Influence of nano-hydroxyapatite on bone regeneration by human bone marrow mesenchymal stem cells

Abstract:AIM: To explore the growth and differentiation features and bone regeneration of human bone marrow mesenchymal stem cells (hMSCs) cultured on nano-hydroxyapatite scaffold. METHODS: The experiment was performed in the medical laboratory of Xiangya Hospital of Central South University from March 2004 to July 2005. ①hMSCs were harvested by density gradient centrifugation, and cultivated in low-glucose DMEM medium containing 0.1 volume fraction fetal bovine serum. The cells were passaged when they almost fused. The third passage cells were inoculated onto the surface of nano-hydroxyapatite (Nanometer Program Team of National Hepatobiliary & Enteric Surgery Research Center of Ministry of Health) and induced differentiation with conditioned medium containing 1mmol/L dexamethasone, 50 mg/L ascorbic acid and 10 mg/L beta-sodium glycerophosphate. ②The proliferation of the cultured cells was evaluated by MTT on days 2, 4, 6, and 8 after migrated. Differentiation was observed by modified calcium-cobalt staining method on day 7 after ossified induction. The growth of cells on scaffolds was observed under scanning electron microscope on day 8. RESULTS: The absorbance value detected in MTT was increased with the culture time (P < 0.05). hMSCs adhered to the materials showed the typical appearance of osteoblasts after being induced differentiation for 7 days. The results of coloration showed that grey and black particles or block sedimentation were positive, and the cells presented polygon or short rectangle. In the scanning electron microscopic study, cells had grown into the porous coral after 8 days culture. CONCLUSION: hMSCs grow well on nano-hydroxyapatite material, indicating this material is appropriate to the growth and differentiate of hMSCs, and can be used as the carrier of bone tissue engineering in vitro.
