Abstract:Five unique metabolites, xyloketals A (1), B (2), C (3), D (4), and E (5), and the known 6 were isolated from mangrove fungus Xylaria sp. (no. 2508), obtained from the South China Sea. The structures of these compounds were elucidated by spectroscopic and X-ray diffraction experiments. Xyloketal A is a ketal compound with a C-3 symmetry and xyloketals B-E are its analogues. It was found that xytoketal C slowly rearranged to xytoketal B in DMSO-d(6) Solution at room temperature. Xyloketal A exhibited the activity of inhibiting acetylcholine esterase.

Abstract:In recent decades, technological advantages have allowed scientists to isolate medicinal compounds from marine organisms that exhibit unique structure and bioactivity. The mangrove fungus Xylaria sp. from the South China Sea is rich in metabolites and produces a potent therapeutic compound, xyloketal B. Since its isolation in 2001, xyloketal B has been extensively studied in a wide variety of cell types and in vitro and in vivo disease models. Xyloketal B and its derivatives exhibit cytoprotective effects in cardiovascular and neurodegenerative diseases by reducing oxidative stress, regulating the apoptosis pathway, maintaining ionic balance, mitigating inflammatory responses, and preventing protein aggregation. Xyloketal B has also shown to alleviate lipid accumulation in a non-alcoholic fatty liver disease model. Moreover, xyloketal B treatment induces glioblastoma cell death. This review summarizes our current understanding of xyloketal B in various disease models.

Abstract:The metabolites of the mangrove fungus Eutypa sp. (#424) from the South China Sea have been studied. A new compound eutypoid A (1), which is a derivative of alpha,beta-unsaturated gamma-lactone, has been isolated from the mycelium. Its structure has been elucidated by spectroscopic methods, mainly 2D NMR techniques. This kind of compounds possess diverse pharmacologically activities, such as inhibitors of cyclin-dependent kinases. The oily liquid from the mycelium is analyzed with GC / MS, the contents of unsaturated fatty acid esters are 57% of total oily liquid.

Abstract:Marine-derived fungi have been reported to have great potential to produce structurally unique metabolites. Our investigation on secondary metabolites from marine-derived fungi resulted in the isolation of seven new polyketides (phomopsiketones D–G (1–4) and letendronols A–C (5–7)) as well as one known xylarinol (8) in the cultural broth of Letendraea sp. Their structures and absolute configurations were elucidated using a set of spectroscopic and chemical methods, including HRESIMS, NMR, single-crystal X-ray diffraction, ECD calculation, and a modified version of Mosher’s method. Compound 2 showed weak inhibition against nitric oxide production in lipopolysaccaride-activated macrophages with an IC50 value of 86 μM.

Abstract:Two new lactones, 1 and 2, together with five known compounds, 3-7, were isolated from the marine mangrove fungus Xylaria sp. BL321. Their structures were determined by comprehensive analysis of their MS and NMR spectroscopic data. The absolute configurations of 1 and 2 were established on the basis of electronic circular dichroism calculations. It was found that the exocyclic double bond of 1 rearranged into a cyclic double bond to form a new crystal compound (1a) in diluted NaOH solution. Compound 3 was isolated for the first time as a natural product; its absolute configuration was determined by single-crystal X-ray crystallography. Compounds 4-7 displayed cytotoxicity against human breast cancer cell lines MCF-7 and MDA-MB-435, while compounds 1- 3 were inactive (IC(50) > 50 µM).

Abstract:Objective: To obtain structurally novel and bioactive secondary metabolites from the marine fungus Xylariaceae sp. SCSIO41212. Methods: Compounds were purified by comprehensive chromatography methods of silica gel column, Sephadex LH-20, Ostade-cylsilane (ODS) and semi-preparative HPLC, and their structures were elucidated by means of physical and chemical properties, magnefic resonance imaging, spectroscopic analysis, and comparison with the reported literatures. Results: Chemical investigation of the fungus Xylariaceae sp. SCSIO41212 has led to the isolation of two new compounds, xylaolide B (1) and xylaolide C (2), together with nine known compounds (3–11). Their structures were determined on the basis of detailed analysis of the 1D and 2D NMR as well as MS data. Compounds 1–6 were evaluated for their antiviral, and cytotoxic activities, respectively. Compound 1 showed weak cytotoxicities against K562, MCF-7 and SGC7901 respectively. Conclusion: This study enriches the diversity of secondary metabolites of marine-derived fungi and provides an important reference for novel promising drug leads research.

The compounds were isolated by column chromatography and identified on the basia of physic-chemical constants and spectral analysis. Abstract:A new isocoumarin glycoside, 3R-(+)-5-O-[6'-O-acetyl]-α-D-glucopyranosyl-5-hydroxymellein (1), and a new phenylethanol glycoside, (−)-phenylethyl-8-O-α-L-rhamno-pyranoside (2), were isolated from the ethyl acetate extract of the fungus Xylaria sp. cfcc 87468, together with five known steroids, β-sitosterol (3), stigmast-4-en-3-one (4), ergosterol (5), (22E)-cholesta-4,6,8(14),22-tetraen-3-one (6), and 4α-methyl- ergosta-8(14),24(28)-dien-3β-ol (7). The structures of compounds 1 and 2 were elucidated by MS, extensive 1D and 2D NMR spectroscopy, and the circular dichroism (CD) spectroscopy.

Abstract:One new metabolite 3,7-dimethyl-9-(-2,2,5,5-tetramethyl-1,3-dioxolan-4-yl)nona-1,6-dien-3-ol, together with nine known compounds, were isolated from the strain Xylaria sp., an endophytic fungus of Taxus mairei. Their structures were deduced from 1D and 2D NMR data. In vitro cytotoxicity and antibacterial activity of these compounds were evaluated. Some of them exhibited substantial activity.

Abstract:OBJECTIVE:The secondary metabolites of the fungus ZZF13 isolated from the leaves of the mangrove sample Kandelia candel in Zhanjiang and Guignardia sp. 4382 isolated from bark of Kandelia candel (endophyte) of Mai Po, Hong Kong were studied.METHODS:The compounds were isolated by siliga gel, and their structures were identified by IR, MS and NMR.RESULTS:Four compounds were isolated from the culture of this strain. Their structures were identified as Bacillpsporin C (1), 5-carboxymellein (2), 5-methylmellein (3) and 1-(2,6-dihydroxyphenyl) butanone (4).CONCLUSION:The compounds 2 - 4 are isolated from the Guignardia sp. of Marine fungi for the first time.

Abstract:new xanthone derivative, 8-hydroxy-3-methyl-9-oxo-9 H -xanthene-1-carboxylic acid methyl ether ( 1 ), was isolated from the co-culture broth of two mangrove fungi (strain No. K38 and E33) isolated from the South China Sea coast. The structure of 1 was determined by comprehensive spectroscopic methods, especially 2D NMR techniques. Primary bioassays showed that compound 1 has inhibitory activity against five microorganisms, including Gloeasporium musae and Peronophthora cichoralearum etc.

Abstract:Four novel tetranortriterpenoids, xylogranatins A-D (1-4), with an unusual 9,10-seco skeleton were isolated from the seeds of a Chinese marine mangrove Xylocarpus granatum. Their structures were determined by spectroscopic and chemical means. Xylogranatin A (1) featured by a unique 1,9-oxygen bridge was confirmed by single-crystal X-ray diffraction, and xylogranatin D (4) with an unprecedented skeleton of C-30-C-9 linkage was postulated biogenetically from 3 via an alpha-hydroxyl ketone rearrangement and was chemically mimicked.
