Abstract:Objective The purpose of this study was to explore the application of synchrotron radiation X-ray phase-contrast imaging in morphology. Methods The dissected kidneys of rats, liver of pig and the lungs of mice, femoral heads of rabbits were imaged using the synchrotron radiation X-ray phase-contrast imaging at the Beijing Synchrotron Radiation Facility(BSRF). Histologic slides of the specimens were made and compared with the phase-contrast imaging of the specimens. Results The straight collecting ducts and papillary tubules were visible clearly in dissected kidney of rats, the central veins and the boundaries for liver lobular of pig could also be observed by diffraction enhanced image(DEI). The bronchia tree and the image created by multiple layers of alveoli in the mouse lung could be displayed in DEI too. The tubules were visible clearly down to approximately 30?μm in these images. With in-line holography imaging, the articular cartilage and trabecular bone of the femoral head could also be imaged. Conclusion Not only the pattern structure but also the relationship between the pattern structure and the surrounding tissue which could not be observed by optical microscope and conventional radiography could be identified by the synchrotron radiation X-ray phase contrast imaging. The synchrotron radiation X-ray phase contrast imaging was suitable for morphologic study.
