Protective effect of parthenolide on human lens epithelial cells against oxidative stress-induced apoptosis

Abstract:Objective: The apoptosis of lens epithelial cells (LECs) is the common basis of cataract formation, and oxidative stress is one of major causes of LECs apoptosis. This study was performed to investigate the protective effect of the herbal constituent parthenolide on human HLECs against oxidative stress-induced apoptosis and possible molecular mechanism. Methods: Human LECs were incubated in DMEM medium and 15% bovine serum with H 2O 2 in the absence or presence of different doses of parthenolide (10, 20 and 50 μmol;/L). The apoptosis of cells were assessed by Annexin V-propidium iodide (PI) double staining flow cytometry. Expression of caspase-3 and caspase-9 were assayed by real-time PCR and Western blot. Results: The percentage of apoptotic human LECs significantly increased in H 2O 2 treatment group (46.17% ± 4.08%) compared with control group (1.93% ± 0.41%) (P = 0.000). Expression levels of caspase-3 and caspase-9 mRNA and protein were enhanced in H 2O 2 treatment group compared with control group. The percentages of apoptotic cells in H 2O 2 + parthenolide of 10,20 and 50 μmol/L treatment group was 37.43% ± 2.69%, 11.43% ± 2.48% and 6.21% ± 2.32%, respectively, showing a significant reduce in comparison with H 2O 2 treatment group in a dose-dependent manner (P < 0.01). The expressions of caspase-3 and caspase-9 by H 2O 2 activated human LECs were significantly reduced in different doses of parthenolide groups. Conclusion: Parthenolide prevents human LECs from oxidative stress-induced apoptosis through inhibiting the activation of caspase-3 and caspase-9, suggesting a potential protective effect of parthenolide against cataract formation.
