Abstract:The Suzuki cross-coupling reaction is a powerful and versatile method for the generation of unsymmetrical biaryls from arylboronic acids and aryl halides in a single step. However, the reaction is usually performed in the presence of Pd catalyst along with phosphine ligand, which sometimes creates practical problems because organophosphines tend to be expensive, poisonous, and air sensitive. Recently, phosphine-free ligands, such as nitrogen or sulfur-containing ligands and N-heterocyclic carbenes have been applied in Suzuki reaction effectively. Nevertheless, the syntheses of these ligands or related catalysts are often time consuming, difficult, and/or require the use of expensive starting materials, which significantly limits their industrial application. In addition, the use of water as solvent for Suzuki reaction has received much attention due to environmental, economical and safe reasons. In 1994 the phosphine-free Suzuki reaction involving water as the cosolvent was reported by Wallow. Later, Badone and Bussolari independently reported that Pd(OAc)2 in combination with Bu4NBr could catalyze Suzuki coupling in water to provide the products in good yields. However, the Suzuki reaction in the absence of additives is greatly limited by the substrate solubility and reactivity in aqueous media. Therefore, the development of economical and efficient catalytic systems in aqueous phase remains a highly desirable goal in Suzuki coupling reaction. In recent years, the development of immoblized transition metal catalysts which can be more easily separated from organic products and recycled has also attracted a great deal of attention in Suzuki reaction, such as palladium supported on carbon, silica, zeolites and polymers. Resins are one class of polymeric supports originating from organic precursors which are widely employed as catalyst carriers and also in solid-phase organic synthesis and resin-supported palladium complexes having high activity and selectivity are currently attracting a great interest due to their easy separation and recovery. Furthermore, although imidazoles, oxazoline and tetrazole have been evaluated as nonphosphine ligands for Pd-catalyzed C―C bond formation reactions, there has been no report on 4-amino-1,2,4triazole resin(abbreviated as 4-ATR), which contains a functional group of triazole and can coordinate directly to metal ions via the nitrogen atom, as a ligand for the coupling reaction so far. Herein lie a simple synthesis of palladium complex immobilized on 4-amino-1,2,4-triazole resin, designated 4-ATR-Pd(0) and its successful application to the Suzuki reaction of various aryl or heteroaryl halides and phenylboronic acid in the absence of phosphine ligand at a low load catalyst in aqueous phase(Scheme 1).
