Abstract:Objective To investigate the effect of atorvastatin-a HMG-CoA reductase inhibitor on the expression and activity of matrix metalloproteinase-9(MMP-9) and tissue factor(TF) on human monocytes induced by Oxidized LDL(oxLDL).Methods Human monocytes-derived macrophages were incubated with oxLDL in the presence or absence of different concentration atorvastatin.At the end of incubation,MMP-9 expression and its activity was assessed by ELISA and zymography analysis respectively.TF expression and its procoagulant activity was assessed by ELISA and a clotting assay respectively.Results The results showed that atorvastatin inhibited human monocytes-derived macrophages proliferation which can be eliminated by adding 100 μmol/L mevalonate.0.1 μmol/L and 0.5 μmol/L atorvastatin can inhibit MMP-9 expression(1.32±0.06 vs 2.57±0.06 μg/L,P0.05;1.25±0.04 vs 2.57±0.06 μg/L,P0.01,respectively)and its activity of human monocytes-derived macrophages significantly,and inhibit TF expression(3.34±0.66 vs 5.78±0.86 μg/L,P0.05;2.20±0.60 vs 5.78±0.86 μg/L,P0.01,respectively) and its procoagulant activity in a concentration-dependant manner.Conclusion Atorvastatin inhibit the proliferation of human monocytes-derived macrophages,and also inhibit MMP-9 and TF expressionsand their activities induced by oxLDL,which can lead to collagen degradation and blood clotting.
Tissue Inhibitor of Metalloproteinases-4 Suppresses Vascular Smooth Muscle Cell Migration and Induces Cell Apoptosis

Abstract:In a previous study, we have demonstrated that overexpression of the tissue inhibitors of metalloproteinases-4 (TIMP-4) can inhibit the neointima formation in the rat carotid model. To define the functions of tissue inhibitor of metalloproteinases-4 (TIMP-4) in SMCs, we transduced human TIMP-4 cDNA into rat aortic SMCs by using adenoviral vector. Overexpression of TIMP-4 blocked the conversion of pro-MMP-2 to the active form and inhibited basic fibroblast growth factor-induced migration by 56.7% (p < 0.01). Overexpression of TIMP-4 markedly increased apoptotic cell death without changing their proliferation. Importantly, overexpression of human TIMP-4 in the wall of balloon-injured rat carotid artery also increased SMC apoptosis. The percentages of TUNEL-positive cells of total cells increased significantly in AdTIMP-4 infected group compared with AdGFP infected group. These findings demonstrate that TIMP-4 can inhibit SMCs migration and induce apoptosis in vitro and in vivo, which may generate new targets for prevention and treatment of vascular diseases.
Effect of Tissue Inhibitor of Metalloproteinase 1 on the Biological Behaviors of Rat Dermal Fibroblast with High Matrix Metalloproteinase 9

Abstract:Objective To explore the effect of tissue inhibitor of metalloproteinase 1 (TIMP-1) on the biological behaviors of rat dermal fibroblast with high matrix metalloproteinase 9 ( MMP-9),in order to provide an experimental basis for looking for a new way to treat diabetic foot.Methods The cell model of skin fibroblast was established by using dermal fibroblast cell line CRL-1213 from SD rats with high expression of MMP-9 by high glucose (22.0 mmol/L) + high homocysteine (100 μmol/L) co-culture.Exogenous TIMP-1 was used to inhibite the activity of MMP-9.Realtime PCR and ELISA were used to detect the expression of MMP-9 mRNA and protein.Gelatin zymography was used to detect the activity of MMP-9.Flow cytometry was used to detect cell proliferation. CCK-8 was used to detect cell viability.ELISA assay was used to detect collagen (hydroxyproline) secretion. Scratch test was used to evaluate horizontal migration of cells.Transwell method was used to evaluate vertical migration of cells.ANOVA was used for data analysis.Results The expression difference of MMP-9 mRNA and protein between TIMP-1 group and high MMP-9 group had no statistically significant (P ＞ 0.05).But the activity of MMP-9 protein in TIMP-1 group was lower than that in high MMP-9 group ( 1.47 ± 0.13 vs 0.43 ± 0.13,t =12.495,P ≤ 0.01 ).Compared with control group,the proportion of S phase cells [ ( 9.31 ± 0.24 ) ％ vs ( 6.54 ± 0.29 ) ％ ],proliferation index [ ( 13.8 ± 0.5 ) ％ vs ( 11.3 ± 0.6) ％ ],cell viability ( 1.76 ± 0.13 vs 1.35 ± 0.12),collagen (hydroxyproline) secretion [ (1126± 63 ) vs (353 ± 61 ) μg/L],6 h horizontal migration rate [ (38.7 ±2.6)％ vs (21.3 ±2.1 )％ ] and the number of vertical migration cells (91 ±4 vs 71 ±4) in high MMP-9 group were decreased (t =16.433,7.328,5.113,19.847,11.529 and 8.124 repectively,all P ＜ 0.05).After interfered by TIMP-1,the proportion of S phase cells [ (6.54 ± 0.29 ) ％ vs (7.75 ±0.27)％],proliferation index [(11.3 ±0.6)％ vs (12.5 ±0.5)％],cell viability (1.35 ±0.12 vs 1.64 ± 0.14 ),collagen ( hydroxyproline ) secretion [ ( 353 ± 61 ) vs ( 829 ± 59 ) μg/L ],horizontal migration rate [ (21.3 ± 2.1 ) ％ vs (31.5 ± 2.7) ％ ] and the number of vertical migration cells (71 ± 4 vs 85 ± 4 ) were increased compared with those in high M MP-9 group( t =6.881,3.292,3.615,12.590,6.644and 6.015 respectively,all P ＜ 0.05).Conclusion The biological behaviors of skin fibroblasts with high expression of MMP-9 is inhibited.The inhibited effect can be decreased by exogenous TIMP-1.
Effects of Homocysteine on Matrix Metalloproteinase-1 and Tissue Inhibitors of Metalloproteinase-1 in Vascular Smooth Muscle Cells

Abstract:BACKGROUND: Homocysteine (HCY) has been verified as an independent risk factor of atherosclerosis and atherothrombosis of cardiovascular disease.OBJECTIVE: To observe the effects of HCY on the secretion and activity of matrix metallopotinase-1 (MMP-1) and tissue inhibitors of metalloproteinase-1 (TIMP-1) in vascular smooth muscle cells (VSMCs).DESIGN : Auto-control observation.SETTING: Pathology Room, Institute of Regeneration Medical Sciences, Jilin University.MATERIALS: In vitro cultured vascular smooth muscles cells (VSMCs) of rats were obtained from male Wistar rats with the body mass of about 150 g from weeks 4-6 supplied by Laboratory of Animals, Norman Bethune Medical Sciences Division, Jilin University.METHODS: The experiment was performed at the Pathology Room, Institute of Regeneration Medical Sciences, Jilin University from May 2001 to May 2003. VSMCs ofin vitro cultured rats were adopted and divided into 5 groups, 0(control group), 0.10, 0.25, 0.50 and 1.00 mmol/L HCY were added, respectively for 48 hours. Effect of HCY on activity of MMP-1 was observed with zymography. The secretions of MMP-1 and TIMP-1 and their mRNA expressions were studied with Western blot and semi-quantitative reverse transcription polymerase chin (RT-PCR).MAIN OUTCOME MEASURES: Activity of MMP-1, secretions of MMP-1 and TIMP-1 and their mRNA expressions.RESULTS: ①Secretions of MMP-1 and TIMP-1: The secretion of MMP-1 in the 0.25, 0.50 and 1.00 mmol/L HCY groups was lower significantly than that in the control group (P ＜ 0.05-0.01). The secretion of TIMP-1 in the 0.10, 0.25, 0.50 and 1.00 mmol/L HCY groups was higher than that in the control group (P＜ 0.05-0.01). ②The MMP-1 activity decreased with the increase of HCY, but reduced obviously in the 0.50 and 1.00 mmol/L HCY groups (P ＜ 0.01). ③The expression of MMP-1 mRNA in the 4 HCY groups was lower markedly than that in the control group (P ＜ 0.01). The expression of TIMP-1 mRNA in the 0.25 mmol/L HCY group was higher than that in the control group (P ＜ 0.05), and it was higher remarkably in the 0.50 and 1.00 mmol/L HCY groups than that in the control group (P ＜ 0.01).CONCLUSION: HCY can inhibit enzyme activity, decrease collagen degradation and induce collagen accumulation by inhibiting the secretion of MMP-1, which indicates that reduction of collagen degradation induced by HCY is one of the pathogenesies of atherosclerosis.
Expressions of TIMP-2, MMP-2 and MT1-MMP in Activated Hepatic Stellate Cells and Their Effects on the Synthesis and Secretion of Extracellular Matrix

Abstract:Objective To investigate the relationship among tissue inhibitors of metalloproteinase-2 (TIMP-2), matrix metalloproteinase-2 (MMP-2) and membrane type 1 matrix metalloproteinase (MT1-MMP) and study their effects on the synthesis and secretion of extracellular matrix (ECM). Methods HSCs were isolated from Sprague-Dawley rats by collagenase and pronase E perfusion of liver from through portal vein and by 12% Nycodenz gradient centrifugation. After activation of HSCs, HSCs were treated with the quantity ranging from 40 pmol to 160 pmol of chemically modified small interfering RNA (siRNA) targeting TIMP-2. The levels of hyaluronic acid (HA), procolagen type Ⅲ (PCⅢ) and hydroxyproline (Hyp) in supernatant were measured. Expressions of TIMP-2 mRNA, MMP-2 mRNA, MT1-MMP mRNA, MMP-13 mRNA, COL Ⅰ mRNA and COL Ⅲ mRNA were detected by quantitative mat-time PCR. Protein levels of TIMP-2, MT1-MMP and MMP-13 were evaluated by western blotting, and the content of MMP-2 was examined by gelatin zymography. Results The levels of HA, PCⅢ and Hyp in three groups treated with siRMA was decreased markedly compared with that in normal group. The expression of TIMP-2, MMP-2, MT1-MMP, COL Ⅰ and COL Ⅲ in three treated groups were significantly lower than that in normal group, but the expression of MMP-13 was significantly higher than that in normal group. Conclusion TIMP-2 mediated by MT1-MMP regulated MMP-2 activation. The siRNA targeting TIMP-2 reduces the expression of TIMP-2, which associated with down-regulation of MT1-MMP and MMP-2, could suppress the synthesis and secretion of ECM in activated HSCs.

Abstract:Tissue inhibitor of metalloproteinase 3 (TIMP3) is unique among the four TIMPs due to its extracellular matrix (ECM)-binding property and broad range of inhibitory substrates that includes matrix metalloproteinases (MMPs), a disintegrin and metalloproteinases (ADAMs), and ADAM with thrombospondin motifs (ADAMTSs). In addition to its metalloproteinase-inhibitory function, TIMP3 can interact with proteins in the extracellular space resulting in its multifarious functions. TIMP3 mRNA has a long 3' untranslated region (UTR) which is a target for numerous microRNAs. TIMP3 levels are reduced in various cardiovascular diseases, and studies have shown that TIMP3 replenishment ameliorates the disease, suggesting a therapeutic potential for TIMP3 in cardiovascular diseases. While significant efforts have been made in identifying the effector targets of TIMP3, the regulatory mechanism for the expression of this multi-functional TIMP has been less explored. Here, we provide an overview of TIMP3 gene structure, transcriptional and post-transcriptional regulators (transcription factors and microRNAs), protein structure and partners, its role in cardiovascular pathology and its application as therapy, while also drawing reference from TIMP3 function in other diseases.

Effects of Antisense Tissue Imhibitor of Metalloproteinase-1 Expressing Plasmid on Porcine Serum-Induced Rat Liver Fibrosis

Abstract:Objective To observe the effects of antisense tissue inhibitor of metalloproteinase-1 (TIMP-1) expressing plasmid on rat liver fibrosis.Methods RT-Nest-PCR and gene recombinant techniques were used to construct the rat antisense TIMP-1 recombinant plasmid which could be expressed in eucaryotic cells. The recombinant plasmid and empty vector (pcDNA3) were encapsulated by glycosyl-poly-L-lysine and then transduced into the porcire serum-induced liver fibrosis rat model. Expression of exogenous transfected gene was assessed by Northern blot, and the hepatic expression of TIMP-1 was evaluated by RT-PCR and Western blot. We also tested the hepatic interstitial collagenase activity with FITC-labled type Ⅰ collagen, and used the immunohistochemistry of hepatic type Ⅰ and Ⅲ collagen and VG staining for pathological study.Results The exogenous antisense TIMP-1 could be expressed in vivo, and could block the gene and protein expression of TIMP-1 in the liver of porcine serum-induced hepatic fibrosis rats at mRNA level, which might elevate the active and latent hepatic interstitial collagenase activity (P0.01); decrease the hydroxyproline content of liver tissue (P0.01), and promote the degradation of collagens tyep Ⅰ and Ⅲ (P0.01). Its expression also improved the hepatic pathology to a certain extent (P0.01). Conclusion The results demonstrate that the antisense TIMP-1 recombinant plasmid has some reversal effect on liver fibrosis which enables it to be a possible candidate of gene therapy in the future.

Abstract:Objective To study the expressing status of antisense tissue inhibitor of metalloproteina se-1(TIMP-1) in hepatic stellate cells (HSC) constructed in vitro, and to eval u ate the effects on the production of type Ⅰ and Ⅲ collagens secreted by activated rat HSC. Methods HSC were extracted from normal rat liver by pronase and co llagenase digestion and purified by centrifugal elutriation, and were cultured pla stic until they were activated to a myofibroblastic phenotype after 7-10 days. RT-nest-PCR and gene recombinant techniques were used to construct the rat ant isense TIMP-1 expression plasmid which can express in eukaryotic cells, and seg uenced after being counstructed. The expressing plasmid and the pcDNA3 empty pla smid were transfected into HSC by Effectene reagent separately. The cells were sel ected after growing in DMEM containing 400 μg/ml G418 for 3 to 4 weeks. Exp ression of TIMP-1 in HSC was d etermined by Northern blot and Western blot. We tested the interstitial collagen ase activity in culture media with FITC-labled type Ⅰ collagen as substrate. U ltimately, we quantified the typeⅠ and type Ⅲ collagen in HSC by Wester n blot. Results The exogenous antisense TIMP-1 recombinant plasmid could block the expression of TIMP-1 greatly, while there were not the same outcome i n pcDNA3 empty plasmid g roup and non-transfecting control group. The ratio of TIMP-1/GAPDH was 0.67, 2 .41 and 2.97 respectively at mRNA level( P 0.05); the ratio of TIMP-1/β-actin was 0.31, 0.98 and 1.32 respectively at protein leve l ( P 0.05). It may also elevate interstitial collagenase activity, the collage nase activity was 0.3049, 0.1411 and 0.1196 respectively( P 0.05), which lead s to decrease the amount of type Ⅰ, Ⅲ collagen in HSC. The ratio of collagen Ⅰ/β-actin was 0.63, 1.78 and 1.92 respectively ( P 0.05), and the ratio of collagen Ⅲ/β-actin was 0.59, 1.81 and 1.98 respectively( P 0.05 ). Conclusions The antisense TIMP-1 recombinant plasmid could express in HSC stably, and increase the interstitial collagenase activity in culture m edia, which might promote the capability of degrading typeⅠ, Ⅲ collagens. Also , it decreased the total amount of typeⅠ, Ⅲ collagen in HSC, diminishing the e xtracellular matrix over-deposition at the same time. It could be a novel metho d to reverse hepatic fibrosis in future.
Effects of monocyte-endothelium interactions on the expression of type IV collagenases in monocytes.

Abstract:The adhesion of monocytes to endothelial cells is one of the early stages in the development of atherosclerosis. The expression of type IV collagenases, which include matrix metalloproteinase (MMP)-2 and MMP-9, in monocytes is hypothesized to play an important role in monocyte infiltration and transformation into foam cells. The aim of the present study was to examine the effects of monocyte-endothelium interactions on the expression levels of type IV collagenases and their specific inhibitors in monocytes, and to investigate the roles of tumor necrosis factor (TNF)-alpha and interleukin (IL)-1 beta in this process. Monocytes were single-cultured or co-cultured with endothelial cells. The expression of the type IV collagenases, MMP-2 and MMP-9, and their specific inhibitors, tissue inhibitor of metalloproteinase (TIMP)-1 and TIMP-2, in monocytes was determined by immunohistochemistry followed by image analysis. The expression levels of MMP-2 and MMP-9 were found to be low in the single-culture monocytes, but increased significantly when the monocytes and endothelial cells were co-cultured. However, treatment with monoclonal INF-alpha or IL-1 beta antibodies partially inhibited the upregulatecl expression of MMP-2 and MMP-9 in the co-cultured monocytes. Expression of TIMP-1 and TIMP-2 was observed in the single monocyte culture, and a small increase in the expression levels was observed when the monocytes were co-cultured with endothelial cells. Therefore, monocyte-endothlium interactions were shown to increase the expression of type IV collagenases in monocytes, resulting in the loss of balance between MMP-2 and -9 with TIMP-1 and -2. In addition, INF-alpha and IL-1 beta were demonstrated to play important roles in this process.
Inhibition Of Adjuvant-Induced Arthritis By Systemic Tissue Inhibitor Of Metalloproteinases 4 Gene Delivery

Abstract:Objective. An imbalance in the matrix metalloproteinase:tissue inhibitor of metalloproteinases (MMP: TIMP) ratio in favor of MMP appears to be an important determinant of tissue damage in arthritis. We undertook this study to explore whether reversal of this imbalance in favor of TIMP would alter this process and to examine the mechanism of this alteration.Methods. We administered human TIMP-4 by electroporation-mediated intramuscular injection of naked DNA using the rat adjuvant-induced arthritis (AIA) model.Results. Intramuscular naked TIMP-4 gene administration resulted in high circulating TIMP-4 levels and completely abolished arthritis development in the rat AIA model. This inhibition was associated with significantly decreased MMP activity in the joint tissue as well as with significantly decreased serum and tissue tumor necrosis factor a levels and serum interleukin-la levels compared with animals with arthritis. The mutation of cysteine at position 1 of TIMP-4 failed to block the development of AIA.Conclusion. Our data indicate that TIMP-4 is a potent anttinflammatory agent, and that its antiarthritis function may be mediated by MMPs. Arthritis-inhibiting effects of TIMP-4 may suggest a unique application of this gene therapy method for arthritis.
Effect of Salvia Miltiorrhiza on MMP-1 and TIMP-1 Mrna Expression in Human Adhesion Tissue Fibroblasts

Abstract:Objective To study the effect of Salvia miltiorrhiza and its chemical compositions on the expression of MMP-1 and TIMP-1 mRNA in human adhesion tissue fibroblasts(ATF).Method ATF from human peritoneal adhesions were harvested by primary culture.The expression of MMP-1 and TIMP-1 mRNAs in ATF treated with Salvia miltiorrhiza extract,sodium danshensu,and sodium tanshinone ⅡA sulfonate were determined by real-time PCR.Dexamethasone was used as positive control.Results Compared with the control group,the levels of MMP-1 mRNA increased significantly in the animal treated with Salvia miltiorrhiza extract and sodium danshensu at the dosage of 10 or 40 μg·mL-1(P0.01).Meantime,the levels of TIMP-1 mRNA enhanced markedly in the animal treated with sodium danshensu at the dosage of 40 μg·mL-1(P0.01).There were no difference between the control and other groups(P0.05).Conclusion Salvia miltiorrhiza could upregulate MMP-1 mRNA expression in ATF,which inhibit the over synthesis of collagen and prevent the formation of adhesion.Danshensu play a major role in this process.
Tissue inhibitor of metalloproteinase-1 and -3 improves cardiac function in an ischemic cardiomyopathy model rat

Abstract:Matrix metalloproteinases (MMPs) and a family of tissue inhibitors of metalloproteinases (TIMPs) may contribute to myocardial remodeling in heart failure. TIMPs are the main inhibitors of MMPs and have other MMP-independent functions. Because little is known of the role of TIMPs in the heart, we examined the effects of TIMPs on cardiac fibroblasts (CFs) and cardiomyocytes. In vitro, TIMP-1-4 enhanced smooth muscle actin (SMA) expression in CFs, and TIMP-1 and TIMP-3 enhanced the expression of phosphorylated Smad-3 and phosphorylated transforming growth factor (TGF)-β type 1 receptor in CFs; this effect was inhibited by TGF-β receptor blocker SB-505124. TIMPs-1, -3, and -4 also inhibited the FAK, AKT, and ERK pathways that induce cardiac hypertrophy. TIMP-1 and TIMP-2 suppressed apoptosis in cardiomyocytes; in contrast, TIMP-4 induced apoptosis in CFs. TIMP-2 stimulated collagen synthesis. Collagen gels containing TIMP-1 or TIMP-3, which exhibit cardioprotective effects in vitro, were transplanted to the left ventricular anterior wall of a rat heart model of myocardial infarction. Gel-released TIMP-1 and TIMP-3 significantly improved cardiac function and myocardial remodeling and enhanced SMA expression in the infarcted area in ischemic cardiomyopathy model rats. Further, the transplantation of TIMP-1 or TIMP-3 gels inhibited apoptosis in the ischemic myocardium and reduced MMP-2 activity. TIMPs may be an ideal target of cardiac regeneration therapy.
Effects of Matrix Metalloproteinase-1 and Antisense Tissue Inhibitor of Metalloproteinase-1 Recombinant Plasmid on Rat Liver Fibrosis

Abstract:Objective To study the effects of matrix metalloproteinase 1 (MMP 1) and antisense tissue inhibitor of metalloproteinase 1 (TIMP 1) recombinant plasmid on rat liver fibrosis. Methods We constructed the rat's MMP 1 and antisense TIMP 1 recombinant plasmids by reverse transcription nested polymerase chain reaction (RT Nested PCR) and the technique of DNA recombination. After encapsulated by lipofectin, the MMP 1 and/or antisense TIMP 1 recombinant plasmids was transferred to the rats with liver fibrosis in vivo intraperitoneally, then the effects of the plasmids on the hepatic fibrosis were observed by RT PCR for MMP 1 and TIMP 1, immunohistochemical assay for type Ⅰ and type Ⅲ collagen, and Masson staining for the observation of pathology. Results The recombinant plasmids of MMP 1 and antisense TIMP 1 could enhance the degradation of type Ⅰ and type Ⅲ collagen in the rat's fibrotic liver compared with fibrotic modal group ( P 0.01 ) or compared with colchicine treated group ( P 0.05). Moreover the recombinant plasmids of MMP 1 and antisense TIMP 1 have some reverse effects on the hepatic fibrosis compared with fibrotic modal group ( P0.01～P0.05 ), but no obvious difference was found between the groups of plasmid of MMP 1 and colchicine ( P 0.05). Conclusion The recombinant plasmids of MMP 1 and antisense TIMP 1 have some reverse effects on hepatic fibrosis to a greater or less degree. Using the two plasmids together has the stronger effect than using the plasmid of antisense TIMP 1 alone and using the plasmid of MMP 1 only lead to a limited changes.
Eeffect of tectorigenin on myocardial fibrosis in rats and its mechanism

Abstract:Objective:To explore the effect of tectorigenin on myocardial fibrosis(MF) in the rats and clarify the related mechanism,and to provide reference for its clinical application. Methods:Sixty Wistar rats were randomly divided into normal control group,model group,positive drug (capropril) control group, and low,middle,high doses of tectorigenin groups(n=10).Except normal control group, the rats in other groups were used to construct MF models by subcutaneous injection of 5 mg·kg -1·d -1 isoproterenol (Iso) for 7 d.The rats in tectorigenin groups and captopril group were intragastricly administrated with different doses of tectorigenin (25,50,100 mg·kg-1·d-1)and captopril(10 mg·kg -1·d -1) from the second day after modeling for consecutive 28 d.Bl-420E+ biological function experiment system was used to detect the heart function;Heart mass index (HMI) and left ventricular mass index (LVMI) were measured after experiment.UV detection was used to measure the levels of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) in myocardial tissue.Microplate reader was used to measure the activities of lactic dehydrogenase(LDH) and creatine kinase(CK) and the levels of nitric oxide (NO)in serum.ELISA were used to detect the levels of collagen typeⅠ(ColⅠ) and collagen type Ⅲ (Col Ⅲ) in myocardium tissue of the rats.The pathological changes of myocardium tissue of the rats in various groups were observed by HE staining.Results:Compared with normal control group,the HR of rats in model groups was increased,and the left ventricular systolic pressure (LVSP) was decreased(P<0.01);the HMI and LVMI were increased(P<0.05),the levels of MDA in left ventricular myocardial tissue was increased(P<0.01),and the activity of SOD was decreased,the levels of serum ColⅠ,Col Ⅲ and the activities of LDH , CK were also increased(P<0.01);the level of NO in serum was decreased(P<0.01).Compared with model groups, the HR were decreased,LVSP were increased, and HMI and LVMI of the rats in different doses of tectorigenin groups were decreased in a dose-dependent manner;the levels of MDA were reduced;the activities of SOD were increased in myocardium tissue,and the CK activities and the ColⅠ and ColⅢ levels were decreased(P<0.05 or P<0.01);the LDH activities in middle and high doses of tectorgenin groups were decreased(P<0.01);and the levels of NO in serum in different doses of tectorigenin groups were significantly increased(P<0.05 or P<0.01) .Conclusion:Tectorigenin could inhibit the MF induced by Iso in the rats, and its mechanism may be related to antioxidation,scavenging free radical and inhibition of collagen synthesis.

Abstract:In response to physical, chemical, and/or biological stimuli, considerable tissue self-degradation occurs in abalone, causing severe post-harvest quality loss. During this process, the extracellular matrix (ECM) is greatly degraded by endogenous proteases. The main component of the ECM is collagen, primarily type I collagen. Although the activity of matrix metalloproteinases (MMPs), which can specifically degrade collagen, is precisely regulated by tissue inhibitors of MPs (TIMPs), indicating that MMPs and TIMPs play crucial roles in the regulation of tissue self-degradation, few studies have reported the interaction between MMPs and TIMPs. In this study, we reveal collagenases to participate in postmortem tissue self-degradation of Haliotis discus hannai by degrading type I collagen. The recombinant MMP-1 catalytic domain (rMMP1c) of abalone with high purity and enzyme activity is expressed using a prokaryotic expression system. The optimum temperature and pH for rMMP1c are 37 °C and 7.0, respectively. The thermal denaturation temperature of rMMP1c is 67.0 ± 0.9 °C. Ethylenediamine tetraacetic acid (EDTA) and 1,10-phenanthroline can completely inhibit rMMP1c activity, while Ba2+, Ca2+, and Mg2+ can significantly elevate it. TIMP is also expressed using HEK 293F cells. Recombinant TIMP (rTIMP) shows good inhibitory activity toward rMMP1c. Inhibition kinetics analyses reveal rTIMP to be a competitive inhibitor of rMMP1c. Biolayer interferometry reveals that rTIMP can effectively bind with rMMP1c, with an equilibrium dissociation constant value of 263 nM. rMMP1c effectively degrades type I collagen γ-β-α chains in turn, and rTIMP can significantly inhibit rMMP1c degradation activity. These results provide a theoretical basis for the study of MMP and TIMP interaction and elucidate the possible mechanism for abalone tissue self-degradation.

Abstract:AIM To examine the expressions of matrix metalloproteinases-2 (MMP-2) and tissue inhibitor of metalloproteinases-1 (TIMP-1) in rat fibrotic liver and in normal rat hepatic stellate cells, and to investigate the changes in their expressions in response to treatment with interleukin-10 (IL-10) and platelet-derived growth factor (PDGF). METHODS Rat models of CCl4-induced hepatic fibrosis were established and the liver tissues were sampled from the rats with or without IL-10 treatment, and also from the control rats. The expressions of MMP-2 and TIMP-1 in liver tissues were detected by S-P immunohistochemistry, and their expression intensities were evaluated in different groups. Hepatic stellate cells (HSCs) were isolated from normal rat and cultured in vitro prior to exposure to PDGF treatment or co-treatment with IL-10 and PDGF. MMP-2 and TIMP-1 levels were measured by semi-quantitative reverse transcriptional polymerase chain reaction (RT-PCR). RESULTS CCl4- induced rat hepatic fibrosis models were successfully established. The positive expressions of MMP-2 and TIMP-1 increased obviously with the development of hepatic fibrosis, especially in untreated model group (84.0% and 92.0%, P<0.01). The positive signals decreased significantly following IL-10 treatment (39.3% and 71.4%, P<0.01 and P<0.05) in a time-dependent manner. TIMP-1 mRNA in PDGF-treated group was significantly increased time-dependently in comparison with that of the control group, but PDGF did not obviously affect MMP-2 expression. No difference was noted in TIMP-1 and MMP-2 expressions in HSCs after IL-10 and PDGF treatment (P>0.05). CONCLUSION MMP-2 and TIMP-1 expressions increase in liver tissues with the development of fibrosis, which can be inhibited by exogenous IL-10 inhibitor. PDGF induces the up-regulation of TIMP-1 but not MMP-2 in the HSCs. IL-10 inhibits TIMP-1 and MMP-2 expressions in HSCs induced by PDGF.
Effect of Transfected Tissue Inhibitor of Metalloproteinase-2 on Nuclear Factor-Κb and Protein Kinase C of the Injured Rabbit Vascular Smooth Cells

Abstract:Objective To study the expression changes of matrix metalloproteinases (MMPs), protein kinase C (PKC) and nuclear factor-κB (NF-κB ) on the injured rabbit vascular smooth muscular cells (VSMC) transfected with tissue inhibitor of metalloproteinase-2 (TIMP-2) vector. Methods Lipofectin method was used to transfect TIMP-2 vector into VSMC, Western blot analysis to detect TIMP-2 peptides and zymography assay to determine MMPs. The activities of MMPs and NF-κB were detected by electrophoretic mobility shift assay (EMSA). The mRNA and protein expressions of PKCα were determined by RT-PCR and immunocytochemistry. Results The injured VSMC showed increased enzyme activity of MMP-2. There was very lower level expressions of PKCα and NF-κB in the normal VSMC but high in the injured VSMC. However, in injured VSMC transfected with TIMP-2 vector, the activity of MMP2/9 was suppressed and the expressions of PKCα and NF-κB decreased (P0.05). Conclusions PKCα and NF-κB play an important role in synthesis of MMPs and mobility of the injured VSMC.TIMP-2 prevents activation of MMPs and restenosis via blocking expressions of PKCα and NF-κB.

Abstract:Objective To investigate the effects of different intensities of treadmill running on the expression of matrix metalloproteinase-3 (MMP-3),collagen-Ⅱ (COL-Ⅱ) and tissue inhibitor of metalloproteinase-3 (TIMP-3) in the articular cartilage of the knee joints of rats.Methods Twenty four male SpragueDawley rats were randomly divided into three groups: control group (n=8),low-intensity running (LIR) group (n=8) and high-intensity running (HIR) group (n=8).The rats in LIR and HIR group underwent treadmill running exercise.After 6 weeks,the bilateral knee joints were harvested.The left knee joints were obtained to observe the change of COL-Ⅱ in the cartilage by immunohistochemistry.However,the right knee joints were used to observe the expression of mRNA and protein of MMP-3,COL-Ⅱ and TIMP-3 by RT-PCR or Western bolt.Results The optical density of COL-Ⅱ protein in HIR group was significantly lower than those in control and LIR groups.The MMP-3 and COL-Ⅱ mRNA expression levels in HIR group was significantly higher than those in control and LIR groups.However,the TIMP-3 mRNA expression level in HIR group was lower than those in control and LIR groups.Although MMP-3 and COL-Ⅱ protein expression increased and TIMP-3 protein expression decreased with the increase of exercise intensity,COL-Ⅱ protein expression level in HIR group was lower than that in LIR group.Conclusion Moderate-intensity running could enhance the metabolism of cartilage matrix by regulating the expression of MMP-3,COL-Ⅱ and TIMP-3.Running intensity may be a key factor to regulate cartilage change.
[Effect of anti-fibrosis compound contained serum on procollagen Type I and IV, matrix metalloproteinase and its tissue inhibitor-1 gene expression in HSC-LI90 cell line]

Abstract:Objective: To study the effects of Anti-fibrosis Compound contained serum (AFCS) on procollagen type I and IV (ProC-I and ProC-IV), matrix metalloproteinase (MMP) and its tissue inhibitor (TIMP-1) gene expression in hepatic stellate cell line LI90 (HSC-LI90). Methods: AFCS was prepared by gastric infusing different dosage (0.5 g/kg, 2.0 g/kg and 4.0 g/kg) of Anti-fibrosis Compound Recipe to rats. After HSC-LI90 cells were exposed to AFCS for 48 hrs, levels of ProC-I, ProC-IV, gene expression of MMP-2, MMP membrane type 1 (MT1-MMP) and TIMP-1 in the cells were detected by Northern blot, and gelatinase activity of MMP-2 was measured by zymography. Results: AFCS of different concentrations could inhibit ProC-I and ProC-IV and TIMP-1 gene expression (P < 0.05 or P < 0.01), increase MT1-MMP gene expression (P < 0.01), but it showed no effect on gene expression and activity of MMP-2 (P > 0.05). Conclusion: Anti-fibrosis Compound Recipe has anti-liver fibrosis action, its effects in inhibiting TIMP-1 gene expression of HSC-LI90 cells and promoting degradation of collagen might be one of the mechanisms of the action.
