Abstract:Fluorescence immunoassay technology is widely recognized as the most sensitive detection technology, now widely used in clinical testing. The research of quantitative analyzer for fluorescence immune chromatography strips is of great significance. In this thesis, a quantitative analyzer for fluorescence immune chromatography strips has realized, based on photoelectric detection principle and immunochromatographic. The system uses LPC1788 and MSP430 as processors. Using some algorithms such as low-pass filtering, least square method and area method to analyze the detected waveform and eliminate noise with high accuracy. In our experiment, the minimum detection concentration is at several mIU/ml levels, the relative standard deviation is less than 2% and it has good linear response characteristics in a concentration range from 0.5125 to 952mIU/ml while the correlation coefficient R2 is 0.981. The instrument has a good prospect.

Abstract:In this article, a kind of microfluidic method based on MEMS technology combined with gold immunochromatographic assay (GICA) is developed and discussed. Compared to the traditional GICA, this method supplies us convenient, multi-channel, in-parallel, low cost and similar efficiency approach in the fields of alpha-fetopro-tei (AFP)detection. Firstly, we improved the adhesion between the model material SU-8 and Silicon wafer, optimized approaches of the fabrication of the SU-8 model systematically, and fabricate the PDMS micro fluid chip with good reproduction successfully. Secondly, Surface modification and antibody immobilization methods with the GICA on the PDMS micro fluid analysis chip are studied, we choose the PDMS material and transfer GICA to the PDMS micro fluid chip successfully after researching the antibody immobilization efficiency of different materials utilized in fabrication of the micro fluid chip. In order to improve the reaction efficiency of the immobilized antibody, we studied the characteristics of micro fluid without the gas drive, and the fluid velocity control in our design; we also design structure of grove to strengthen the ability of immobilizing the antibody. The stimulation of the structure shows that it achieves great improvement and experiments prove the design is feasible.
Antigen Detection Based on Background Fluorescence Quenching Immunochromatographic Assay.

Abstract:Gold immunochromatographic assay (GICA) has been around for quite a while, but it is qualitative in the vast majority of applications. A fast, simple and quantitative GICA is in call for better medicine. In the current study, we have established a novel, quantitative GICA based on fluorescence quenching and nitrocellulose membrane background signals, called background fluorescence quenching immunochromatographic assay (bFQICA). Using model analyte alpha-fetoprotein (AFP), the present study assessed the performance of bFQICA in numerous assay aspects. With serial dilutions of the international AFP standard, standard curves for the calculation of AFP concentration were successfully established. At 10 and 100ngmL(-1) of the international AFP standard, the assay variability was defined with a coefficient of variance at 10.4% and 15.2%, respectively. For samples with extended range of AFP levels, bFQICA was able to detect AFP at as low as 1ngmL(-1). Fluorescence in bFQICA strips stayed constant over months. A good correlation between the results from bFQICA and from a well-established Roche electrochemiluminescence immunoassay was observed in 27 serum samples (r=0.98, p<0.001). In conclusion, our study has demonstrated distinctive features of bFQICA over conventional GICA, including utilization of a unique fluorescence ratio between nitrocellulose membrane background and specific signals (F1/F2) to ensure accurate measurements, combined qualitative and quantitative capabilities, and exceptionally high sensitivity for detection of very low levels of antigens. All of these features could make bFQICA attractive as a model for antigen-antibody complex based GICA, and could promote bFQICA to a broad range of applications for investigation of a variety of diseases.
Facile Fabrication of Highly Quantum Dot/AuNP-Loaded Tags for a Dual-Modal Colorimetric/Reversed Ratiometric Fluorescence Immunochromatographic Assay

Abstract:Developing an easily-prepared, sensitive, and accurate point-of-need immunochromatographic assay (ICA) is significant in food safety screening, clinical diagnosis, and environmental monitoring. However, the current single-modal ICAs are limited in certain instinct drawbacks that restrict analytical performances. Herein, we introduce an ultrasensitive dual-modal colorimetric/reversed ratiometric fluorescence ICA based on facilely prepared immunoprobes with a high loading capacity of red quantum dots and AuNPs. By smartly integrating these red-colored/fluorescent signal probes with an immobilized green quantum dot antigen on the test lines, discrete "turn-on" visual inspection and reversed ratiometric quantification via a portable smartphone-based analyzer were accomplished. As an application, this method was employed to detect 11 phosphodiesterase-5 inhibitors in health foods with ultralow detection limits (0.0028-0.045 ng/mL), high repeatability (coefficient of variations of 0.3-1.91%), and reasonable accuracy (recoveries of 86.6-107%). The proposed method was further validated by the authorized liquid chromatography with tandem mass spectrometry method in actual sample detection. This new assay format can be extended to ultrasensitive flexible detection of other food contaminants, environmental pollutants, or tumor biomarkers within minutes, and it just requires simply prepared signal reporters, easy-to-operate procedures, and a low-cost miniaturized analyzer.
Development of a colloidal gold-based immunochromatographic test strip for the rapid, on-site detection of Pseudomonas aeruginosa in clinical samples

Abstract:BACKGROUND: Current procedures for the detection of Pseudomonas aeruginosa require sophisticated equipment, skilled technicians, and a great deal of time. Immunochromatography assays (ICA) are simple and rapid diagnostic procedures that can be performed and interpreted on the spot or at the bedside without a machine.METHODS: We developed a rapid, 1-step immunochromatographic test strip that is well suited to the on-site detection of P. aeruginosa with high sensitivity and specificity. In brief, a monoclonal antibody targeting the outer membrane protein F (OprF) of P. aeruginosa, 3C3B5, was conjugated to colloidal gold and used as a detection antibody. An OprF polyclonal antibody was developed as the capture antibody. Eighty-three clinical samples were examined for P. aeruginosa by rapid 1-step ICA and compared with Multiplex-polymerase chain reaction (M-PCR).RESULTS: The detection limit of this method is 5 × 10(5) CFU/ml for P. aeruginosa and 10 ng/ml for the OprF protein. The immunochromatographic strip test demonstrated a slightly lower sensitivity (84.8%), but a similar specificity (100%), to multi-PCR, which is an accurate method for the detection of P. aeruginosa in the laboratory. We observed no cross-reactivity with non-P. aeruginosa bacterial microbes. The detection of P. aeruginosa by the ICA strip can be completed within 5-10 min and is at least 10-fold faster than M-PCR.CONCLUSIONS: The ICA test strip developed in this study has proved to be a rapid, simple, effective and economical method for the detection of P. aeruginosa infection in clinical samples. To our knowledge, this is the first report of an ICA method being used to detect P. aeruginosa.

Abstract:Rapid, quantitative detection of tumor markers with high sensitivity and specificity is critical to clinical diagnosis and treatment of cancer. We describe here a novel portable fluorescent biosensor that integrates quantum dot (QD) with an immunochromatography test strip (ICTS) and a home-made test strip reader for detection of tumor markers in human serum. Alpha fetoprotein (AFP), which is valuable for diagnosis of primary hepatic carcinoma, is used as a model tumor marker to demonstrate the performance of the proposed immunosensor. The principle of this sensor is on the basis of a sandwich immunoreaction that was performed on an ICTS. The fluorescence intensity of captured QD labels on the test line and control line served as signals was determined by the home-made test strip reader. The strong luminescence and robust photostability of QDs combined with the promising advantages of an ICTS and sensitive detection with the test strip reader result in good performance. Under optimal conditions, this biosensor is capable of detecting as low as 1ng/mL AFP standard analyte in 10min with only 50μL sample volume. Furthermore, 1000 clinical human serum samples were tested by both the QD-based ICTS and a commercial electrochemiluminescence immunoassay AFP kit simultaneously to estimate the sensitivity, specificity and concordance of the assays. Results showed high consistency except for 24 false positive cases (false positive rate 3.92%) and 17 false negative cases (false negative rate 4.38%); the error rate was 4.10% in all. This demonstrates that the QD-based ICTS is capable of rapid, sensitive, and quantitative detection of AFP and shows a great promise for point-of-care testing of other tumor markers.

Abstract:Quantitative detection of fluorescence immunoassay is widely used in clinical detection. For this reason, through a reasonable design of optoelectronic circuits real-time detection system, designing a set of fully functional fluorescence immunochromatographic quantitative detection system is necessary, which integrates a large number of peripheral modules, including fluorescence signal collecting, motion control, fluorescence signal processing data, quantitative curve fitting and testing results. After the improved threshold wavelet denoising through experiments, the system performance is very good, quantitative results are accurate. So, the system of fluorescence analysis has a vital significance.

Abstract:An immunochromatographic assay (ICA) based on red silica nanoparticles (red-SiNPs) was developed for quantitative detection of hepatitis B surface antigen (HBsAg) in serum. Red SiNPs have unique optical properties and remarkable stability compared to red latex microspheres. Red-SiNPs functionalized with anti-HBsAg mAb1 were employed as detection immuno-probes. The sandwich-type immunoreaction structures were formed on ICA strips, and the accumulation of red SiNPs-mAb1 on the test line produced characteristic red color bands, enabling visual detection of HBsAg without instrumentation. Quantitative detection was performed by researching the relationship between the HBsAg concentration and the corresponding peak area of the gray value of the test line with a camera and image analysis software Image J. Under optimal conditions, the visual limit of detection (vLOD) of the strip for the qualitative detection of HBsAg is 1 ng mL(-1), which is lower than that of the routinely used red latex microsphere-based ICA. In the quantitative detection, the peak area of the gray value of the test line and the HBsAg concentration show a good linearity when the HBsAg concentration is in the range of 5-500 ng mL(-1). The calibration equation was y = 370.6118 + 9.6673x (R2 = 0.9962) and the limit of detection (LOD) of the method was 0.97 ng mL(-1) (S/N = 3). Our proposed method provides promising and versatile opportunities for the rapid and quantitative screening of other proteins at relatively sensitive levels by combining a colored label with ICA.
Ultra-sensitive Fluorescent Immunoassay Based on Bright AIE Nanoparticle for Biomarkers Detection and Quantification

Abstract:Fluorescence immunoassay (FIA) is one of the most immunoassay diagnosis methods in clinic for biomarker quantification in recent years. The development of reliable FIA strategy with high-sensitivity, large-rang and outstanding efficiency for biomarkers rapid diagnosis, especially in complicated samples, is still a great challenge. Herein, an ultra-sensitive FIA combined with magnetic separation was established to achieve efficient point-of-care diagnosis in quantitative behavior based on bright aggregation-induced emission nanoparticles (AIE NPs). By utilizing swelling method, AIE NPs with controllable size, relatively high luminescence efficiency and large Stokes shift were prepared, moreover, the obtained NPs also exhibit narrow size distribution and well-defined spherical morphology. Stability studies prove that AIE NPs not only possess quite high photostability, but also present good resistance towards harsh acid-alkaline steadily. By using AIE NPs as the reporter, a reliable and efficient FIA sensor was successfully constructed. The precise detection of C-reaction protein (CRP) antigen was acquired with the linear range of 0.5 ngmL(-1) similar to 1000 ngmL(-1) and the limit of detection (LOD) of 0.35 ngmL(-1), which is comparative with the conventional chemiluminescence immunoassays. Moreover, the whole detection operation could be accomplished within 10 min, which is relatively rapid and desirable for point-of-care diagnosis. In addition, this strategy also presented excellent specificity towards CRP antigens among various inflammatory antigens. These results confirm that AIE NPs-based FIA strategy provides an ideal and reliable approach for precise and quantitive biosensing of in vitro diagnostics.

Abstract:Rapid and sensitive detection of hazards in clinical, food, and environmental samples is of great importance in the field of public health. Immunochromatographic assay (ICA), as a rapid diagnostics platform, is currently the most attractive point-of-care sensor due to its rapidness, user-friendly, and low-cost. Although, a variety of "head-to-toe" improvement strategies have sprung up to provide ICA with superior analytical performance in recent years, their respective strengths and weaknesses are difficult to interconnect and integrate, hampering their widespread deployment in ensuring public health. In this review, the momentous strategies to address the inherent limitations of ICA covering increasing sensitivity, enabling quantitative detection of analytes, and multiplexing are presented. Particularly, significant attention is focused on synergistically breaking the bottleneck of practical feasibility of ICA. Furthermore, the unique superiority and latest applications in clinical diagnosis, food safety, and environmental monitoring, as well as perspectives in the development process of the new generation of ICA are also elaborated. Eventually, the main challenges and future-oriented innovations in this field have also received attention with the profound development of the new generation of ICA in public health.

Abstract:Alpha-fetoprotein (AFP), commonly employed for early diagnosis of liver cancer, serves as a biomarker for cancer screening and diagnosis. Combining the high sensitivity and specificity of fluorescence immunoassay (FIA), developing a low-cost and efficient immunoassay system for AFP detection holds significant importance in disease diagnosis. In this work, we developed a miniaturized oblique laser-induced fluorescence (LIF) immunoassay system, coupled with a microfluidic PMMA/paper hybrid chip, for rapid detection of AFP. The system employed an avalanche photodiode (APD) as the detector, and implemented multi-level filtering in the excitation light channel using the dichroic mirror and optical trap. At first, we employed the Savitzky–Golay filter and baseline off-set elimination methods to denoise and normalize the original data. Then the cutoff frequency of the low-pass filter and the reverse voltage of the APD were optimized to enhance the detection sensitivity of the system. Furthermore, the effect of laser power on the fluorescence excitation efficiency was investigated, and the sampling time during the scanning process was optimized. Finally, a four-parameter logistic (4PL) model was utilized to establish the concentration–response equation for AFP. The system was capable of detecting concentrations of AFP standard solution within the range of 1–500 ng/mL, with a detection limit of 0.8 ng/mL. The entire immunoassay process could be completed within 15 min. It has an excellent potential for applications in low-cost portable diagnostic instruments for the rapid detection of biomarkers.

Abstract:Semiconductor quantum dots (QDs) have drawn considerable attention since its emergence because they have high brightness and a long luminous time. Our aim is to develop a new assay based on QDs to determine Alpha fetal protein (AFP) by immunofluorescence assay (IFA). Two different monoclonal antibodies of anti-AFP were labeled by QDs and the magnetic beads. After combination with AFP antigen, the fluorescence intensity of QDs was detected by the fluorospectrophotometer. The method was linear to 400 ng/mL AFP with a detection limit of 1.2 ng/mL. The intra-assay imprecision of two different level serums were 5.81% and 5.53%; the inter-assay imprecision values were 6.26% and 6.15%. The proposed method may be applied in the clinical lab.

Abstract:Because of their simplicity, rapidity, and cost-effectiveness, immunochromatographic strips (ICTs) have been widely used as an effective tool in various fields. However, typical strips for the preliminary screening suffer from limited detection sensitivity, particularly in biomarker detection with trace concentration. Herein, to tackle this challenge, we integrated homemade gold-decorated Fe3O4 nanoparticles (Au/Fe3O4 NPs) with flexible strips, exploring the excellent peroxidase-like activity of this labeled material, and then enhancing the detection sensitivity via signal amplification. The limit of detection (LOD) of the strips is as low as 0.05 mIU mL(-1) when human chorionic gonadotropin (hCG) is as a biomarker model, which is 500 times lower than that of the traditional color-based strip. Overall, our results demonstrated the potential for Au/Fe3O4 NP based-ICTs for the rapid detection of the biomarker in an instrument-free and point-of-care testing format.

Abstract:Potentiometric immunoassays have been utilized for the quantitative detection of alpha-fetoprotein (AFP) in hepatocellular carcinoma, but most of them involve low sensitivity and enzyme labels, and thus are unfavorable for routine use. In this work, we report the proof-of-concept of a sensitive and powerful ion-selective potentiometric sensing method for AFP detection with an in situ amplified signal readout. This potentiometric immunoassay mainly contains a silver nanocluster-functionalized single-stranded DNA (AgNC-DNA), a short DNA primer and two antibodies. A sandwich-type immunoreaction is employed for AFP determination on an anti-AFP capture antibody-coated microplate using a biotinylated human AFP secondary antibody. Coupling with a typical biotin-avidin system, the biotinylated DNA initiator strands are conjugated on the microplate in the presence of AFP to induce the rolling cycle amplification (RCA) reaction, followed by AgNC-DNA hybridization. Upon addition of HNO3, the hybridized AgNCs are dissolved into numerous Ag(I) ions, which can be readily determined on a portable handheld silver-ion selective electrode (Ag-ISE). Under optimal conditions, the electrode potential increases with an increase in AFP concentration and exhibits a good linear range of 0.01-100 ng mL-1 at a detection limit of 7.9 pg mL-1. Moreover, the Ag-ISE-based potentiometric immune assay also shows good reproducibility, high specificity and long-term storage stability. Importantly, 18 human serum specimens containing the AFP analyte are screened using the potentiometric immunoassay, giving well-matched experimental results relative to the referenced enzyme-linked immunosorbent assay (ELISA) method.
An integrated immunochromatographic device for C-reactive protein detection using hierarchical dendritic gold nanostructure films

Abstract:Immunochromatographic test strips typically consist of sample pad, conjugate pad, nitrocellulose membrane, and absorbent pad. Even minute variations in the assembly of these components can lead to inconsistent sample-reagent interactions, thereby reducing reproducibility. In addition, the nitrocellulose membrane is susceptible to damage during assembly and handling. To address this issue, we propose to replace the sample pad, conjugate pad, and nitrocellulose membrane with hierarchical dendritic gold nanostructure (HD-nanoAu) films to develop a compact integrated immunochromatographic strip. The strip uses quantum dots as a background fluorescence signal and employs fluorescence quenching to detect C-reactive protein (CRP) in human serum. A 5.9 μm thick HD-nanoAu film was electrodeposited on an ITO conductive glass by the constant potential method. The wicking kinetics of the HD-nanoAu film was thoroughly investigated, and the results indicated that the film exhibited favorable wicking properties, with a wicking coefficient of 0.72 μm ms-0.5. The immunochromatographic device was fabricated by etching three interconnected rings on HD-nanoAu/ITO to designate sample/conjugate (S/C), test (T), and control (C) regions. The S/C region was immobilized with mouse anti-human CRP antibody (Ab1) labeled with gold nanoparticles (AuNPs), while the T region was preloaded with polystyrene microspheres decorated with CdSe@ZnS quantum dots (QDs) as background fluorescent material, followed by mouse anti-human CRP antibody (Ab2). The C region was immobilized with goat anti-mouse IgG antibody. After the samples were added to the S/C region, the excellent wicking properties of the HD-nanoAu film facilitated the lateral flow of the CRP-containing sample toward the T and C regions after binding to AuNPs labeled with CRP Ab1. In the T region, CRP-AuNPs-Ab1 formed sandwich immunocomplexes with Ab2, and the fluorescence of QDs was quenched by AuNPs. The ratio of fluorescence intensity in the T region to that in the C region was used to quantify CRP. The T/C fluorescence intensity ratio was negatively correlated with the CRP concentration in the range of 26.67-853.33 ng mL-1 (corresponding to 300-fold diluted human serum), with a correlation coefficient (R2) of 0.98. The limit of detection was 15.0 ng mL-1 (corresponding to 300-fold diluted human serum), and the range of relative standard deviation: 4.48-5.31%, with a recovery rate of 98.22-108.33%. Common interfering substances did not cause significant interference, and the range of relative standard deviation: 1.96-5.51%. This device integrates multiple components of conventional immunochromatographic strips onto a single HD-nanoAu film, resulting in a more compact structure that improves the reproducibility and robustness of detection, making it promising for point-of-care testing applications.
Rapid and Quantitative Detection of C-reactive Protein Using Quantum Dots and Immunochromatographic Test Strips

Abstract:Background: Rapid immunochromatographic tests can detect disease markers in 10-15 minutes, which facilitates clinical diagnosis and treatment programs. However, most immunochromatographic tests employ gold nanoparticles as reporters, and these have only moderate sensitivity and act as qualitative methods for analyzing high biomarker concentrations.Methods: In this study, we introduce quantum dots (QDs) as fluorescent probes and immunochromatographic strips to develop quantitative fluorescence point-of-care tests (QF-POCT) to analyze C-reactive protein (CRP) levels. Goat anti-rabbit IgG and rabbit IgG were used as control antibodies, and mouse monoclonal CRP antibody pairs were used for disease marker detection. One monoclonal CRP antibody was conjugated with QDs and served as a signal antibody, and the other monoclonal CRP antibody was dispensed onto the nitrocellulose membrane and served as a capturing antibody. In the presence of CRP, the fluorescence intensity of the monoclonal antibody-CRP- monoclonal antibody sandwich complex captured on the nitrocellulose membrane was determined using the fluorescence strip reader.Results: QF-POCT assays could quantitatively analyze the concentration of CRP in 15 minutes had a detection limit of 0.25 mg/L, and had a wide detection linearity range (0.5-300 mg/L). The intra-assay and interassay coefficients of variation were 8.95% and 9.86% at 0.5 mg/L, 6.47% and 8.66% at 10 mg/L, and 6.81% and 9.10% at 60 mg/L, respectively. In a comparison between clinical samples, the results of this QD-based assay of CRP levels were significantly correlated with those of an Immulite 2000 assay (R=0.993, P<0.001).Conclusion: Our results demonstrated that the QD-based immunochromatographic test is a rapid, sensitive, accurate, and quantitative method for the detection of disease biomarkers.

Abstract:As the most widely used disposable paper-based diagnostic tool in the world, immunochromatographic test strips (ICTS) have occupied more and more positions in the field of rapid diagnosis due to their ease of operation and affordability. Therefore, the development of an easily prepared, sensitive, and accurate signal reporter is of great significance for the detection of some low-abundance biomarkers in clinical diagnosis. Herein, Mg/Fe layered double hydroxide nanoflowers (MF NFs) were selected as adsorption templates and sulfhydryl-functionalized, followed by one-step loading of hydrophobic CdSe/ZnS quantum dots in the organic phase via a metal-thiol covalent bond. After coating the reporter with branched polyethyleneimine (PEI), a novel ICTS fluorescent reporter was prepared. The modification of PEI not only improved the hydrophilicity of MF@CdSe/ZnS NFs but also introduced amino functional groups on the surface of the reporter for subsequent conjugation with antibodies. X-ray photoelectron spectroscopy, UV-vis absorption, X-ray diffraction, fluorescence spectroscopy, and infrared spectroscopy were used to characterize the composition of MF@CdSe/ZnS NFs. Under the optimal experimental conditions, the detection range of MF@CdSe/ZnS@PEI-ICTS for the model analyte HCG was 0.1–500 mIU/mL, and the limit of detection (LOD) reached was 0.1 mIU/mL. The potential for practical application was validated by the detection of HCG in spiked healthy human serum, showing overall recoveries between 90.48 and 116.1% with coefficients of variation that ranged from 3.66 to 12.91%.

Abstract:A novel photoelectrochemical immunosensor based on TiO2/CdS hybrid modified electrode was developed. The TiO2/CdS hybrid modified electrode was obtained by alternately dipping the TiO2 modified indium–tin oxide (ITO) electrode into the [Cd(NH3)4]2+ and S2− solution repeatedly. Compared with the routine method using Cd2+ solution for CdS deposition, the as obtained TiO2/CdS electrode showed enhanced photocurrent intensity with fewer coating times. After the ITO/TiO2/CdS electrode was coated with chitosan (CS), α-fetoprotein (AFP) antibodies were covalently conjugated on the surface of the electrode. Thus, a label-free photoelectrochemical immunosensor for the detection of AFP was developed by monitoring the changes in the photocurrent signals of the electrode resulting from the immunoreaction. The immunosensor displayed a linear response to AFP in the ranges from 50pg/mL to 50ng/mL with a relatively low detection limit of 40pg/ml. The photoelectrochemical results for the detection of AFP in five human sera showed acceptable accuracy. The method is simple, sensitive and specific. Moreover, the studied immunosensor possessed acceptable reproducibility and storage stability. The proposed methodology was potentially attractive for clinical immunoassay.

Abstract:Simplifying the procedure of immunoassay is still a challenge due to problems such as multiple washing processes, complicated chemical modification and expensive cost. In this study, we developed a portable centrifugal microchip fluorescence immunoassay for washing-free, rapid, quantitative and point-of-care (POC) detection of protein. The designed microchip was fabricated by polycarbonate and assembled by double-sided adhesive tape using injecting molding with high scalability and low cost. The centrifugal strategy is capable of washing-out the bio-fluid and improving signal-to-noise ratio. Matrix nano-spotting method was employed to facilitate satisfactory immunological binding sites with the advantage of high capture efficiency and reproducibility. The proposed approach was capable of sensitively detecting procalcitonin (PCT) with a wide dynamic ranging from 0.10 ng/mL to 70.00 ng/mL within 10 min. Furthermore, this novel integrated diagnostic tool was successfully applied to detect PCT in 101 clinical samples with good consistency with Roche’s method, indicating its attractive practical application capability. With favorable simplicity, rapidity, low cost and excellent analytical performance, our method holds great promise for POC diagnostics of proteins.
