Experiment on autologous platelet-rich plasma instead of animal serum to induce and culture human bone marrow stromal cells in vitro

Abstract:AIM: To discuss a new method of bone marrow stromal cells BMSC culture in order to satisfy the need for various cytokines in cell culture. Meanwhile, to try to reduce the lead-in of extraneous antigenic substances in the process of cell culture so as to reach the demand of quantity and biological characteristics of the cells in the clinical use of tissue engineering bone. METHODS: The experiment was conducted at the tissue-engineering lab of Southern Medical University between March 2004 and March 2005. ①Harvesting of autologous platelet-rich plasma: The 200 mL autologous plasma mixed with anticoagulant, and centrifugated for twice. The first centrifugation was done at 20 ℃, 3 000 r per minute for 10 minutes. These at 3 mm under supernatant and separating surface were put into another centrifuge tube. The second centrifugation was done at 20 ℃, 3 600 r per minute for 15 minutes. Three fourths of the above layer was abandoned, and the others were shocked evenly in whirlpool oscillato, and that was platelet-rich plasma. The platelet-rich plasma was mixed with catayzer solution composed of 100 g/L calcium chloride and 400 IU/mL thrombase in the proportion of nine to one. After being mixed evenly, it was filtered with 0.22 μm filtering membrane, and that was extraction of platelet-rich plasma compound factor. ②Harvesting and cultivation of human BMSC: There were ten healthy adult volunteers excluded other systemic diseases, including 7 males and 3 females, who were aged from 15-35 years with an average of 27.3 years. 5 mL bone marrow was extracted. The obtained karyotes at 2×107 were inoculated in 10 cm2 culture bottle 100 mL/L platelet-rich plasma matched with low glucose DMEM. Passaged cells were cultured with 100 mL/L platelet-rich plasma, 50 mg/L ascorbic acid, 1×10-8 mol/L desamethasone and 1×10-3 mol/L β-sodium glycerophosphate. After rapid amplification, form and proliferation of cells were observed with inverted phase contrast microscope and scanning electron microscope. Biological characteristics of the cells was detected with alkaline phosphatase staining and calcium node staining, etc. RESULTS: ①Result of morphological observation of human BMSC: Two to four hours after inoculation of human BMSC, the cells adhered to wall, which showed multiple angle and shuttle shape after 24 hours. Six to eight days later, the cells extended the bottom of bottle and showed simple-layer cell fusion. After subculturing, the cells grew rapidly. After passaged induction, the inonulated cells that enlarged the bottom of bottle between 1 and 2 days showed long shuttle shape or irregular polygon and the pseudopodium were stretched out. Nucleus lied at on side of the cells. The growth became slow when the cells were in the 8th generation. There were granule-like deposits from the 10th generation with cell shedding floated in the liquid. Scanning electron microscope indicated that adherent cells showed shuttle shape or multiple angle shape, and the multiple apophysis showed irregular shape. ②Result of calcium node alizarin bordeaux staining: After the fusion of the 3rd generation cell, they were cultured till forming intensive cluster, appearing deposition of cell matrix in center. The alizarin bordeaux staining could show clearly the calcium node. ③Alkaline phosphatase staining: After the 3rd generation culture, there was a large amount of gray-black granule or block deposit, which showed positive alkaline phosphatase staining. CONCLUSION: ①The autologous platelet-rich plasma instead of animal serum to induce human BMSC in vitro. The results of the alkaline phosphatase staining and the calcium node staining show that the cells have good biological characteristics of osteoblasts. ②The cultured quantity and biological characteristics of the cells can reach the demand of clinical application rapidly, which is a good cultured method.
