Abstract:OBJECTIVE:To study the expression of human VEGF165 cDNA in Pichia pastoris and to obtain high-level expression of recombinant human VEGP165 (hVEGF165) with good biological activity.METHODS:Amplifying hVEGF165 cDNA by PCR, after confirmed by DNA sequence analysis, the gene was inserted into the Pichia pastoris expression vector pPIC9K containing AOX1 promoter and a secreting signal peptides, the recombinant expression plasmid pPIC9K/VEGF165 was constructed and transformed into KM71. The multiple insert transformants were screened, fermented in flasks and induced by 1% methanol.RESULTS:After 4 days of methanol induction, the expressed hVEGF165 came up to 30% of total proteins in supernatant by SDS-PAGE. The expressed hVEGF165 was further proved having good antigenicity and high specificity by ELISA and Western blot assay, and having good biological activity to stimulate HUVEC proliferation.CONCLUSIONS:High-level expression of secreted hVEGF165 had been successfully achieved in Pichia pastoris expression system.

Abstract:The extracellular domain 2-4 loop cDNA of quail vascular endothelial growth factor receptor quek1 (qVEGFR2) was obtained from plasmid carrying qVEGFR2 by PCR. Then it was cloned into expression vector pPICZalphaA of Pichia pastoris. To construct recombinant expression plasmid pPICZalphaA-qVEGFR2, linearized pPICZalphaA-qVEGFR2 with SacI was transformed to electroporated Pichia pastoris GS115. Subsequently, positive clone was selected by PCR and its phenotype was determined. SDSPAGE and Western blot assays of culture medium from a methanol-induced expression strain demonstrated that recombinant qVEGFR2 proteins were expressed and secreted into the culture medium. These results could provide a basis for further researches on tumor protein vaccine as well as for the preparation of tumor protein vaccine with Pichia pastoris.

Abstract:OBJECTIVE:To develop a secreted expression system of xenogeneic homologous epidermal growth factor receptor (EGFR) ectodomain using Pichia pastoris (P. pastoris), thus providing a basis for subsequent antitumor immunological researches on EGFR subunit protein vaccine. METHODS:In order to construct the secreted expression vector pPICZaA-mer and pPICZaA-her, we recombined the cDNA of xenogeneic (human) EGFR ectodomain (her) and the corresponding control mouse EGFR ectodomain (mer) with the empty plasmid pPICZaA, respectively. Then the recombinant plasmids were linearized by Sac I and transformed into P. pastoris strain GS115 by electroporation. The positive transformants were selected on YPDS plate including Zeocin and were induced to express by methanol. Consequently, the proteins in the culture supernatant were assayed with SDS-PAGE and Western-blot. So we could choose the high-level secreted expression recombinant strain to produce protein vaccine vastly. RESULTS:We got the secreted expression of mouse and human EGFR ectodomain in P. pastoris. The relative molecular mass of the two aimed proteins is about 95 x 10(3). The Western-blot analysis demonstrated that the expression proteins have much good antigenicity and specificity. CONCLUSION:The recombinant plasmids pPICZaA-mer and pPICZaA-her could be induced to express the EGFR aimed proteins by methanol in methanol-trophic yeast expression system.

Abstract:Objective To express human vascular endothelial growth factor receptor Flt-1 extracellular domain 1-3 loop cDNA in Pichia. pastroris, and to purify the expressed product and detect its biological activity. Methods By inserting human Flt-1 ( 1-3 loop) cDNA coding 316 amino acid residues into Pichia pastoris expression vector pPIC9K containing AOX1 promoter and the sequences of a secreting signal peptides, a recombinant expression plasmid pPIC9K/Flt-1 (1-3) was constructed and transformed to yeast host strain GS115, then His(+) Mut(9) phenotype transformant was screened out and cultured in flasks, and Flt-1 ( 1-3) was expressed under the induction of 1% methanol. Results SDS-PAGE showed that after being induced with 1% methanol for 4d, the expressed product existed in supernatant in the form of soluble molecule and contained 60% of total protein expressed. Western blot showed good antigenicity and specificity of expressed product. After being purified by CM-Sepharose FF and Sephacryl S-100 chromatography, the purity of the expressed product reached above 90%. Biological assay proved that the expressed product could bind to hVEGF(165) and inhibit the proliferation of HUVEC stimulated by hVEGF(165). Conclusion Human vascular endothelial growth factor receptor Flt-1 extracellular domain 1-3 loop was successfully expressed. The study lays a foundation for further application of the expressed product in the treatment of vasoformation related diseases, such as tumor and diabetic retinopathy.

Abstract:AIM: A recombinanted DNA Pichia pastroris was constructed to express human vascular endothelial growth factor (hVEGF 165 ) with Pichia pastroris expression system. METHOD:A re comb inant expression plasmid pPIC 9K/hVEGF165 was constructed with pPIC9K vector and human VEGF 165 cDNA prepared from human tumor tissue,then transformed to yeas t host strain GS115 The positive transformants were screened out and cultured in flasks. The supernatant of culture was tested by SDS-PAGE and Western-blot. RESULT:PCR,DNA sequencing and enzyme breakage results showed that the recombined plasmid was constructed successfully. SDS-PAGE and Western-blot re sul t showed that hVEGF was expressed successfully. CONCLUSION: hV EG F 165 was successfully expressed by Pichia pastroris expression system .

Abstract:Objective:To establish a Pichia yeast expression system expressing EGF-like domains of thrombomo-dulin domain 2(TM-D2) to obtain recombinant TM-D2 protein. Methods: The nucleotide sequence encoding TM-D2 was amplified by PCR before being ligated into pPICZaB vector. Then TM-D2-pPICZaB vector was constructed and con-firmed by PCR and sequencing. After linearization, this recombinant vector was transformed into yeast GS115 by electric shock. Methanol was used to induce the expression of TM-D2, and the TM-D2 was identified by SDS-polyacrylamide gel electrophoresis and Western blotting. Results: Sequencing and PCR revealed that recombinant fragment of TM-D2 with his-tag was cloned into pPICZaB vector, and SDS-PAGE and Western blotting confirmed that TM-D2 protein was present in the supernatant. Conclusions: The recombinant vector TM-D2-pPICZaB is successfully constructed, and TM-D2 is well expressed in Pichia pastoris.

Abstract:Objective To express the tumor necrosis factor receptor-immunoglobulin G1 Fc fusion(TNFR-Fc) in Pichia pastoris and study the effect of N-glycoengineered process on the protein bioactivity.Methods The cDNA encoding TNFR-Fc fusion protein was subcloned into pPIC6αA vector to construct the expression plasmid pPIC6-TF that was transformed into the GSB,a glycoengineered P.pastoris strain.Positive colonies were selected by YPD culture containing blasticidin and further screened by induced expression on cellulose acetate and nitrocellulose membrane with HRP labeled human IgG.The TNFR-Fc/GSB57 strain,whose expression level was higher,was cultured in 10L fermentor.Purified TNFR-Fc was obtained by Protein A affinity chromatography from the fermentation.Results Bioactivity analysis showed that the purified protein could neutralize the cytotoxicity of 5×104U /L TNFα to L929 cells(EC50 1.48 μmol/L).Conclusion The activity of recombinant TNFR-Fc expressed in glycoengineered P.pastoris is higher than that of the wild type one,but still lower than that expressed in CHO.N-glycan should be further modified.

Abstract:The yeast Pichia pastoris with efficiently expressed recombinant human epidermal growth factor(rhEGF)was constructed and selected and fermentations were conducted in a 30 Lfermenter.The quantity of expression reached up to 1g/L.With the purification using chromatography methods,rhEGF could reach the purity of more than 95% and the yield was above 40%.MALDITOF MS and N-terminal protein sequence analyzer were utilized to analyze the molecular weight and N-terminal sequence of rhEGF.Using Balb/c 3T3 cell proliferation/MTT colorimetric method,the activity of rhEGF was determined,up to 1.0×106 IU/mg,equivalent to the WHO standard.This study screened out rhEGF P.pastoris strains for production and could purify rhEGF to satisfy a certain quality standard,which might provide a basis for the industrialized production of rhEGF.

Abstract:Vascular endothelial growth factor (VEGF) plays a key role in angiogenesis through binding to its specific receptors, which mainly occurs to VEGF receptor 2 (VEGFR-2), a kinase insert domain-containing receptor. Therefore, the disruption of VEGFR-2 signaling provides a promising therapeutic approach for the treatment of cancer by inhibiting abnormal or tumorinduced angiogenesis. To explore this potential, we expressed the catalytic domain of VEGFR- 2 (VEGFR-2-CD) as a soluble active kinase in Escherichia coli. The recombinant protein was purified and the VEGFR-2-CD activity was investigated. The obtained VEGFR-2-CD showed autophosphorylation activity and phosphate transfer activity comparable to the commercial enzyme. Furthermore, the IC50 value of known VEGFR-2 inhibitor was determined using the purified VEGFR-2-CD. These results indicated a possibility for functional and economical VEGFR-2-CD expression in E. coli to use for inhibitor screening.

Abstract:To prepare the human placental growth factor 2 (PIGF-2), human PIGF 2 gene was cloned into pPIC9K vector to construct the recombinant pPIC9K-PIGF-2 vector. Linearized recombinant pPIC9K-PIGF-2 was transformed into Pichia pastoris by electroporation. YPD-Geneticin plate was used to screen geneticin hyper-resistant colonies. The positive colonies were verified by PCR. Results of SDS-PAGE and Western blot showed that recombinant human PIGF-2 was expressed after being induced by methanol. Using its characteristic heparin binding, recombinant human PIGF-2 was successfully purified by heparin affinity column chromatography.

Abstract:Objective:To gain recombinant endostatin in Pichia pastoris.Methods:The total mRNA was isolated from Chinese fetal liver,endostatin cDNA was synthesized with RT PCR,then cloned into pGEM T vector.The positive clones were screened with PCR,restriction map and DNA sequencing.Endostatin cDNA was subcloned into Pichia expression vector pGAPZa.Pichia pastoris GS115 cells were transformed with recombinant DNA.SDS-PAGE was used to assay the expression of endostatin in recombinant GS115.After purified with G25 and heparin,the recombined protein was used in the proliferation assay of ECV304 cells.Results:DNA sequencing result showed the endostatin cDNA was successfully cloned.After transformation with recombined DNA,GS115 cells expressed recombinant human endostatin.MTT assay implicated the recombinant protein could inhibited the proliferation of ECV304 cell sejectivity.Conclusion:Recombinant human endostatin protein was sa cesefuely expressed in Pichia pastoris.

Abstract:Objective To construct a eukaryotic expression vector in Pichia pastoris containing human fibrinogen gene, in order to achieve high level secretory expression in extracellular.Methods Expression plasmid,pGAPZαA-FGB-FGG-FGA-AOX1,was constructed by inserting the synthesized sequence encoding human fibrinogen(FGA, FGB,FGG) and then introduced into Pichia pastoris SMD1168H by electroporation.Transformants were availably screened by Zeocin resistance,the expression of recombinant protein was identified by SDS-PAGE and Western blot analysis, the protein yield was tested by ELISA assay.After ultrafiltration and purification, the biological activity of protein was detected.Results The crude yield of human fibrinogen in Pichia pastoris supernatant reached 15 mg/L in flask and the biological aggregation activity was determined.Conclusion The human fibrinogen gene was obtained and successfully expressed in Pichia pastoris and the active products were secreted into the medium.

Abstract:Vascular endothelial growth factor is a specific stimulator for endothelial cells;it has shown a potential application in tissue ischemia field.This study has muted the heparin sensitive cleavage site and fused the heparin binding domain to the C terminus of VEGF165.Then it carries out yeast expression and purification of the yeast expression product and conducts a preliminary identification of biological activity.The results show that change structure of the reconstructed VEGF protein still able to form protein dimers,and retains the angiogenesis in vitro.The successful expression of reconstructed VEGF expression product has laid the foundation for further research its function.

Abstract:Objective To study efficient expression of Homo sapiens HER2/neu extracellular domain (ECD) in Pichia pastoris, and to offer a technical prerequisite for the further study of HER2/neu ECD as a vaccine in tumor immunotherapy. Methods Her2/neu ECD DNA obtained from human RNA by RT-PCR was cloned into vector pPICZα. After the sequence was confirmed , the recombinant vector was transformed into Pichia pastoris via electroporation. Pichia pastoris secreting HER2/neu ECD in culture supernatant were obtained to extract genomic DNA of transformed X-33 and perform PCR and clone primers. SDS-PAGE and Western blot were used to screen high expressed HER2/neu ECD engineering bacteria and to identify HER2/neu ECD in culture supernatant induced by methanol. Results The sequence of her2/neu ECD DNA obtained by RT-PCR cloning was identical with that published on GeneBank. Recombinant expression vector was constructed to obtain positive Zeocin resistance clone by electrotransformation. SDS-PAGE and Western blot analysis showed HER2/neu ECD in culture supernatant induced by methanol, up to 120 mg/L. Pichia pastoris effectively and stably expressed HER2/neu ECD. Conclusions Pichia pastoris engineering bacteria effectively expressing HER2/neu ECD could be constructed and screened by cloning her2/neu ECD.

Abstract:The gene of Endostatin that consisted of 20 codons of preference in Pichia pastoris was artificial synthesized according to its mature peptide sequence in human collagenⅩⅧ.The artificial gene was cloned into methanol inducible expression vector pPICZαA.The linearized recombinant plasmid was transformed into chromosome of Pichia pastoris SMD1168 strain by electroporation.5 recombinant yeast strains were fermented in flasks and 1 recombinant yeast strain P.pastoris Endo V was performed for high cell density fermentation.The expression of Endostatin in supernate was 80mg/L and 125mg/L,respectively.After purification,the Endostatin could restrain the proliferation of endothelium cells in vitro. ;

Abstract:AIM: To optimize the expressi on condition of recombined human vascular endothelial growth factor(hVEGF 16 5) with Pichia pastroris expression system,then to purify and to determine its ac tivity. METHOD: The inducing concentration of methanol an d inducing period were reviewed in this paper.The supernatant of culture was purified wit h Heparin-Sepharose CL6B affinity chromatography. Biological activities of pur if ied product were tested by CAM experiment and HMVECnd proliferation. Furtherm o re,antigenicity and specificity of expressed product were compared with human VEGF 165 control by ELISA. RESULT: After being induced by 1% methanol for 5 d,recombinant protein was expressed best. The purity of product was ab ov e 90% after being purified by Heparin-Sepharose CL6B affinity chromatography. Results showed that expressed protein could stimulate vascular endothelial growth and ha d the similar antigenicity to human VEGF 165. CONCLUSION:Recombined human VEGF 165 was expressed and purified successfully.

Abstract:A fusion expression vector pPIC3.5K-PDGFRβ was constructed to express recombinant receptor tyrosine kinase PDGFRβ and the right Pichia pastoris transformants were screened on his-deficient plates and YPD-G418 plates by turns after electroporation of strain GS115, a high yield strain named M3 was screened. The strain M3 was cultured in a 5 L fermentor and His-GFP-PDGFRβ fusion protein was purified by Ni2+ chelating affinity chromatography. One distinct peak was obtained after elution with 250 mmol/L imidazole. Fusion protein was proved to be 90.08 kD by western blotting, and have tyrosine kinase activity by ELISA. Results showed that the receptor tyrosine kinase PDGFRβ was successfully expressed in P. pastoris and could be used as a target for small molecule selective inhibitors screening.

Abstract:Classical swine fever(CSF) is a highly contagious disease with high death rate,and usually causes huge loss in pig industry,which is classified in List A disease of OIE.E2 protein is the main protective antigen of CSF,so considerable research has focused on genetically engineering vaccines based on E2.In this study,so as to provide technological support in producing CSFV vaccine,we used the recombinant plasmid pMD18-T-E2 as the PCR template and designed one pair of primers to amplify the E2 gene without TMR.The PCR products were inserted into secretory Pichia pastoris expression vector pPICZαA,then the vector was linearised and transformed into Pichia pastoris X33 by electrotransformation.Transformants with high copies were acquired when selected under Zeocin and were induced with methonl.SDS-PAGE and Western blot analysis showed that the recombinant E2 protein was successfully expressed in Pichia pastoris,which laid the foundation for developing the subunit vaccine.

Abstract:The expression condition of recombinant human vascular endothelial growth factor 121 in E.coli expression system is optimized in this paper.After being induced by 2% lactose for 6h,recombinant protein was expressed.Results showed that gene highly expressed its product in inclusion body. By denaturation,purification with DEAE-cellulose chromatography and renaturation,the inclusion body was purified.The purity of product was above 90% analyzed by SDS-PAGE.It can be concluded that the recombinant human VEGF121 was expressed and purified successfully.
