Abstract:Three 20(S)-protopanaxatriol-type saponins, ginsenoside-Rg(1) (1), notoginsenoside-R-1 (2), and ginsenoside-Re (3), were transformed by the fungus Absidia coerulea (AS 3.3389). Compound 1 was converted into five metabolites, ginsenoside-Rh-4 (4), 3 beta, 2 beta, 25-trihydroxydammar-(E)-20(22)-ene-6-O-beta-D-glucopyranoside (5), 20(S)-ginsenoside-Rh-1 (6), 20(R)-ginsenoside- Rh-1(7), and a mixture of 25-hydroxy-20(S)-ginsenoside-Rh-1 and its C-20(R) epimer (8). Compound 2 was converted into 10 metabolites, 20(S)-notoginsenoside-R-2 (9), 20(R)-notoginsenoside-R-2 (10), 3 beta, 12 beta, 25-trihydroxydammar( E)-20(22)-ene-6-O-beta-D-xylopyranosyl-(1 -> 2)-beta-D-glucopyranoside (11), 3 beta, 12 beta-dihydroxydammar-(E)-20(22), 24-diene-6- O-beta-D-xylopyranosyl-(1 -> 2)-beta-D-glucopyranoside (12), 3 beta, 12 beta, 20,25-tetrahydroxydammaran-6-O-beta-D-xylopyranosyl(1 -> 2)-beta- D-glucopyranoside (13), and compounds 4-8. Abstract:The biotransformation of the major saponins in Panax notoginseng, including the ginsenosides Rg1, Rh1, Rb1, and Re, by endophytes isolated from P. notoginseng was studied. One hundred and thirty‐six endophytes were isolated and screened for their biotransformational abilities. The results showed that five of the tested endophytes were able to transform these saponins. These five strains were identified based on their ITS or 16S rDNA sequences, which revealed that they belonged to the genera Fusarium, Nodulisporium, Brevundimonas, and Bacillus genera. Ten transformed products were isolated and identified, including a new compound 6‐O‐[α‐L‐rhamnopyranosyl‐(1→2)‐β‐D‐glucopyranosyl]‐20‐O‐β‐D‐glucopyranosyldammarane‐3,6,12,20,24,25‐hexaol (3), and nine known compounds, compound K (1), ginsenoside F2 (2), vinaginsenoside R13 (4), vinaginsenoside R22 (5), pseudo‐ginsenoside RT4 (6), (20S)‐protopanaxatriol (7), ginsenoside Rg1 (8), vinaginsenoside R15 (9), and (20S)‐3‐O‐β‐D‐glucopyranosyl‐6‐O‐β‐D‐glucopyranosylprotopanaxatriol (10). This is the first study on the biotransformation of chemical components in P. notoginseng by endophytes isolated from the same plant.

Abstract:A new minor oleanane-type saponin, ginsenoside-R(OA) (1), has been isolated from the extract of Kaixin-San, a prescription composed of Panax ginseng and other medicinal herbs, together with 15 known ginsenosides. The structure of 1 was established as 3-O-beta-D-glucopyranosyl-(1-->2)-beta-D-glucuronopyranosyl oleanolic acid 28-O-beta-D-glucopyranosyl-(1-->6)-beta-D-glucopyranoside by chemical and spectroscopic evidence. In addition, the presence of ginsenoside-R(OA) in ginseng was confirmed by HPLC-ESI-MS.
Cloning and heterologous expression of UDP-glycosyltransferase genes from Bacillus subtilis and its application in the glycosylation of ginsenoside Rh1

Abstract:Bacillus subtilis CCTCC AB 2012913 can transform ginsenoside Rh1 to 3-O-β-D-glucopyranosyl-6-O-β-D-glucopyranosyl-20(S)-protopanaxatriol. Based on its genome sequence, strain B. subtilis 168 contains three UDP-glycosyltransferase genes. Here, we cloned the three UDP-glycosyltransferase genes (ydhE1, yojK1 and yjiC1) from B. subtilis CCTCC AB 2012913 and expressed in Escherichia coli BL21 (DE3) with His-tag. The crude enzyme extracts were assayed, respectively, for their activities to transform ginsenoside Rh1. Extracts containing enzymes YojK1 and YjiC1 could use ginsenoside Rh1 as a substrate to produce 3-O-β-D-glucopyranosyl-6-O-β-D-glucopyranosyl-20(S)-protopanaxatriol, which had an additional glucopyranosyl linked with C-3 over the substrate. Enzyme YjiC1 was purified by affinity chromatography on Ni-NTA His Binding resin. The molecular mass of purified YjiC1 was c. 47 kDa as determined by SDS-PAGE. This is the first report of an in vitro biotransformation of ginsenoside Rh1 to 3-O-β-D-glucopyranosyl-6-O-β-D-glucopyranosyl-20(S)-protopanaxatriol using the recombinant UDP-glycosyltransferase. Significance and impact of the study: The Chinese traditional medicinal plant Panax is reported to have multiple health benefits. Its main active ingredient is saponin, and different saponins have different activity spectrum. In the study, three UDP-glycosyltransferase genes, ydhE1, yojK1 and yjiC1, were cloned from Bacillus subtilis CCTCC AB2012913 and the three genes were expressed in Escherichia coli BL21 (DE3). The enzyme YjiC1 was purified and converted ginsenoside Rh1 to 3-O-β-D-glucopyranosyl-6-O-β-D-glucopyranosyl-20(S)-protopanaxatriol in vitro. The compound is the first saponin possessing β-glucopyranosyl at both C-3 and C-6 sites. We showed that the in vitro biotransformation was effective, and the reaction condition was easy to control. Our research suggests that a diversity of saponins could be generated through efficient and directed enzymatic biotransformation.

Abstract:Ganoderma sp. contains high amounts of diverse triterpenoids; however, few triterpenoid saponins could be isolated from the medicinal fungus. To produce novel Ganoderma triterpenoid saponins, biotransformation-guided purification (BGP) process was applied to a commercial Ganoderma extract. The commercial Ganoderma extract was partially separated into three fractions by preparative high-performance liquid chromatography, and the separated fractions were then directly biotransformed by a Bacillus glycosyltransferase (BsUGT489). One of the biotransformed products could be further purified and identified as a novel saponin: ganoderic acid C2 (GAC2)-3-O-β-glucoside by nucleic magnetic resonance (NMR) and mass spectral analyses. Based on the structure of the saponin, the predicted precursor should be the GAC2, which was confirmed to be biotransformed into four saponins, GAC2-3-O-β-glucoside, GAC2-3,15-O-β-diglucoside and two unknown GAC2 monoglucosides, revealed by NMR and mass spectral analyses. GAC2-3-O-β-glucoside and GAC2-3,15-O-β-diglucoside possessed 17-fold and 200-fold higher aqueous solubility than that of GAC2, respectively. In addition, GAC2-3-O-β-glucoside retained the most anti-α-glucosidase activity of GAC2 and was comparable with that of the anti-diabetes drug (acarbose). The present study showed that the BGP process is an efficient strategy to survey novel and bioactive molecules from crude extracts of natural products.

Abstract:An Aspergillus niger strain was isolated from the soil around ginseng fruit. In vitro enzyme assays showed that this strain had the ability to transform total ginsenosides (TGS) into several new products. In a further biochemical study, a beta-glucosidase gene isolated from this strain, bgl1, was expressed in Saccharomyces cerevisiae. His-tagged BGL1 protein (similar to 170 kD) showed the ability to transform ginsenoside Rf into Rh-1.

Abstract:Abstract Aim To study of structural modification in steroidal saponin by biotransformation technology. Methods Structural modification was carried out of glycosyl residues of dichotomin at C-3 position by Pectinex BE XXL (PBX). Cytotoxicity against HL-60 cells of all products was investigated. Results The PBX could hydrolyze the glycosyl residues of dichotomin one-by-one to produce nine transformed compounds. The transformed product (mixture) and four secondary spirostanosides among them exhibited cytotoxicity. Compounds P5, P6 and P7 with α-1, 2 end-rhamnosyl residues showed a potential cytotoxic effect on HL-60 cells; P8 with α-1, 4 end-rhamnosyl residue had a weak effect on HL-60 cells. Dichotomin, secondary furostanosides and aglycone were not cytotoxic. Conclusion PBX has various hydrolyzation to the glycosidic bonds of dichotomin. Nine obtained transformed compounds widen steroidal saponin library. Meanwhile, screening activity affords data for further pharmacological study.
Preparation of 20 (S)-protopanaxadiol saponins and 20 (S)-ginsenoside- Rh2 from 20 (S)-protopanaxadiol saponins

Abstract:OBJECTIVE: To prepare 20(S)-protopanaxadiol saponins and change them into 20 (S)-ginsenoside Rh2 having antitumor activity. METHODS: From the crude saponins of the stems and leaves of American ginseng grown in China, we prepared 20(S)-protopanaxadiol saponins by extraction and 20(S)-ginsenosiok- Rh2 by the alkaline hydrolysis of protopanaxadiol saponins and column chromatography. RESULTS: The yield of 20(S)-protopanaxadiol saponins was 41.5%, and the yield of 20(S)-ginsenoside-Rh2 was 9.64% from the 20(S)- protopanaxadiol saponins. CONCLUSION: The method is simple and has a high yield.

Abstract:The strains isolated from 7 year-old ginseng root under the forest were tested to transform ginseng saponins from the ginseng root,and found the strain GS 1-33 could transform ginseng saponin from ginseng root into rare ginsenoside C-K and Rh1 effectively.The following optimum conditions were determined.On water medium with pH value3.0,the maximum yields of ginsenoside C-K and Rh1 were 14% and 25%.Found rare ginsenoside C-K was the products of ginsenoside Rb1,Rc and Rd by biotransformation,and Rh1 of ginsenoside Rg1 and Re.

Abstract:A new triterpenoid saponin containing five sugars was isolated from the ethanolic root extract of Dipsacus japonicus Miq., and its structure was established as 3-O-[beta-D-glucopyranosyl (1-->4)] [alpha-L-rhamnopyransoyl (1 --> 3)]-beta-D-glucopyranosyl (1-->3)-alpha-L-rhamnopyranosyl (1-->2)-alpha-L-arabinopyranosyl-oleanoic acid. The sites of glycosylation and the sequence of sugars in the glycoside can be determined unambiguously and total assignment of severely overlapping proton resonance of sugar residues were achieved by a combined use of the 1D-SEMDY and NOE difference spectroscopy in rotating frame techniques, without having recourse to chemical degration or modification. The results showed that these new NMR techniques are very effective and convenient for the structure determination and spectral assignment of this class of compounds.

Abstract:A novel ginsenoside-hydrolyzing strain was isolated from ginseng-cultivation soil in Changbai Mountain (China). The strain was identified as Cellulosimicrobium sp. 21 by 16S rDNA sequencing. Using the beta-glucosidases secreted from Cellulosimicrobium sp. 21, protopanaxatriol-type ginsenoside Re was converted to the highly active neuroprotective molecule (S)-Rg2 by removal of the C-20-glucopyranosyl residue. The alpha-L-rhamnopyranosyl-(1 2)-beta-D-glucopyranose at the C-6 position of Rg2 was not further attacked by Cellulosimicrobium sp. 21, so the transformation shows high specificity. To simplify the transformation and product-preparation process, a simple and efficient transformation system was developed in a phosphate buffer system instead of organic media. The optimum conditions for transforming ginsenoside Re into Rg2 by Cellulosimicrobium sp. 21 were determined through single-factor experiments and response surface methodology. Under the optimized conditions: transformation buffer, 50 mM phosphate buffer, at pH: 7.00; temperature: 27.6 degrees C; substrate concentration: 0.50 mg/ml; biotransformation period: 12 h; the biotransformation efficiency reached 89.8% (molar ratio) in 2-L reaction system. This simple biotransformation with high specificity and efficiency has potential for use in Rg2 preparation in the pharmaceutical industry.

Abstract:Protopanaxatriol saponins obtained with AB-8 macroporous resin mainly consisted of ginsenosides Rg(1) and Re. A novel mono-ester of ginsenoside-Rh-1 (ginsenoside-ORh1) was synthesized through further enzymatic hydrolysis and octanoyl chloride modifications. A 53% yield was obtained by a facile synthetic method. The structures were identified on the basis of 1D-NMR and 2D-NMR, as well as ESI-TOF-MS mass spectroscopic analyses. The isolated and synthetic compounds were applied in an anti-tumor bioassay, in which ginsenoside ORh1 showed moderate effects on Murine H22 Hepatoma Cells.
