Abstract:OBJECTIVE:To observe the expression of the enhanced green fluorescent protein (EGFP) induced by hypoxia-responsive elements (HRE) in SPCA1 and A549 lung cancer cells exposed to hypoxia, and examine the toxicity of the promoter to control expression of HSVtk-mediated gene-directed enzyme prodrug therapy (GDEPT) to kill lung cancer cells. METHODS: Transient transfected cells with EGFP-constructs were exposed to hypoxia 24 h after transfection. Relative EGFP fluorescence was measured by a flow cytometer; HSVtk-constructs were transfected into hypoxic SPCA1 and A549 cells, then the cells were exposed to media containing ganciclovir (GCV) for 24 h. Two days later, the cell growth rate was detected by MTT assay. RESULTS: Compared with the corresponding oxic sample, the folds of induction of EGFP expression levels in hypoxic SPCA1 and A549 cells transiently transfected with the pDNA.HRE.EGFP plasmid were 2.02±0.23 and 2.37±0.24-fold; the cell survival of SPCA1 and A549 cells transiently transfected with the pDNA.HRE.HSVtk plasmid in hypoxia and GCV exposure group reduced by (29.9±1.60)% and (42.83±2.57)%, respectively. CONCLUSIONS: The promoters constructed are responded to hypoxia. The addition of GCV to hypoxic treatment significantly reduces plasmid-transfectesd cell survival.
