Unilateral lesion of the nigrostriatal pathway changes the neuronal activity of the reticular thalamic nucleus in the rat

Abstract:Objective: To investigate the changes in neuronal activity of the thalamic reticular nucleus (TRN) following the unilateral lesion of the nigrostriatal pathway. Methods: The changes in the firing rate and firing pattern of the TRN GABAergic neurons were examined with extracellular recording methods in control and Parkinson's disease (PD) rats. Results: The results showed that the firing rates of TRN neurons in control and PD rats were 14.7 ± 1.6 Hz (n = 23) and 33.8 ± 3.0 Hz (n = 29) , respectively, and the firing rate of TRN neurons in PD rats was significantly increased when compared to that of control rats (P < 0.001). In control rats, 17% of the neurons fired irregularly and 83% in bursts. In PD rats, 7% of the neurons fired regularly, 45% irregularly and 48% in bursts, the percentage of the neurons firing in bursts being siginificant lower than that of control rats (P < 0.05). Conclusion: Unilateral lesion of the nigrostiatal pathway induces an increase of the firing rate on the TRN GABAergic neurons and a decrease of the percentage of the neurons firing in bursts, suggesting that the changes may be associated with the increase of the cortical-TRN excitatory neurotransmission and decrease of the globus pallidus-TRN inhibitory neurotransmission.
