Factors affecting root growth and metabolite production in Salvia miltiorrhiza adventitious root cultures

Abstract:Aim Root growth and secondary metabolite formation in Salvia miltiorrhiza adventitious root cultures were affect ed by factors including sucrose concentration, plant growth regulators, nitrogen sources and ultrasound. Methods. Under various sucrose concentrations, the maximum biomass and production of tanshinone IIA and protocatechuic aldehyde were obtained with 60 g center dot L-1, 20 g center dot L-1, 30 g center dot L-1 sucrose in culture medium, respectively. Plant growth regulator combinations of IBA and KT had different effects in this culture. Results. The results indicated that the highest growth rate, ie. 4.17 by dry weight, a relatively high productivity of tanshinone E[A and protocatechuic aldehyde, being 0.19 mg center dot g(-1) and 0.32 mg center dot g(-1), respectively, were reached with combinations of the growth regulators of IBA 2-KT 0, EBA 2-KT 1 and EBA 0-KT 0.5, respectively. When adding various ratios of NH4+/NO3- to the medium and keeping the total initial nitrogen source to 60 mmol center dot L-1, a relatively high biomass and maximum secondary metabolite production were achieved with nitrate as the sole nitrogen source. Conclusion. As for ultrasound-induction, higher power level, Le. 300 W and 500 W for long exposed time (3-4 min) inhibited root growth.
