Effects of L-arginine on expression of soluble vascular cell adhesion molecule 1 in atherosclerosis rabbits

Abstract:Aim: To investigate the effects of L-arginine on soluble vascular cell adhesion molecule 1 of atherosclerosis rabbits, and explore relation between level of soluble vascular cell adhesion molecule 1 and expression of local vascular cell adhesion molecule 1. Methods: 1 The e xperiment was conducted at the laboratory of Department of Cardiology, Shanghai First People's Hospital from September 2003 to April 2004. Twenty-four healthy New Zealand male rabbits were selected, and randomly divided into 3 groups: model group, L-arginine group and control group with 8 rabbits every group. 2 Rabbits of model group and L-arginine group were fed with hyper-cholestorol diet 10 days before and then balloon denudation in right iliac artery. All the animals were killed at the 30th day after procedure. From the 2nd day after operation, those in the L-arginine group were given 1 g L-arginine every day through drinking. Those in the control group were treated with common feed. 3 Level of soluble vascular cell adhesion molecule 1 in serum was detected with double antibody sandwich enzyme-linked immunoadsordent assay before experiment, before operation and 30 days after operation. Expression of mRNA of vascular cell adhesion molecule 1 from tissue freezing section was tested with in situ hybridization. Protein was extracted to perform immunoblot test. 4 Difference of measure data was compared with t-test. Results: Twenty-four rabbits wer e all involved in the result analysis. 1 Expression of mRNA of vascular cell adhesion molecule 1: After 30 days of injury of blood vessel, it became more significantly in the model group, and a few in the L-arginine group, and showed negative in the control group. 2 Expression of protein of vascular cell adhesion molecule 1: After injury of blood vessel for 30 days, it increased in the model group, and less in the L-arginine group than that in the model group, and few in the control group. 3 Level of soluble vascular cell adhesion molecule 1 in serum: There was no difference at three time points in every group (P > 0.05). After operation for 30 days, it was higher markedly in the model group than that before operation (P < 0.05); It was higher remarkably in the model group than that in the control group and L-arginine group (P < 0.01, 0.05), while it had insignificant difference between L-arginine group and control group (P > 0.05). Conclusion: Orally taken L-arginine can lower serum level of sol uble vascular cell adhesion molecule 1 in atherosclerosis rabbits, and its change reflects local expression of soluble vascular cell adhesion molecule 1 in saccule injured artery.
