Abstract:Residual pesticides in soil may be taken up by crops and negatively affect food safety. The uptake mechanism of imidacloprid and propiconazole was studied using wheat roots. The factors affecting root uptake were also studied with different crops and in different soils. Imidacloprid and propiconazole were taken up by wheat roots mainly through the symplastic and apoplastic pathways, respectively. Root protein and lipid contents were the main factors affecting the uptake and accumulation of imidacloprid and propiconazole by different crop roots, respectively. The uptake of imidacloprid and propiconazole in soil by wheat plants was linearly correlated with their concentrations in soil pore water, which were governed by soil characteristics. These results are helpful for understanding and estimating crop uptake of residual pesticides in soils.

Abstract:The application of the herbicide chlorimuron-ethyl has a lasting toxic effect on some succession crops. Here, a bacterium capable of utilizing chlorimuron-ethyl as the sole source of nitrogen was isolated from the contaminated soil and was identified as Enterobacter ludwigii sp. CE-1, and its detoxification and degradation of the herbicide were then examined. The biodegradation of chlorimuron-ethyl by the isolate CE-1 was significantly accelerated with increasing concentration (1–10mg/l) and temperature (20–40°C). The optimal pH for the degradation of chlorimuron-ethyl by the isolate CE-1 was pH 7.0. A pathway for the biodegradation of chlorimuron-ethyl by the isolate CE-1 was proposed, in which it could be first converted into 2-amino-4-chloro-6-methoxypyrimidine and an intermediate product by the cleavage of the sulfonylurea bridge and then transformed into saccharin via hydrolysis and amidation. The plant height and fresh weight of corn that had been incubated in nutrient solution containing 0.2mg/l of chlorimuron-ethyl significantly recovered to 83.9% and 83.1% compared with those in the uninoculated control, although the root growth inhibition of chlorimuron-ethyl could not be alleviated after inoculation for 14 d. The results indicate that the isolate CE-1 is a promising bacterial resource for the biodegradation and detoxification of chlorimuron-ethyl.
Subcellular Distribution Governing Accumulation and Translocation of Pesticides in Wheat (triticum Aestivum L.)

Abstract:Root uptake, translocation, and subcellular distribution of six pesticides (dinotefuran, thiamethoxam, imidacloprid, imazethapyr, propiconazole, and chlorpyrifos) with K-ow ranging from -0.549 to 4.7 were investigated in wheat to study transportation and accumulation of pesticides. The root bioconcentration factor (RCF) of pesticides decreased with water solubility (R-2 = 0.6121) and increased with hydrophobicity (when the pH-adjusted log K-ow > 2, R-2 = 0.925), respectively. The translocation of neutral pesticides from roots to shoots increased positively with water solubility (R-2 > 0.6484) but decreased with hydrophobicity (R-2 > 0.8039). The subcellular fraction concentration factor (SFCF) increased linearly with hydrophobicity of the tested pesticides (R-2 > 0.958). The log RCF was positively correlated with log SFCF in root cell walls (R-2 = 0.9894) and organelles (R-2 = 0.9786). Transportation of the pesticides from roots to stems and stems to leaves was adversely affected by the log SFCF of cell walls and organelles of roots (R-2 > 0.7997) and stems (R-2 > 0.6666), respectively. Hydrophobicity-dependent SFCF is a factor governing accumulation of pesticides in roots after uptake and their subsequent upward translocation. (C) 2020 Elsevier Ltd. All rights reserved.

Abstract:Intensive and indiscriminate use of insecticides in agroecosystems causes phytotoxic disturbances in non-target crops. However, the mechanisms by which plants reprogram cellular metabolites to resist and tolerate such agrochemicals remain unclear. Here, the interaction between lettuce plants with imidacloprid and fenvalerate was investigated by the complementary use of physiological and metabolomic analyses. Neither imidacloprid nor fenvalerate induced overt phytotoxicity in lettuce seedlings. The plant biomass, chlorophyll fluorescence, lipid peroxidation, and membrane integrity were not significantly affected by the selected insecticides. Flavonoid content decreased by 25% in lettuce leaves under fenvalerate exposure, whereas polyphenol and flavonoid contents were not significantly altered by imidacloprid. Although the content of most of the nutrient element in the leaves remained the same following pesticide treatment, iron content decreased by 28.1% under imidacloprid exposure but increased by 22.8% under fenvalerate exposure. Metabolomic analysis revealed that the selected insecticides induced extensive metabolic reprogramming in lettuce roots and shoots. Imidacloprid dramatically increased the metabolism of several amino acids (arginine, cysteine, homoserine, and 4-hydroxyisoleucine), whereas markedly decreased the metabolism of various carbohydrates (glucose, raffinose, maltotetraose, maltopentaose, and stachyose). Fenvalerate did not significantly alter amino acid metabolism but decreased carbohydrate metabolism. Additionally, the relative abundance of most organic acids and polyphenolic compounds decreased significantly after pesticide exposure. These results suggest that plants might program their primary and secondary metabolism to resist and tolerate insecticides. The findings of this study provide important information on how neonicotinoid and pyrethroid insecticides affect the health and physiological state of plants, which are ultimately associated with crop yield and quality.

Abstract:The responses of trace elements and secondary metabolites to stress can reflect plant adaptation to the environment. If and how the imperative trace element Fe and the defensive secondary metabolite 2,4-dihydroxy-7-methoxy-2H-1,4-benzoxazine-3(4H)-one (DIMBOA) mediate the toxicity of chiral herbicides to nontarget plants remains inconclusive. We found that the herbicidal-active imazethapyr enantiomer [(R)-IM] stimulated heme oxygenase-1 activity, triggered the release of the catalytic product Fe2+, increased reactive oxygen species production, decreased the DIMBOA content, and increased the DIMBOA-Fe content. XAFS analyses and in vitro Fenton assays demonstrated that DIMBOA could relieve phytotoxicity by chelating excessive Fe3+ to restore Fe homeostasis. The free radical scavenging ability of the chelate of DIMBOA and Fe was also involved. This work refines the dual role of DIMBOA and Fe in mediating the enantioselective phytotoxicity of chiral herbicides, which provides a new direction for improving the herbicide resistance of crops.
The Dissipation Behavior and Risk Assessment of Carbendazim under Individual and Joint Applications on Peach (amygdalus Persica L.).

Abstract:Dissipation, residue levels, and ingestion risks of carbendazim in peach (Amygdalus persica L.) were investigated with individual and joint applications in the present study. The dissipation kinetics of carbendazim, chlorpyrifos, prochloraz, and imidacloprid were evaluated by the first-order kinetics. When carbendazim was individually applied, the final residual concentration was 2.97 mg kg-1 and the half-life was 17.4 d. In the joint application of carbendazim with chlorpyrifos, prochloraz, and imidacloprid, the residual concentrations at 35 d after spraying were 7.16, 7.50, and 4.26 mg kg-1 and the half-lives were 30.8, 23.7, and 23.2 d, respectively, which showed an increase of 1.3-1.8 times compared with the single application of carbendazim. In addition, the effects of household processing of rinsing and peeling were investigated, and a high removal rate of 54.6% and 76.5% were found. Furthermore, the carbendazim ingestion risk assessment was conducted, which indicated that the acute health risk (aHI) and hazard quotient (HQ) of carbendazim were all within acceptable levels ranging from 21.7% to 40.9%. However, a higher ingestion risk of carbendazim was found under the joint application. This study provides some preliminary guidance for the joint application and risk assessment of carbendazim in peach, which is worth further investigation.

Abstract:Objective:To study on residue trend of imidacloprid in fruits of Citrus grandis.Methods:It was examined by high performance liquid chromatography(HPLC) after trunk injection.Results:The residue of imidacloprid remained a low period in fruits of C.grandis after injection with 0.8 mL·cm-1 diluted of imidacloprid.When the trunk was injected with 1.5 mL·cm-1 diluted of imidacloprid,the residue level of imidacloprid in fruits increased on the 7th day immediately.And the peak level was detected on the 14th day after the treatment.The level of imidacloprid in fruits was 0.835 mg·kg-1 in the 30th day,which was lower than MRLs(1 mg·kg-1).Conclusions:When imidacloprid through trunk injection was applied to control bore pests of C.grandis,which was diluted with 1.5 mL·cm-1,the safe interval period was more than 30 d.

Abstract:Fluorescent labeling techniques have been increasingly used to study the transport of pesticides in crops instead of radioisotope techniques due to the high cost, low resolution and handling and disposal of radioisotope materials. Nevertheless, the use of fluorescent labeling techniques to study the uptake and translocation mechanisms of pesticides in crops has rarely been reported. The distribution, translocation and metabolism of imidacloprid in the vegetable pakchoi were studied via a combination of fluorescence microscopy and UPLC-MS/MS. The translocation rate of imidacloprid-loaded fluorescence double hollow shell mesoporous silica nanoparticles () under sunlight conditions were observed by fluorescence microscopy to be higher than that under the dark. The was mainly transported from the ducts to the stem and leaf parts through transpiration under hydroponic conditions. In contrast, was transported downward mainly through the sieve tubes of the phloem driven by photosynthesis after the foliar spray. Moreover, fewer metabolites of imidacloprid from were produced than those from 70% Im granules. This work revealed how was absorbed, distributed and translocated as well as how the released imidacloprid was metabolized in pakchoi plants.

Sorption and Degradation of Imidacloprid in Soil and Water

Abstract:Imidacloprid, the major component of many widely used insecticide formulations, is highly persistent in soils. In this study, the sorption of imidacloprid by six soils as well as its photodegradation and hydrolysis in water were studied. The soils differed significantly in organic matter content and other physical and chemical properties. Sorption increased with increasing soil organic matter content but was not significantly correlated with other soil properties. Removal of organic matter via H2O2 oxidation decreased the sorption. By normalizing the Freundlich coefficients (K-f) to organic matter contents, the variability in obtained sorption coefficient (K-om) was substantially reduced. These results indicate that soil organic matter was the primary sorptive medium for imidacloprid. The low heat of sorption calculated from K-om suggests that partition into soil organic matter was most likely the mechanism. The photodegradation and hydrolysis of imidacloprid in water followed pseudo-first-order kinetics; however, the latter process needed a six-time-higher activation energy. While both processes produced the same main intermediate, they occurred via different pathways. The hydrolysis of imidacloprid was not catalyzed by the high interlayer pH in the presence of metal-saturated clays, which appeared to result from the lack of the pesticide adsorption in the interlayers of clays.
Impact of Microorganisms, Humidity, and Temperature on the Enantioselective Degradation of Imazethapyr in Two Soils.

Abstract:Imazethapyr (IM) is a chiral herbicide composed of an (-)-R-enantiomer and an (+)-S-enantiomer with differential herbicidal activity. In this study, the effects of microbial organisms, humidity, and temperature on the selective degradation of the (-)-R- and (+)-S-enantiomers of IM were determined in silty loam (SL) and clay loam (CL) soil with different pH values. The (-)-R-enantiomer of IM was preferentially degraded in two soils under different microorganism, humidity, and temperature conditions. The average half-lives of R-IM ranged from 43 to 66.1 days and were significantly shorter (P < 0.05) than those of S-IM, which ranged from 51.4 to 79.8 days. The enantiomer fraction (EF = (+)-S-enantiomer/((-)-R-enantiomer + (+)-S-enantiomer)) values were used to describe the enantioselectivity of degradation of IM were >0.5 (P < 0.05) in two unsterilized soils under different humidity and temperature conditions. The highest EF values were observed at unsterilized CL soil samples under 50% maximum water-holding capacity (MWHC) and 25 °C environmental conditions. The EF values of the IM enantiomers were significantly higher (P < 0.05) in CL soils (higher pH = 5.81) and were 0.581 (unsterilized) and 0.575 (50% MWHC; 25 °C) compared with those recorded in SL soil (lower pH = 4.85). In addition, this study revealed that microbial organisms preferentially utilized the more herbicidal active IM enantiomer.
Translocation and metabolism of imidacloprid in cabbage: Application of 14C-labelling and LC-QTOF-MS

Abstract:Imidacloprid (IMI) is a widely used neonicotinoid insecticide effective against sucking and some chewing insects. Translocation and metabolism of IMI in plants are related to food safety. In this study, 14C-labeled IMI was used to investigate its translocation, transformation, radioactive IMI metabolites and possible metabolic pathways in cabbage. The amount of IMI accumulated in the edible part of cabbage accounted for 80.3–95.4% of the applied amounts by foliar application. There was a tendency to transport from edible parts to inedible parts. The proportions of extractable IMI decreased gradually from 92.4% to 83.0% in edible parts, greater than that in inedible parts over the experiment (0–19 days), while the bound residues showed an opposite trend. The half-life of IMI was determined as 33.0 and 63.0 days in the edible parts and whole plant, respectively. Five radioactive components including the parent IMI were detected by HPLC-LSC. The relative content of M1 was less than 0.01 mg kg−1, which was not required to identify according to the metabolic scheme proposed by the US Environmental Protection Agency. The metabolites N-nitro(1-6-chloro-3-pyridylmethyl)-4,5-dihydroxyimidazol-2-imine (M2), N-nitro(1-6-chloro-3-pyridylmethyl)-4/5-hydroxyimidazole-2-imine (M3) and 1/3-(1-6-chloro-3-pyridylmethyl)-2,4-imidazodione (M4) were identified by LC-QTOF-MS. The primary metabolism of IMI in cabbage included hydrolysis and oxidation. The residue level and daily intake values of IMI in cabbage were estimated to be 0.033–0.078 mg kg−1 and 9.56–20.01 ng d−1 kg−1, respectively, which were far below the maximum residue level and allowable daily intake values.

Abstract:This study aimed to develop a method, followed by gas chromatography-mass spectrometry, for detecting 37 pesticides in Chrysanthemum indicum (C. indicum) and investigating the decrease in the matrix-induced enhancement effect. The influence of QuEChERS extraction and matrix solid-phase dispersion (MSPD) on the recovery and matrix effect (ME) was compared. extraction and matrix solid-phase dispersion (MSPD) on the recovery and matrix effect (ME) was compared to decrease the ME. The cleanup sorbents, volume and type of solvent, and treatment time were optimized. The accuracy (as recovery), precision (as relative standard deviation, RSD), linearity, limit of quantitation, and limit of detection were determined. The recoveries at the three levels using mixed standard solution ranged between 76% and 120% with RSD ≤15%, and 76% and 120% with RSD ≤11% for MSPD and QuEChERS extraction, respectively. The results suggested that the ME for 21 pesticides was in the range of 80%–120% after MSPD and 15% after QuEChERS extraction. QuEChERS extraction was simpler and faster than MSPD. This methodology was applied in the analysis of 27 C. indicum samples; phorate was most frequently detected (63.0% of the sample).

Abstract:BACKGROUND Cyromazine and acetamiprid are widely applied as pesticides in agriculture, causing increasing concerns about their residues in crops. In this study, cyromazine, acetamiprid and their mixture were applied to cowpea to investigate their degradation dynamics and perform a dietary risk assessment. RESULTS The dissipation behavior of cyromazine and acetamiprid in the single- and mixed-pesticide groups followed first-order kinetics, with a linear correlation coefficient of 0.910-0.987. The half-lives of cyromazine and acetamiprid were 1.56-11.18 days in the four different matrices. The half-life of cyromazine in the mixed-pesticide group was similar to or even shorter than that in the single-pesticide group. The highest levels of cyromazine and acetamiprid in cowpea occurred with a preharvest interval (PHI) of 7 days and after two or three applications. These levels are below the maximum residue limits (MRLs) recommended by the Chinese Ministry of Agriculture for cyromazine and acetamiprid in cowpea. The risk quotient (RQ) of cyromazine and acetamiprid ranged from 0.0018 to 0.0418, and the NESTI values of the cyromazine and acetamiprid were far below the ARfD as recommended by the EFSA. CONCLUSION These results suggest that the use of cyromazine and acetamiprid and a cyromazine-acetamiprid mixture in cowpea is safe under the Good Agricultural Practices (GAP) for Chinese fields, and the use of a cyromazine-acetamiprid mixture affords even better results than the application of cyromazine alone. Moreover, the residue dynamics information will support the label claims for the application of cyromazine, acetamiprid and a cyromazine-acetamiprid mixture to cowpea fruit. Abstract:The degradation capacity of the four chlorimuron-ethyl-degrading strains, D310-1, LCY-2, LCY-3 and LCY-4, which identified as Rhodococcus sp.,Stenotrophomonas maltophilia sp,Bacillus subtilis sp., and Rhodotorula mucilaginosa sp., respectively, combined in different ways was examined in this study. Then the optimal degradation conditions and mitigation phytotoxicity of chlorimuron-ethyl on sensitive crop were investigated by both orthogonal test and pot culture experiment. The experimental results showed that the combination of four strains was adopted, and the removal rate of chlorimuron-ethyl reached the maximum of 90% in 30 days. Then the optimal degradation conditions were inoculum size 2.5%, temperature 28°C, pH 7.0 and the soil moisture 35%, and the mixed strains could eradicate 92.57% of chlorimuron-ethyl within 30 days. Furthermore pot culture experiment indicated that inoculation of mixed strains could also mitigate the phytotoxic effects of chlorimuron-ethyl on the growth of cucumber seedling.

Abstract:Artificial aphid infestation experiments on the three chrysanthemum cultivars 'Keiun', 'Han6' and 'Jinba' showed that the three cultivars vary markedly in their resistance. Of the three cultivars, the most resistant ('Keiun') produced the longest, highest and densest trichomes, the largest and fullest gland cells, and the most wax on the lower leaf epidermis. Superoxide dismutase (EC 1.15.1.1), peroxidase (EC 1.11.1.7), ascorbate peroxidase (EC 1.11.1.11), polyphenol oxidase activity (EC 1.14.18.1) and phenylalanine ammonia lyase (EC 4.3.1.5) were enhanced by aphid herbivory. In the two more resistant cultivars ('Keiun' and 'Han6'), the activity of superoxide dismutase and ascorbate peroxidase enzymes rapidly increased following infestation, and their levels remained high from seventy-two to one hundred and sixty-eight hours after inoculation. We suggest that these two antioxidant enzymes contribute to aphid resistance of these chrysanthemum cultivars. All three cultivars showed quick responses to aphid infestation by increasing polyphenol oxidase and phenylalanine ammonia lyase activities during the early period after inoculation. Activities of these two defense enzymes were higher in the two resistant cultivars after 72h after inoculation, suggesting involvement of these two enzymes in aphid resistance.
Dissipation and residues of fluroxypyr-meptyl in rice and environment

Abstract:A simple and reliable analytical method for analysis of fluroxypyr-meptyl residue in rice, soil and water was investigated. The residue levels and dissipation rates of fluroxypyr-meptyl (34% AS) were determined by GC-MSD in selected ion monitoring mode. In the supervised field trials, the half-life of fluroxypyr- meptyl in rice plant was in the range 1.25-4.13 days. The half-lives of fluroxypyr-meptyl in soil and water were 2.29 and 1.73 days, respectively. In order to assess the dissipation of fluroxypyr-meptyl in the environment, a laboratory simulated experiment was carried out in soil and water. The terminal residues of fluroxypyr-meptyl in rice plant, husked rice, rice hull and soil samples were undetectable at harvest. It showed that fluroxypyr-meptyl may be safe to use according to the recommended dosage suggested by the corporation.

Abstract:Pollen and nectar can be contaminated with a range of pesticides, including insecticides, fungicides, and herbicides. Since these matrices are important food sources for pollinators and other beneficial insects, their contamination can represent a key route of exposure. However, limited knowledge exists with respect to pesticide residue levels and their dynamics in these matrices for many crops and active ingredients (AIs). We used controlled glasshouse studies to investigate the residue dynamics of a systemic (cyprodinil) and a contact (fludioxonil) fungicide in the floral matrices and other plant parts of courgette/zucchini (Cucurbita pepo L.). We aimed to better understand the processes behind residue accumulation and decline in pollen and nectar. Each AI was applied to plants, either by spraying whole plants or by targeted spraying onto leaves only. Samples of pollen, nectar, anthers, flowers, and leaves were taken on the day of application and each subsequent morning for up to 13 days and analysed for residues using LC-MS/MS. Significant differences in residue levels and dynamics were found between AIs and floral matrices. The present study allowed for the identification of potential routes by which residues translocate between tissues and to link those to the physicochemical properties of each AI, which may facilitate the prediction of residue levels in pollen and nectar. Residues of the contact AI declined more quickly than those of the systemic AI in pollen and nectar. Our results further suggest that the risk of oral exposure for pollinators may be considerably reduced by using contact AIs during the green bud stage of plants, but application of systemic compounds could still result in a low, but continuous long-term exposure for pollinators with limited decline.
Evaluating the Safety of Imidacloprid FS Seed Treatment Use in Potato Production: A Case Study from China

Abstract:This study evaluated the residue behavior and dissipation dynamics of a new imidacloprid FS 600 seed treatment in potato cultivation systems in Shandong and Jilin, China. Sensitive and accurate UPLC-MS/MS methods were established to quantify imidacloprid residues in potatoes, potato plants, and soil. Results showed that imidacloprid dissipation followed a first-order kinetic model, with half-lives ranging from 6.9 to 26.7 days in plants and 19.8 to 28.9 days in soil. At harvest, the highest average residues in potatoes and soil were 0.778 mg/kg and 0.149 mg/kg, respectively. The dietary risk assessment indicated a chronic risk quotient (CRQ) of 39.73% for adults, indicating minimal risk to human consumers, while the ecological risk quotient (ERQ) and ecotoxicity exposure ratio (TER) revealed low to moderate toxicity to earthworms, warranting caution in the use of this formulation. This research provides valuable data for assessing the safety of imidacloprid FS seed treatment in potato cultivation.

Abstract:Imidacloprid is a broad-spectrum neonicotinoid class insecticide with systemic action, widely used on vegetables in India for the management of sucking insect pests. The overall pesticide usage profile in gourds growing districts of Tamil Nadu showed that imidacloprid as the most commonly used insecticide. The present study aimed to develop and validate an analytical approach for detecting imidacloprid and 6-chloronicotinic acid residues in bitter gourd fruit, juice and soil using LC-EI-MS (liquid chromatography coupled with electron ionization mass spectrometry) was undertaken. The persistence pattern, effect of household processing and risk assessment of imidacloprid on bitter gourd was studied by conducting field trials at single and double doses of 20 and 40 g a.i ha-1. Calibration curves showed a good linear relationship (r2>0.99) with the concentrations (0.0025–0.5 μg mL-1) of imidacloprid and 6-chloronicotinic acid. The limit of detection and quantification of the method were 0.008 and 0.025 mg kg-1, respectively. Accuracy of imidacloprid and 6-chloronicotinic acid residue recovery was in the range of 88–101 per cent with RSD of less than six per cent in all the matrices of bitter gourd. Initial deposits of imidacloprid at 20 and 40 g a.i ha-1 were 0.68 and 1.25 mg kg-1 and the residues persisted up to 10 and 15 days with their respective half-lives of 2.51 and 3.13 days. Simple decontamination techniques showed 33 to 80 per cent reduction of residues in samples collected up to 10 days after treatment. The estimated RQ was less than one indicating the level of risk to the consumer is negligible.
