Effects of glucocorticoid receptor gene transfection on the expressions of inflammatory cytokines in rat alveolar macrophages

Abstract:AIM: To explore the regulatory role of glucocorticoid receptor (GR) gene transfection on the expressions of inflammatory cytokine in rat alveolar macrophages. METHODS: The experiment was conducted in the laboratory of respiratory diseases, General Hospital of Nanjing Military Command of Chinese PLA between October 2005 and March 2006. ①Ten healthy adult Wistar rats were selected either male or female. pCMV-rGR were used to transfer the in vitro cultured alveolar macrophages, which were divided into transfected group and non-transfected group. The cells of the transfected group were harvested after the plasmid was transfected for 24 hours, and observed at different time points: normal control with matching normal saline, lipopolysaccharide (LPS) and Dexamethasone (Dex) stimulated for 4, 8 and 12 hours with concentration of 100 μg/L and 1×10-5 mol/L, respectively. ②Twenty-four hours after transfection, the expressions of GR in total proteins and nuclear proteins were measured with Western Blotting. At the same time, tumor necrosis factor α (TNF-α) and interleukin 1 β (IL-1 β) in the supernatant of cell culture solution were measured with enzyme linked immunosorbent assay (ELISA). ③Analysis of variance was used for comparison among groups, and SNK test for comparison between two groups. RESULTS: ①Expression changes of GR after transfection of alveolar macrophages with plasmid: The GR expressions in total proteins of the transfected group were higher than those of the non-transfected group (3.75±0.25, 1.21±0.16, P < 0.01); but GR expressions in nuclear proteins of transfected group had no difference from the non-transfected group (1.15±0.12, 1.09±0.11, P > 0.05). ②GR expressions in nuclear proteins after treated with LPS and Dex: After LPS and Dex treatment for 4, 8 and 12 hours, the expressions of the transfected group were 4.02±0.09, 2.13±0.08 and 1.22±0.15, respectively, as were 4.08±0.13, 2.09±0.11 and 1.19±0.17 in the non-transfected group. The two groups had the same tendency in the changes of GR expressions, which reached the peak at 4 hours, but significantly decreased at 8 hour, and reached the lowest level at 12 hour. At the same time point, there were no significant differences between the two groups (P > 0.05). ③Expressions of cytokines in the supernatant after LPA and Dex treatment: After LPS and Dex treatment at 4 , 8 and 12 hours, the contents of TNF-α in the supernatant of the transfected group were (32.16±3.99), (56.82±8.43) and (73.34±9.97) ng/L, respectively, as were (34.28±4.25), (59.82±9.65) and (71.41±8.35) ng/L in the non-transfected group. The mass concentrations of IL-1 β in the supernatant of the transfected group were (67.57±18.37), (83.17±8.55) and (97.55±8.27) ng/L, as were (62.23±15.64), (85.72±9.37) and (99.07±9.25) ng/L in the non-transfected group. The content of TNF-α and IL-1 β in the supernatant increased gradually with time, and reached the peak at 12 hours. There were no obvious differences between the two groups at the same time point (P > 0.05). CONCLUSION: ①Exogenous GR in alveolar macrophages are over-expressed through gene transfection, which locates in cytoplasm. ②After Dex treatment, the GR expressions in nuclear proteins are significantly increased, but gradually decreased. ③After transfection, the over-expressed exogenous GR is not found nucleus shifting, and only endogenous GR is detected. ④Over-expressed exogenous GR through gene transfection cannot suppress the expressions of inflammatory cytokine in condition of inflammation.
