Gene polymorphisms of tumor necrosis factor and the susceptibility to nasopharyngeal carcinoma in Han population in Southwest China

Abstract:Aim: To explore the association of polymorphism distribution of tumor necrosis factor (TNF) genes with the susceptibility to nasopharyngeal carcinoma in Han population of Southwest China. Methods: 100 patients with nasopharyngeal carcinoma treated in the First Affiliated Hospital of West China University of Medical Sciences between October 2000 and September 2005, who were diagnosed by biopsy in pathological department, of which 44 cases were not treated, 37 treated by radiotherapy, and 19 by radiotherapy and chemotherapy; at the same time, 100 healthy physical examinees were selected as control. All subjects were Han population in the Southwest of China and agreed to the detection. The allelic frequency of the TNF-α (-308) and TNF-β (+252) loci were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) to analyze the relationship between the polymorphism of the two loci and the susceptibility to nasopharyngeal carcinoma. Results: The frequency of G/A genotype of TNF-β (+252) locus in the nasopharyngeal carcinoma patients was significantly higher than that of the control group (57%, 29%, P < 0.01); that of G/G genotype was lower than the control group (23%, 51 %, P < 0.01); that of A allele of TNF-β (+252) locus was higher than the control group (48.5%, 13%, P < 0.01). There were no significant differences in the frequencies of genotype and allele of TNF-α (-308) locus between the two groups. Conclusion: Association is not found between the polymorphism of TNF-α (-308) locus and the susceptibility to nasopharyngeal carcinoma, but is found in TNF-β (+252); the A allele might be the susceptible factor in the pathogenesis of nasopharyngeal carcinoma, and people with allele G/A mutation inlocus site are susceptible to nasopharyngeal carcinoma.
