Organosilicon Material in Combination with Structure-Controlling Agents as a Basis for Immobilization of the Enzyme Glucose Oxidase

Abstract:In this work, the glucose oxidase enzyme was immobilized in a sol-gel matrix based on dimethyldiethoxysilane and tetraethoxysilane in a volume ratio of 50/50%. Polyvinyl alcohol and chitosan were used as structure control agents. A number of physicochemical methods were used to study the morphology of the resulting immobilized biomaterials. The formation of organosilicon polymers was confirmed using IR spectroscopy. Differences in the morphology of the resulting sol-gel matrix structures were determined by SEM. The distribution of pores in samples of synthesized materials was determined using the method of low-temperature nitrogen adsorption. The catalytic activity of immobilized enzymes was studied using the biosensor method. Glucose oxidase is characterized by similar sensitivity characteristics when encapsulated in a sol-gel matrix using the structure-controlling agents chitosan and polyvinyl alcohol (sensitivity coefficients (3.8 × 10 –3 and 3.0 × 10 –3 , mg O 2 min –1 mmol –1 , respectively). However, the addition of chitosan to the matrix allows the performance of the enzyme biosensor to be increased up to 17 days, as well as reducing the relative standard deviation, thereby increasing the stability of the biosensor.
