Abstract:Objective: To determine the effects of silibinin on inhibiting the proliferation of 5637 cell in vitro and to investigate the related mechanism. Methods: The cell growth activity was evaluated by MTT assay. Morphological changes were studied through the inverted phase contrast microscope. The expression of survivin (inhibitor of apoptosis protein) mRNA was determined by reverse transcription-polymerase chain reaction (RT-PCR). Results: Silibinin treatment of 5637 cells resulted in a moderate to strong growth inhibition in both a dose-and a time-dependent manner. There were significant differences between these groups (P < 0.01). These morphological changes of 5637 which were treated with silibinin: cells shrank and turned round, condensed cytoplasma and pycnosised nucleus were observed through the inverted phase contrast microscope. There was also significant difference in the positive expression of survivin mRNA between the 5637 cell treated by differentiated silibinin. Conclusion: Silibinin could inhibit the growth of human bladder cancer 5637 cells and down-regulate the expression of survivin.
