Construction in vitro of tissue engineered compound with urothelium cells and collagen membrane

Abstract:AIM: To vaccinate the cultured porcine urothelial cells onto the bovine typeⅠcollagen membranes modified by polyvinyl alcohol, so as to investigate the feasibility of construction in vitro of tissue engineered compound for urothelium rebuilding on neobladders.METHODS: The experiment was conducted in the Medical Research Center of Guangdong Provincial People's Hospital between March and May 2005. ①Porcine bladder urothelial cells were isolated to perform primary culture and subculture. ②The reproduction rate of the bladder urothelial cells in collagen membranes extraction: The 2nd and 3rd generation cells were selected to make suspension with keratinocyte serum-free medium (KSFM), and were cultured in control group (normal KSFM) and experimental group (extraction), respectively. The reproduction rate of cells was measured with MTT assay (A490 value in experimental group/A490 value in control group ×100%) on the 1st, 3rd, 7th days, respectively, and the toxicity of the collagen membranes was evaluated according to the rules of relation between reproduction rate and the classification of toxicity from the Pharmacopoeia of the United States of America (USP). ③Cell adhesion and cell growth were observed by histological observation and scanning electron microscopy. RESULTS: ①Porcine bladder urothelial cells could be cultured to the 9th generations in vitro. ②The relative reproduction rate on the 1st, 3rd, 7th days was 94.59%, 100.95% and 82.42%, respectively: There was no significant difference between the experimental group and the control group (P > 0.05). The toxicity of the collagen membranes cells was in gradeⅠ, which meant no toxicity. ③The state of cell adhesion, growth and proliferation observed during the culture was good. CONCLUSION: Porcine bladder urothelial cells can be cultured successfully in this way, and the bovine typeⅠcollagen membranes is well compatible with urothelial cells, which can construct urinary bladder mucosa substitute with collagen membranes, so as to rebuild the urothelial layers on neobladders.
