Association of lipoprotein lipase gene Hind Ⅲ polymorphism with blood lipid and coronary heart disease in type 2 diabetes mellitus patients

Abstract:AIM: To observe the distribution of the single nucleotide polymorphism in intron 8 (T495G) of the lipoprotein lipase (LPL) in the Han population in Tianjin and to explore the association between gene distribution and type 2 diabetic mellitus. METHODS: Totally 104 patients with type 2 diabetic mellitus were selected from Outpatient Clinic and Department of Endocrinology, General Hospital of Tianjin Medical University as diabetic group from January to June 2004. They all accorded with the Diagnostic Criteria of American Diabetes Association in 1997, including 70 males and 34 females, with an average of (50±5) years. There were 18 patients combined with coronary heart disease, which met the international diagnostic criteria of coronary heart disease. A total of 94 normal persons, who did body examination during the same period, were enrolled as control group, including 62 males and 32 females, with an average of (49±6) years, fasting blood glucose < 6.1 mmol/L. Content of glucose in plasma, levels of cholesterol, triacylglycerol and insulin in serum were determined by glucose oxidase-peroxydase method, CHOD-PAP method, GPO-PAP method and radioimmunity method, respectively. The T495G polymorphism of the intron 8 of LPL gene was determined by the polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) methods. According to the designed primers in this trial, the base in this site was T in both homologous chromosomes, indicating TT; the base in this site was G, indicating GG; the bases in this base were T and G, indicating TG. RESULTS: ①There was no significant difference in genotype and allele frequencies of T495G polymorphisms between these two groups. ②The level of triacylglycerol in diabetic patients of TT group was significantly higher than that in the TG/GG group (2.14±1.45,1.55±0.96 mmol/L ,P < 0.05). ③There was no significant difference in TT and TG/GG genotype in coronary heart disease and non-coronary heart disease patients of diabetic group. ④Multiple regression analysis showed that genotype, age and insulin resistance were associated with type 2 diabetic mellitus and high triacylglycerol. ⑤Diabetic patients with triacylglycerol > 1.7 mmol/L as high triacylglycerol group, triacylglycerol ≤ 1.7 mmol/L as low triacylglycerol group. In type 2 diabetic group, TT genotype and T allele was associated with high triacylglycerol, and there was no statistical difference in the control group. CONCLUSION: There is T495G polymorphism of the LPL gene in the Han population of Tianjin. The T495G polymorphism is not associated with type 2 diabetes mellitus, but it is associated with high triacylglycerol level in type 2 diabetes mellitus. The allele T is the independent risk factor of type 2 diabetes mellitus combined with high triacylglycerol level.
