Effect of puerarin on the expression of monocyte chemoattractant protein-1 in ischemia/reperfusion injured retina of rats and its significance

Abstract:Aim: To observe the effect of puerarin on the expression of monocyte chemoattractant protein-1 (MCP-1) in rat's ischemia/reperfusion injured retina and its protection for rat's retina. Methods: The experiment was performed at the Institute of Department of Ophthalmology, Medical College, Nanchang University from October to December 2005. Totally 90 clean SD rats, of either sex, were selected and randomly divided into ischemia/reperfusion group, puerarin group and sham operation group with 30 in each group. The models of retinal ischemia/reperfusion were employed in the ischemia/reperfusion group and puerarin group. The sham operation group received the same intervention with the exception of common carotid artery deligation. The puerarin was used as interventional material in the puerarin group (40 mg/kg). After 1, 6, 12, 24, 48 and 72 hours of ischemia/reperfusion, the rats received alectroretimogram detection and immunohistochemistry detection, 5 rats in each group at each time point. The expression of MCP-1 in retina was determined with immunohistochemical method. The influence of puerarin on the expression of MCP-1 in retina was observed at each time phase. Amplitude of wave of electroretimogram a and b was recorded in rats at each time phase. The ratio of left/right eye's electroretimogram a and b wave was looked as the relatively recovery rate to appraise the retinal function. Results: All the experimental animals were involved in the result analysis. 1 MCP-1 in the ischemia/reperfusion group began to express and augmented gradually at hour 6 after reperfusion, reached the peak at hour 24, mainly expressed in cytoplasm and nucleus of inter and outer nuclear layer in retina. No significant MCP-1 expression appeared in the puerarin group at hour 6 after reperfusion, expressed markedly at hour 12 after reperfusion, reached the peak at hour 24, and the expression at hours 6 and 24 was lower than that in the expression of ischemia/reperfusion retina at the same period (0.724±0.002,1.421±0.231; 2.159±0.307,3.682±0.157;P < 0.05). There was no significant MCP-1 expression in the sham operation group. 2 From hour 6 after ischemia/reperfusion, amplitude of wave of electroretimogram a and b was higher in the puerarin group than that in the ischemia/ reperfusion group at each time phase, but lower than in the sham operation group (P < 0.01). Conclusion: The puerarin can inhibit the expression of MCP-1 in rat's ischemia-reperfusion injured retina, which may be associated with the protection on ischemia-repesfusion injured retina.
