Tooth repair-related transgene study: an experimental observation of founders of pTet-on-CX transgenic mouse at G0 generation

Abstract:Aim: To establish a transgenic mouse line which is a part of tetracycline-responsive regulating transgene system in order to facilitate large scale of transgene investigations. Methods: The experiment was completed in the Shanghai Research Center of Biomodel Organism from June to October 2002. The transgenic vector pTet-on-CX was constructed by substituting cβ-actin promoter for NSE promoter in pTet-on-NSE, and after linearized by XhoI, it was microinjected into the mouse male pronucleus of the zygotes and embryos were implanted into uterine tube of recipient pseudopregnant mice. The tail DNA of pups was tested by PCR and Southern Blotting. Results: Totally 603 injected fertilized eggs survived, and gave birth to 64 pups after implantation of eggs. Six of the 64 pups carrying the transgene were positive. The passage and establishment of the transgenic line are under progress in the pups with positive transgene. Conclusion: The founders of pTet-on-CX transgenic mouse at G0 generation were obtained successfully by microinjection, which provides a basis for large scale of transgene investigations (including tooth repair-related genes, such as dentin sialophosphoprotein gene).
