Effects of homocysteine on the production of collagens and proliferation of rat vascular smooth muscle cells

Abstract:Aim: To investigate the relationship of concentration of homocysteine (Hcy) with production of collagen types I, III, IV, and proliferation of rat vascular smooth muscle cells (VSMCs). Methods: The experiment was carried out in the Laboratory of Cardiology Department, Ruijin Hospital of Shanghai Second Medical University from February 2005 to April 2006. 1 Rat thoracic artery smooth muscle cells were cultured primarily by tissue adhesion method. Then VSMCs were stimulated with different concentrations of Hcy (0.025, 0.05, 0.1, 0.2, 0.5, 1.0, 1.5, 2.0, 3.0, 5.0 mmol/L), while control group (without Hcy) and blank group (without cell) were taken for comparison. 2 After the cells were treated for 12, 24, 48 and 72 hours, cell viabilities were evaluated by MTT, cell DNA synthesis were measured by BrdU, and the effects of Hcy on cell cycle were assessed by flow cytometry. Collagen content was measured by ELISA. Results: 1 MTT showed that VSMCs proliferated significantly at concentration of both 0.025 mmol/L and 0.05 mmol/L Hcy within 12-48 hours. 2 BrdU showed that DNA synthesis was increased at concentration of both 0.025 mmol/L and 0.05 mmol/L Hcy within 24-48 hours (24 hour: 432±33, 774±52; 48 hour: 582±32, 816±62), and the proliferation was also strengthened (P < 0.05); With the increasing of Hcy concentration, DNA synthesis of rat VSMCs was induced to the lowest at 5 mmol/l. 3 Flow cytometry analysis in VSMCs stimulated Hcy for 24 hours elucidated that at concentration of both 0.025 mmol/L and 0.05 mmol/L Hcy, the percent of VSMCs was increased obviously in S phase compared with other groups. 4 ELISA showed that Collagen types I and IV increased significantly in a dose-dependent manner at elevated concentrations of Hcy; the amount of type VI collagen was significantly reduced by high concentration of Hcy in a dose-dependent manner (r2 =0.41; P < 0.001). Conclusion: Low concentration of Hcy can stimulate VSMCs to proliferate more rapidly, meanwhile Hcy may contribute to accelerate progression of atherosclerotic lesions with collagen alterations which transform the injury into fibrotic plaques.
