Abstract:In this paper, we describe the preparation of human serum albumin-coated magnetic particles of about 200nm in diameter with narrow size distribution radiolabeled with 188Re for the purpose of magnetically targeted therapy. The optimum radiolabeling conditions are: SnCl2·2H2O 8mg/ml, citric acid 20mg/ml, vitamin C 8mg/ml, labeling volume 500μl and a reaction time of 3h. The stability of the radiolabeled particles is suitable for in vivo study.

Abstract:Amino-functionalized superparamagnetic iron oxide nanoparticles (SPION) were synthesized by coprecipitation method. The particles were characterized by X-ray diffraction (XRD), vibrating sample magnetometer (VSM), scanning electron micrographs (SEM), transmission electron micrographs (TEM) and atomic force micrographs (AFM). The size of the modified particles varied in the range 10–15 nm and did not change significantly after modification. Hepama-1, an excellent humanized monoclonal antibody directed against liver cancer, was conjugated to the SPION to prepare immuno-magnetic nanoparticles (IMN). A direct labeling method was employed to radiolabel IMN with rhenium-188. The radiolabeling efficiency was about 90% with good in vitro stability. 188 Re labeled IMN could markedly kill SMMC-7721 liver cancer cells. Such SPION might be very useful for bio-magnetically targeted radiotherapy in liver cancer treatment.

Abstract:Superparamagnetic poly(acrylamide) magnetic nanogels produced via photochemical method have been developed. After Hoffmann degradation of carbonyl, the nanogels with amino groups, or poly(acrylamide-vinyl amine) magnetic nanogels, were also obtained. And the magnetic nanogels were further modified by methoxy poly(ethylene glycol) (MPEG) for higher dispersibility and stability. The MPEG-modified magnetic nanogels were characterized by X-ray diffraction (XRD), photo correlation spectroscopy (PCS) and scanning electron microscopy (SEM), respectively. The MPEG-modified magnetic nanogels were labeled by 188Re radiopharmaceuticals and intravenously injected into tails of mice in the presence and absence of a 0.5 T external magnetic field targeted on the bellies. The radioactivity distribution was monitored in vivo. In the absence of magnetic field, the radioactivity was mainly distributed in liver, spleen, kidney, stomach and lung. In the presence of the magnetic field, the radioactivity was mainly accumulated on the targeted point, verifying the magnetically targeted character.

Abstract:Objective To prepare ~(188)Re labeled monoclonal antibody(Herceptin)-coated magnetic uanoparticles for tumor targeting and to study its biodistribution in mice.Methods Herceptin and histidine were covalently linked to the amine group upon silica-coated magnetic nanoparticles modified by N-[3-(tri- methyoxysilyl)propyl]-ethylenediamine using glutaraldebyde method.The Heceptin-coated magnetic nano- particles and Herceptin were radiolabeled with ~(188)Re by a direct labelling method,whereas the histidine-coa- ted magnetic nanoparticles was radiolabeled with ~(188)Re using fac-[~(188)Re(CO)_3(H_2O)_3]~+as a precursor. The labelling efficiency and immunoreactivity as well as labelling stability were determined.Also,the bio- distribution of ~(188)Re-magnetic and ~(188)Re-Herceptin-magnetic nanoparticles were observed in mice.Results Herceptin-coated magnetic nanoparticles was characterized by transmission electron microscope(TEM)with diameter about 60 nm,while histidine-coated magnetic nanoparticles about 30 nm.The labeling efficiency for ~(188)Re-Herceptin,~(188)Re-magnetic nanoparticles and ~(188)Re-Herceptin-magnetic nanoparticles were all> 90% and had a better stability in vitro.The immunoreaetivity of Herceptiu linked to magnetic nanoparticles was still high.The biodistribution in mice was shown that ~(188)Re-magnetic nanoparticles and ~(188)Re-Herceptin- magnetic nanoparticles had higher radioactivity levels in blood.Magnetic nanoparticles with diameter of 30 or 60 nm had a long half-life in blood stream and were accumulated in liver.Conclusion The efficiency and stability of labelling Herceptin-coated magnetic nanoparticles and labeling magnetic nanoparticles with ~(188)Re are suitable for in vivo study in tumor-bearing nude mice models.

Abstract:Magnetite nanoparticles coated with silica were synthesized and characterized by electron microscopic methods (SEM and TEM) and crystal structure analysis (XRD). After surface modification with an amino silane coupling agent, N-[3-(trimethyoxysilyl) propyl]-ethylenediamine (SG-Si900), histidine was covalently linked to the amine group using glutaraldehyde as cross-linker. The magnetic nanoparticles were then radiolabeled with 188Re with a labeling yield of 91.4±0.3% and good stability in vitro. The labeling mechanism is suggested as fac-[188Re(CO)3(H2O)3]+ core complexed with imidazolyl groups.

Abstract:Histidine is covalently linked to the amine group upon silica-coated magnetic nanoparticles modified by N--ethylenediamine using glutaraldehyde as cross-linker. The magnetic nanoparticles are then radiolabeled with ~(188)Re using fac-~+ as a precursor with a good labeling yield of 91.4%±0.3% under optimized conditions (reaction temperature 70 ℃, reaction time 40 min, and buffer solution pH 6.6) and obtained above 80% radioactivity retention of radiolabeled magnetic nanoparticles incubated with wild vibration in calf serum under 37 ℃ within 3 days.

Abstract:Objective:To prepare HSA-coated magnetite nanoparticles labeled with rhenium- 188(188 Re) for the purpose of regional target therapy. Methods: The optimum labeling conditions are that: SnCl2· H2O8 mg/ml, citric acid 20 mg/ml, Vc 8 mg/ml, reaction volume 500 μl and reaction time 3 hours. Results: The labeling efficiency is about 90%. The labeling stability ( 37℃ in PBS at pH 7.4) is about 98% rhenium- 188 is still bound 7 h later, 95 % 24 h later. Conclusions: The efficiency and stability of labeling HAS- coated magnetite nanoparticles with 188Re is suitable for in vivo study.

Abstract:Magnetic targeting is one of the most promising approaches for improving the targeting efficiency by which magnetic drug carriers are directed using external magnetic fields to reach their targets. As a natural magnetic nanoparticle (MNP) of biological origin, the magnetosome is a special "organelle" formed by biomineralization in magnetotactic bacteria (MTB) and is essential for MTB magnetic navigation to respond to geomagnetic fields. The magnetic targeting of magnetosomes, however, can be hindered by the aggregation and precipitation of magnetosomes in water and biological fluid environments due to the strong magnetic attraction between particles. In this study, we constructed a magnetosome-like nanoreactor by introducing MTB Mms6 protein into a reverse micelle system. MNPs synthesized by thermal decomposition exhibit the same crystal morphology and magnetism (high saturation magnetization and low coercivity) as natural magnetosomes but have a smaller particle size. The DSPE-mPEG-coated magnetosome-like MNPs exhibit good monodispersion, penetrating the lesion area of a tumor mouse model to achieve magnetic enrichment by an order of magnitude more than in the control groups, demonstrating great prospects for biomedical magnetic targeting applications.

Abstract:To demonstrate their applications in biological and medical fields such as in immunoassays, magnetic separation of cells or proteins, drug or gene delivery, and magnetic resonance imaging, the template-free syntheses of water-soluble and surface functionalized magnetic nanomaterials have become essential and are challenging. Herein, we developed a facile one-pot template-free method for the preparation of amine-functionalized magnetite nanoparticles and hollow nanospheres by using FeCl(3)(.)6H(2)O as single iron source. These magnetic nanomaterials were characterized by TEM, SEM, XRD, and FTIR technologies. Their magnetic properties were also studied by using a superconducting quantum interference device (SQUID) magnetometer at room temperature. Then the amine-functionalized magnetite nanoparticles were applied to immunoassays and magnetic resonance imaging in live mice.

Abstract:Objective:To prepare Fe3O4 magnetite nanoparticles and HSA- coated magnetite particles for the purpose of regional target therapy. Methods: To adopt partial reduction method to prepare the Fe3O4 nanoparticles:100 ml 0.01 mol/L Na2SO3 was added dropwise into 100 ml 0.06 mol/LFeCl3 solution under nitrogen gas flow.10%(V%) ammonia was added dropwise with rapid stirring until pH of the reaction solution reach 8. Heated at 70℃ with water bath for 15 min. Preparation of HAS- coated magnetite particles:To use microemulsion approach,with oleic acid as oil phase, mixture of HSA and magnetite solution as water phase and sp- 85 as emulsion agent.Results: Fe3O4 magnetite particles with about 10 nm in diameter and X - ray power diffraction show that the nanoparticles is γ- Fe2O3. HAS- coated Fe3O4 magnetite nanoparticles is characterized by TEM with diameter no more than 200 nm. Conclusions: Fe3O4 magnetite nanoparticles and HSA - coated magnetite particles is suitable for researching of regional target therapy.

Abstract:Uniform magnetic nanoparticle-loaded polymer nanospheres with different loading contents of manganese ferrite nanoparticles were successfully synthesized using a flexible emulsion process. The MnFe2O4-loaded polymer nanospheres displayed an excellent dispersibility in both water and phosphate buffer saline. The effect of loading ratio and size of MnFe2O4 nanoparticles within the nanospheres on the specific absorption rate (SAR) under an alternating magnetic field was investigated. Our results indicate that a large size (here 18 nm) and a low loading ratio are preferable for a high SAR. For a smaller particle size (6 nm), the low loading ratio did not result in an enhancement of the SAR value, while a very low SAR value is expected for 6 nm. In addition, the SAR of low-content MnFe2O4 (18 nm)-loaded polymer nanospheres in the agarose gel which is simulated for in vivo environment is the highest among the samples and does not change substantially in physiological environments. This differs largely from the behaviour of singly dispersed nanoparticles. Our results have paved the way for the design of MnFe2O4-loaded polymer nanospheres as magnetic hyperthermia agents for in vivo bio-applications.

Abstract:Highly hydrophilic, uniform, superparamagnetic and nontoxic maltotrionic acid (MA)-coated magnetite nano-particles (MAM) were prepared and characterized by TEM, DLS, XRD and VSM. MA was used to improve the biocompatibility, monodispersity and non-specific intracellular uptake of nanoparticles. Folic acid (FA) was subsequently conjugated to the MAM to preferentially target KB cells (cancer cells) that have folate receptors expressed on their surfaces and to facilitate nanoparticles in their transit across the cell membrane. Finally, fluorescence isothiocyanate (FITC) was added to the nanoparticles to visualize the nanoparticle internalization into KB cells. After the cells were cultured in a media containing the MAM and MAM-folate conjugate (FAMAM), the results of fluorescence and confocal microscopy showed that both types of nanoparticles were internalized into the cells. Nevertheless, the amount of FAMAM uptake was higher than that of MAM. This result indicated that nanoparticles modified with MA and FA could be used to facilitate the nanoparticle uptake to specific KB cells (cancer cells) for molecular imaging.

Abstract:Aqueous-phase Fe3O4 nanoparticles (NPs), with a size of 9.8 +/- 3.0 nm, are directly synthesized by a modified iron salt coprecipitation process, stabilized by the synergistic effect of a combination of poly(vinylpyrrolidone) (PVP), trisodium citrate (TSC), and maleic anhydride (MAH). These NPs show good aqueous-phase dispersion stability and excellent superparamagnetic properties with a saturation magnetization of 48.0 emu/g at a low magnetic field of 0.5 T. Their biocompatibility and metabolism for application in magnetic resonance reticuloendothelial system examinations are investigated by using rabbits as animal models for the determination of optimized dosage and diagnosis time for the enhanced imaging contrast for the early diagnosis of diseases. Results indicate that a significant darkening effect on the liver epithelial net lymph tissue can be observed in less than 30 min, with about 20% reduction of the spin-spin relaxation time of T-2 after injection. A metabolism study on the NPs indicates that they show little toxicity to the living organs examined, and finally enter into the spleen without obvious retention in any related organs after recycling for more than 2 d.
Smart Multifunctional Nanostructure for Targeted Cancer Chemotherapy and Magnetic Resonance Imaging.

Abstract:Targeted chemotherapy and magnetic resonance imaging of cancer cells in vitro has been achieved using a smart multifunctional nanostructure (SMN) constructed from a porous hollow magnetite nanoparticle (PHMNP), a heterobifunctional PEG ligand, and an aptamer. The PHMNPs were prepared through a three-step reaction and loaded with the anticancer drug doxorubicin while being functionalized with PEG ligands. Targeting aptamers were then introduced by reaction with the PEG ligands. The pores of the PHMNPs are stable at physiological pH, but they are subject to acid etching. Specific binding and uptake of the SMN to the target cancer cells induced by aptamers was observed. In addition, multiple aptamers on the surface of one single SMN led to enhanced binding and uptake to target cancer cells due to the multivalent effect. Upon reaching the lysosomes of target cancer cells through receptor-mediated endocytosis, the relatively low lysosomal pH level resulted in corrosion of the PHMNP pores, facilitating the release of doxorubicin to kill the target cancer cells. In addition, the potential of using SMN for magnetic resonance imaging was also investigated.

Abstract:A rapid ligand-exchange method was developed to transfer high quality hydrophobic magnetite nanocrystals into water-soluble NPs by using protocatechuic acid as a ligand via homogenous reaction. After ligand exchange, the magnetite nanocrystals not only exhibited outstanding stability in water, but also maintained high crystallinity and saturation magnetization. Cell viability experiments demonstrated good biocompatibility of the NPs. For 12 nm magnetite nanoparticles (NPs), the small hydrodynamic size of 14 nm enabled a high T-1 relaxivity of 17.8 mM(-1) s(-1) while high saturation magnetization of 77.8 emu g(-1) enabled the NPs to exhibit a high T-2 relaxivity of 220 mM(-1) s(-1) in MRI phantom experiments. In vivo MR imaging experiments further confirmed that the NPs were eminent T-1 and T-2 contrast agents. Moreover, the high quality NPs can be used as excellent magnetic heating agents under an alternating magnetic field. With all those features, including multimodality imaging and magnetic hyperthermia, the NPs can be used as single compound multifunctional agents for various biomedical applications, especially for cancer diagnosis and therapy.

Abstract:A novel bioconjugation of amino saline capped Fe3O4 magnetic nanoparticles (MNPs) with bovine serum albumin (BSA) was developed by applying glutaraldehyde as activator. Briefly, Fe3O4 MNs were synthesized by the chemical co-precipitation method. Surface modification of the prepared MNPs was performed by employing amino saline as the coating agent. Glutaraldehyde was further applied as an activation agent through which BSA was conjugated to the amino-coated MNPs. The structure of the BSA-MNs was confirmed by FTIR analysis. Physico-chemical characterizations of the BSA-MNPs, such as surface morphology, surface charge and magnetic properties were investigated by Transmission Electron Microscopy (TEM), zeta-Potential and Vibrating Sample Magnetometer (VSM), etc. Magnetic inductive heating characteristics of the BSA-MNPs were analyzed by exposing the MNPs suspension (magnetic fluid) under alternative magnetic field (AMF). The results demonstrate that BSA was successfully conjugated with amino-coated MNs mediated through glutaraldehyde activation. The nanoparticles were spherical shaped with approximately 10 nm diameter. Possessing ideal magnetic inductive heating characteristics, which can generate very rapid and efficient heating while upon AMF exposure, BSA-MNPs can be applied as a novel candidature for magnetic nanothermotherapy for cancer treatment. In vitro cytotoxicity study on the human hepatocellular liver carcinoma cells (HepG-2) indicates that BSA-MNP is an efficient agent for cancer nanothermotherapy with satisfied biocompatibility, as rare cytotoxicity was observed in the absence of AMF. Moreover, our investigation provides a methodology for fabrication protein conjugated MNPs, for instance monoclonal antibody conjugated MNPs for targeting cancer nanothermotherapy.

Abstract:Multifunctional magnet–fluorescent nanocomposites are widely applied in biomedical applications. Incorporating biocompatible quantum dots with highly ferrimagnetic magnetic nanoparticles into one nanoplatform for achieving efficient magnetic hyperthermia therapy (MHT) is very important. Herein, we reported an amphiphilic block copolymer with a flowable hydrophobic chain to encapsulate highly ferrimagnetic magnetic nanoparticles and ZnS/InP quantum dots via a facile self-assembly method. The obtained ferrimagnetic fluorescent micelle (FMFM) exhibited a uniform diameter of about 180 nm. In stark contrast, larger aggregation (400 nm in diameter) inevitably occurred using common poly( D , L -lactide) (PLA)-based amphiphilic block copolymer with a rigid hydrophobic chain, which was readily cleared by the reticuloendothelial system (RES). The flowable FMFM exhibited long-term colloidal stability within one month and desired fluorescent stability within 84 h. Benefiting from the high ferrimagnetism, the FMFM revealed excellent magnetic heating effect and magnetic resonance imaging capability. With accurate manipulation under an external magnetic field, FMFM realized in vitro enhanced fluorescence imaging sensitivity and accumulation efficiency at the tumor region, achieving in vitro and vivo improved MHT efficacy.

Abstract:Superparamagnetic Fe3O4 nanoparticles (NPs)-based hyperthermia is a promising non-invasive approach for cancer therapy. However, the heat transfer efficiency of Fe3O4 NPs is relative low, which hinders their practical clinical applications. Therefore, it is promising to improve the magnetic hyperthermia efficiency by exploring the higher performance magnetic NPs-based hybrid nanostructures. In the current study, it presents a straightforward in situ reduction method for the shape-controlled preparation of magnetite (Fe3O4) silver (Ag) hybrid NPs designed as magnetic hyperthermia heat mediators. The magnetite silver hybrid NPs with core-shell (Fe3O4@Ag) or heteromer (Fe3O4-Ag) structures exhibited a higher biocompatibility with SMMC-7721 cells and L02 cells than the individual Ag NPs. Importantly, in the magnetic hyperthermia, with the exposure to alternating current magnetic field, the Fe3O4@Ag and Fe3O4-Ag hybrid NPs indicated much better tumor suppression effect against SMMC-7721 cells than the individual Fe3O4 NPs in vitro and in vivo. These results demonstrate that the hybridisation of Fe3O4 and Ag NPs could greatly enhance the magnetic hyperthermia efficiency of Fe3O4 NPs. Therefore, the Fe3O4@Ag and Fe3O4-Ag hybrid NPs can be used to be as high performance magnetic hyperthermia mediators based on a simple and effective preparation approach.
