Proliferation of focal smooth muscle cells and intimal hyperplasia after injury of carotid artery in rats

Abstract:Aim: To study the effects of hyperhomocysteinemia on the expressions of focal inflammatory factors such as monocyte chemoattractant protein-1 (MCP-1) and intercellular adhesion molecule-1 (ICAM-1) as well as intimal hyperplasia (IH) and focal smooth muscle cells (SMCs) in a rat model of carotid artery injury. Methods: The experiment was conducted in the Institute of Cardiology, Peking: University First Hospital from October 2004 to February 2005. 1 Fifty male Wistar rats were divided randomly into two groups (n=25 per group). Control group was infused with tap water in stomach, and the experiment group was treated with tap water containing 1 g/kg L-methionine via intragastrically injection. After 4 weeks of their respective feed, all animals underwent left common carotid artery balloon denudation, and 3, 7, 14 and 28 days later, the plasma Homocysteine (Hcy) concentrations were measured. The morphology of carotid artery IH was studied. The expressions of MCP-1 and ICAM-1 were also examined. Results: 1 The levels of plasma Hcy in experiment group were significantly higher than that of control group [(106.19 ± 19.75), (4.90±0.10) μmol/L]. 2 Morphology of carotid artery: After 7 days of hematoxylin-eosin staining, IH developed in all balloon-injured arteries of two groups. At days 14 and 28, the intimal thickness, intimal area and intima/media area ratio were increased by 36% and 33%, 41% and 30%, 36% and 21%, respectively, whereas luminal area was reduced by 47% and 61%, separately in the experiment group compared with the control group. The difference was significant in above indexes except media area between the two groups. At day 14, the percentages of intima and media SMC proliferation cell nuclear antigen positive particles of the experiment group were also significantly promoted by 1.7-fold and 2.3-fold compared with the control group, which was less than 0.1% [(84.40±9.05)% vs. (49.00±4.51)%, P < 0.01; (53.30±5.25)% vs. (23.70±6.17)%, P < 0.01]. The expressions of MCP-1 and ICAM-1 in balloon-injured arteries was significantly higher in the experiment group than in the control group, and the former mainly distributed in intima and media while the latter one located in endothelium and intima, which were observed obvious at days 3 and 7, continued for 28 days. Conclusion: Hyperhomocysteinemia can increase the expressions of MCP-1 and ICAM-1, stimulate SMC proliferation and promote IH after balloon-injured carotid arteries.
