Abstract:The algorithmic complexity, the approximate entropy and the power spectrum of rat electroencephalogram (EEG) under different stages of anesthesia were calculated and analyzed. For synchronous EEGs, both complexity values decreased significantly with the development of anesthesia depth from light to deep. Although powers of synchronous EEG in high frequency bands of theta, alpha and beta gradually decreased, its total power and its power in delta band did not decrease until the deepest anesthesia. For de-synchronous EEGs, the complexity values did not change much with the development of anesthesia depth, and after the anesthesia was deep enough the de-synchronous EEGs would not appear any more, which indicated that the activities in the midbrain reticular formation had been inhibited. Powers of desynchronous EEG in high frequency bands of alpha and beta decreased gradually from light anesthesia stages. These findings suggest that more EEG features can be revealed by applying both methods of the complexity measures and the power spectral analysis.

Abstract:By complexity analysis index-fractal dimension,approximate entropy,complexity,heart rate variability signals obtained from44subjects ,ECG during anesthesia and during the state of being conscious are analyzed.The results show that there is an obvious chaos change of heart rate variability during anesthesia,the complexity(P0.05)and approximate entropy(P0.1)of heart rate variability reflecting narcotism are lower than them reflecting consciousness,while fractal dimension(P0.001)reflecting narcotism is higher,It hints that the nonlinear dynamics change of heart rate variability can be an important index for monitoring the depth of anesthesia.

Abstract:Objective: The study aims to investigate the relationship between amplitude modulation (AM) of EEG and anesthesia depth during general anesthesia. Methods: In this study, Holo-Hilbert spectrum analysis (HHSA) was used to decompose the multichannel EEG signals of 15 patients to obtain the spatial distribution of AM in the brain. Subsequently, HHSA was applied to the prefrontal EEG (Fp1) obtained during general anesthesia surgery in 15 and 34 patients, and the alpha-theta and alpha-delta regions of feature (ROFs) were defined in Holo-Hilbert spectrum (HHS) and three features were derived to quantify AM in ROFs. Results: During anesthetized phase, an anteriorization of the spatial distribution of AMs of alpha-carrier in brain was observed, as well as AMs of alpha-theta and alpha-delta in the EEG of Fp1. The total power ( P-total ), mean carrier frequency ( M-FC ) and mean amplitude frequency ( MFAM ) of AMs changed during different anesthesia states. Conclusion: HHSA can effectively analyze the cross-frequency coupling of EEG during anesthesia and the AM features may be applied to anesthesia monitoring. Significance: The study provides a new perspective for the characterization of brain states during general anesthesia, which is of great significance for exploring new features of anesthesia monitoring.
Monitoring Anesthetic Depth Based on Lattice Complexity of Electroencephalogram

Abstract:Based on fine-graining method and two partition methods(permutation partition and average partition), lattice complexity(LC)was used in EEG complexity analysis under anesthesia in this paper.By using the EEG of 40 patients who underwent general anesthesia with propofol,we calculated the Lattice complexity and lempel-ziv(LZ) complexity and compared Pearson's correlation coefficient between LC,LZ and BIS values. Results showed that during anesthesia induction period ,average partition with lattice complexity get better effect. While during anesthesia recovery period,permutation partition with lattice complexity get better result,and the Pearson's correlation coeffi?cient between LC and BIS values was 0.963 6. During the whole anesthesia period,Average partition with lattice com?plexity get better effect,and the the Pearson's correlation coefficient between LC and BIS values was 0.841 6. The fine-graining order take 3 can get good result. All these provide a new method for clinical anesthetic depth monitoring.
Study on the Evaluation Index of Depth of Anesthesia Awareness Based on Sample Entropy and Decision Tree

Abstract:Currently, monitoring system of awareness of the depth of anesthesia has been more and more widely used in clinical practices. The intelligent evaluation algorithm is the key technology of this type of equipment. On the basis of studies about changes of electroencephalography (EEG) features during anesthesia, a discussion about how to select reasonable EEG parameters and classification algorithm to monitor the depth of anesthesia has taken place. A scheme which combines time domain analysis, frequency domain analysis and the variability of EEG and decision tree as classifier and least squares to compute Depth of anesthesia Index (DOAI) is proposed in this paper. Using the EEG of 40 patients who underwent general anesthesia with propofol, and the classification and the score of the EEG annotated by anesthesiologist, we verified this scheme with experiments. Classification and scoring was based on a combination of modified observer assessment of alertness/sedation (MOAA/S), and the changes of EEG parameters of patients during anesthesia. Then we used the BIS index to testify the validation of the DOAI. Results showed that Pearson's correlation coefficient between the DOAI and the BIS over the test set was 0.89. It is demonstrated that the method is feasible and has good accuracy.

Abstract:Phase-amplitude coupling (PAC) plays an important role in anesthetic-induced unconsciousness. The delta-alpha PAC signature during anesthetic-induced unconsciousness is gradually becoming known; however, the frequency dependence and spatial characteristics of PAC are still unclear. Multi-channel electroencephalography (EEG) was performed during the loss and recovery phases of consciousness in patients undergoing general anesthesia using sevoflurane. First, a spectral analysis was used to investigate the power change of the different frequency bands in the EEG signals. Second, PAC comodulogram analysis was performed to confirm the frequencies of the PAC phase drivers. Finally, to investigate the spatial characteristics of PAC, a novel PAC network was constructed using within- and cross-lead PAC, and a K-means clustering algorithm was used to identify PAC network patterns. Our results show that, in addition to the delta-alpha PAC, unconsciousness induced by sevoflurane was accompanied by spatial non-uniform alpha-gamma PAC in the cortical network, and dynamic PAC patterns between the anterior and posterior brain were observed during the unconscious phase. The dynamic transition of PAC network patterns indicates that brain states under sevoflurane-induced unconsciousness emerge from the regulation of functional integration and segregation instantiated by delta-alpha and alpha-gamma PAC.
An integrated information theory index using multichannel EEG for evaluating various states of consciousness under anesthesia

Abstract:Background: The integrated information theory (IIT) of consciousness introduces a measure O to quantify consciousness in a physical system. Directly related to this, general anesthesia aims to induce reversible and safe loss of consciousness (LOC). We sought to propose an electroencephalogram (EEG)-based IIT index OEEG to evaluate various states of consciousness under general anesthesia.Methods: Based on the definition of mutual information, we estimated the OEEG by maximizing the integrated information under various time lags. We used the binning method to cut the nonGaussian EEG data for estimating mutual information. We tested two EEG databases collected from propofol-(n = 20) and sevoflurane-induced (n = 15) anesthesia, and especially, we compared the OEEG of drowsy (n = 7) and responsive participants (n = 13) under propofol anesthesia. We compared the effectiveness of OEEG with the estimated bispectral index (eBIS).Results: In all EEG frequency bands, we observed a negative correlation between OEEG and end-tidal sevoflurane concentration under sevoflurane-induced anesthesia (p < 0.001, BF10 > 6000). Under propofol-induced anesthesia, drowsy participants in moderate sedation (6.96 +/- 0.26(mean +/- SD)) showed decreased alpha-band OEEG compared with baseline (7.40 +/- 0.53, p = 0.016, BF10 = 3.58), no significant difference was observed for responsive participants. Oppositely, the responsive participants in moderate sedation (-5.32 +/- 0.38) showed decreased eBIS compared with baseline (-4.94 +/- 0.40, p = 0.03, BF10 = 2.41).Conclusions: These findings may enable monitors of the anesthetic state that can distinguish consciousness and unconsciousness rather than the changes of anesthetic concentrations. The alpha-band OEEG is promising for deriving the gold standard for depth of anesthesia monitoring.
A Study of Complexity and Power Spectrum of Cortical EEG and Hippocampal Potential in Rats under Different Behavioral States

Abstract:Objective. To study the complexity and the power spectrum of cortical EEG and hippocampal potential in rats under waking and sleep states. Method. Cortical EEG and hippocampal potential were collected by implanted electrodes in freely moving rats. Algorithmic complexity (Kc), approximate entropy (ApEn), power spectral density (PSD) and gravity frequency of PSD of the potential waves were calculated. Result. The complexity of hippocampal potential was higher than that of cortical EEG under every state. The complexity of cortical EEG was lowest under the state of non rapid eye movement (NREM) sleep. The complexity of hippocampal potential was highest under waking state. The total power of both potentials in 0.5- 30 Hz frequency band showed their highest values under NREM state. Conclusion. The values of Kc and ApEn are closely related to the distributions of PSD. When there are evident peaks in PSD, the complexities of signals will decrease. The complexities may be used to distinguish the difference between cortical EEG and hippocampal potential, or large differences between the same kind of potentials under different behavioral states.

Abstract:Background: Recent studies have demonstrated that changes in brain information processing during anesthetic-nduced loss of consciousness (LOC) might be influenced by phase-amplitude coupling (PAC) in electroencephalogram (EEG). However, most anesthesia research on PAC typically focuses on delta and alpha oscillations. Studies of spatial-frequency characteristics by PAC for EEG may yield additional insights into understanding the impaired information processing under anesthesia unconsciousness and provide potential improvements in anesthesia monitoring. Objective: Considering different frequency bands of EEG represent neural activities on different spatial scales, we hypothesized that functional coupling simultaneously appears in multiple frequency bands and specific brain regions during anesthesia unconsciousness. In this paper, PAC analysis on whole-brain EEG besides delta and alpha oscillations was investigated to understand the influence of multiple cross-frequency coordination coupling on information processing during the loss and recovery of consciousness. Method: EEG data from fifteen patients without cognitive diseases (7 males/8 females, aged 43.8 +/- 13.4 years, weighing 63.3 +/- 14.9 kilograms) undergoing lower limb surgery and sevoflurane anesthesia was recorded. To investigate the spatial-frequency characteristics of EEG source signals during loss and recovery of consciousness, the time-resolved PAC (tPAC) was calculated to reflect cross-frequency coordination in different frequency bands (delta, theta, alpha, beta, gamma) and different functional regions (Visual, Limbic, Dorsal attention, Ventral attention, Default, Somatomotor, Control, Salience networks). Furthermore, different patterns (peak-max and trough-max) of PAC were examined by constructing phase-amplitude histograms using phase bins to investigate the different information processing during LOC. The multivariate analysis of variance (MANOVA) and trend analysis were used for statistical analysis. Results: Theta-alpha and alpha-beta PAC were observed during sevoflurane-induced LOC, which significantly changed during loss and recovery of consciousness (E-4,E-70 = 16.553, p < 0.001 for theta-alpha PAC and F-4,F-70 = 12.446, p < 0.001 for alpha-beta PAC, MANOVA test). Simultaneously, PAC was distributed in specific functional regions, i.e., Visual, Limbic, Default, Somatomotor, etc. Furthermore, peak-max patterns of theta-alpha PAC were observed while alpha-beta PAC showed trough-max patterns and vice versa. Conclusion: Theta-alpha and alpha-beta PAC observed in specific brain regions represent information processing on multiple spatial scales, and the opposite patterns of PAC indicate opposite information processing on multiple spatial scales during LOC. Our study demonstrates the regulation of local-global information processing during sevoflurane-induced LOC. It suggests the utility of evaluating the balance of functional integration and segregation in monitoring anesthetized states.

Abstract:Wavelet entropy (WE), a new method of complexity measure for non-stationary signals, was used to investigate the dynamic features of rat EEGs under three vigilance states. The EEGs of the freely moving rats were recorded with implanted electrodes and were decomposed into four components of delta, theta, alpha and beta by using multi-resolution wavelet transform. Then, the wavelet entropy curves were calculated as a function of time. The results showed that there were significant differences among the average WEs of EEGs recorded under the vigilance states of waking, slow wave sleep (SWS) and rapid eye movement (REM) sleep. The changes of WE had different relationships with the four power components under different states. Moreover, there was evident rhythm in EEG WEs of SWS sleep for most experimental rats, which indicated a reciprocal relationship between slow waves and sleep spindles in the micro-states of SWS sleep. Therefore, WE can be used not only to distinguish the long-term changes in EEG complexity, but also to reveal the short-term changes in EEG micro-state.

Abstract:Electroenccephalogram (EEG) is well established for assessing the functional state of the brain. During the anesthesia, the brain activity level changed dramatically, so we could get the depth of anesthesia estimation from EEG recording. In previous research, most achievements were in the frequency-domain. In this paper, besides the frequency-domain features extraction, we have introduced the concept of “Entropy” as well as nonlinear feature. The results show that, with the deepening of anesthesia degree, approximate entropy (ApEn), Shannon entropy (SSE) and Lempel— Ziv complexity from EEG signal decrease gradually. Centre frequency towards to low frequency, total power shows a rising trend. Largest Lyapunov index also decreases, while the correlation dimension has no clear trend.
USING THE MULTI-RESOLUTION WAVELET DECOMPOSITIONTO ANALYZE THE SLEEP SPINDLE IN RAT EEG

Abstract:An algorithm was designed to automatically analyze the sleep spindles in rat sleep EEG based on the multi-resolution wavelet decomposition. This algorithm was used to evaluate the effects of the diazepam and the sleep deprivation on the sleep spindles of rat EEG. Firstly, a simulating EEG signal with sleep spindles was synthesized and used to select a more suitable wavelet function for decomposing sleep spindles from EEG. The results showed that the best result was obtained using Coiflet wavelet. Then, this wavelet function was used to establish the algorithm to extract the sleep spindles from EEG with a threshold method. The EEGs were recorded from freely moving rats with implanted electrodes following the applications of diazepam or following sleep deprivations. The results showed that the duration of sleep spindle was significantly increased in the diazepam sleep but not in the recovery sleep following sleep deprivation, however, the power of sleep spindle in the recovery sleep was significantly increased. Therefore, the method of wavelet decomposition can provide more information in detail on both time and frequency fields than the traditional power spectrum.

Analysis of Rat Electroencephalogram During Slow Wave Sleep and Transition Sleep Using Wavelet Transform.

Abstract:The dynamic features of rat EEGs collected during slow wave sleep (SWS) and transition sleep (TS) were investigated in both time and frequency domains using wavelet transform based on multi-resolution signal decomposition. EEGs of freely moving rats were recorded with implanted electrodes and then decomposed into four components of delta, theta, alpha and beta using wavelet transform. The power and power percentage of each component were calculated as functions of time. In SWS EEGs, the results showed that there existed as much as 26.2% +/- 7.7% time duration in which the delta power percentage was less than 50%. In addition, the powers of other three components in small delta EEGs were significantly larger than those in large delta EEGs. This result revealed a reciprocal relationship between delta oscillation and spindle oscillation. Comparatively, the conventional method of FFT based power spectrum could only show a delta power-dominating (70.6% +/- 6.4%) spectrum of SWS EEGs. In the non-stationary TS EEG, spindle and non-spindle segments were distinguished based on the wavelet components of theta and alpha, and then the average duration of the spindles was estimated. In conclusion, the wavelet transform may be useful in developing novel quantitative time-frequency measures of sleep EEGs as valuable complements of conventional FFT method to analyze the transient changes in sleep EEGs induced by physiological, pathological or pharmacological conditions.

Abstract:Variations of instantaneous heart rate appears regularly oscillatory in deeper levels of anesthesia and less regular in lighter levels of anesthesia. It is impossible to observe this "rhythmic-to-non-rhythmic" phenomenon from raw electrocardiography waveform in current standard anesthesia monitors. To explore the possible clinical value, I proposed the adaptive harmonic model, which fits the descriptive property in physiology, and provides adequate mathematical conditions for the quantification. Based on the adaptive harmonic model, multitaper Synchrosqueezing transform was used to provide time-varying power spectrum, which facilitates to compute the quantitative index: "Non-rhythmic-to-Rhythmic Ratio" index (NRR index). I then used a clinical database to analyze the behavior of NRR index and compare it with other standard indices of anesthetic depth. The positive statistical results suggest that NRR index provides addition clinical information regarding motor reaction, which aligns with current standard tools. Furthermore, the ability to indicates the noxious stimulation is an additional finding. Lastly, I have proposed an real-time interpolation scheme to contribute my study further as a clinical application.

Abstract:Abstract The anesthetic states induced by general anesthesia are accompanied by changes in functional and effective connectivity in brain networks. Considering the perturbations in oscillations produced by anesthesia, we postulated that the capability of information transmission in brain networks has distinct manifestations in different frequency bands. Therefore, we implanted electrodes in brain regions of the rat default mode network (DMN) that are related to anesthetic states to acquire local field potentials. Data were computed with power spectral density, graphic theory analysis, fuzzy entropy, and directed phase transfer entropy. Our results illustrated that the transition patterns of DMN activity are frequency dependent. Collectively, the sedative and general anesthetic states derive from inharmonious information processing ໿instantiated by contrasting lower/higher-frequency changes in DMN activity. These results impart insights into the neural network mechanisms underlying anesthesia and suggest the potential of monitoring depth of anesthesia based on lower/higher frequency synergy at the network level.

Abstract:This paper investigates the problem of automatic depth of sedation (DOS) estimation from electroencephalogram (EEG) recordings. Bispectral index (BIS) is used as a criterion to estimate the new modeling and calculation methods in this paper. Estimation of DOS contains two steps. First, time-frequency domain signal processing technique and nonlinear dynamical techniques based on the concept of chaos have been used to extract multiple features; Second, these features have been employed and modeled by Lasso technique, DOS is calculated and the evaluation models for specific EEG samples are obtained as a result. In emulation and clinical application, DOS are calculated and compared with BIS. Result shows that estimation including the nonlinear features are much closer to BIS than that without nonlinear features, it shows that the nonlinear chaos features have a great advantage in reflecting the chaotic characteristics of the brain during the anesthesia process.

Abstract:We accurately reconstruct the Local Field Potential time series obtained from anesthetized and awake rats, both before and during CO euthanasia. We apply the Eigensystem Realization Algorithm to identify an underlying linear dynamical system capable of generating the observed data. Time series exhibiting more intricate dynamics typically lead to systems of higher dimensions, offering a means to assess the complexity of the brain throughout various phases of the experiment. Our results indicate that anesthetized brains possess complexity levels similar to awake brains before CO administration. This resemblance undergoes significant changes following euthanization, as signals from the awake brain display a more resilient complexity profile, implying a state of heightened neuronal activity or a last fight response during the euthanasia process. In contrast, anesthetized brains seem to enter a more subdued state early on. Our data-driven techniques can likely be applied to a broader range of electrophysiological recording modalities.

Abstract:In conscious states, the electrodynamics of the cortex are reported to work near a critical point or phase transition of chaotic dynamics, known as the edge-of-chaos, representing a boundary between stability and chaos. Transitions away from this boundary disrupt cortical information processing and induce a loss of consciousness. The entropy of the electroencephalogram (EEG) is known to decrease as the level of anesthesia deepens. However, whether the chaotic dynamics of electroencephalographic activity shift from this boundary to the side of stability or the side of chaotic enhancement during anesthesia-induced loss of consciousness remains poorly understood. We investigated the chaotic properties of EEGs at two different depths of clinical anesthesia using the maximum Lyapunov exponent, which is mathematically regarded as a formal measure of chaotic nature, using the Rosenstein algorithm. In 14 adult patients, 12 s of electroencephalographic signals were selected during two depths of clinical anesthesia (sevoflurane concentration 2% as relatively deep anesthesia, sevoflurane concentration 0.6% as relatively shallow anesthesia). Lyapunov exponents, correlation dimensions and approximate entropy were calculated from these electroencephalographic signals. As a result, maximum Lyapunov exponent was generally positive during sevoflurane anesthesia, and both maximum Lyapunov exponents and correlation dimensions were significantly greater during deep anesthesia than during shallow anesthesia despite reductions in approximate entropy. The chaotic nature of the EEG might be increased at clinically deeper inhalational anesthesia, despite the decrease in randomness as reflected in the decreased entropy, suggesting a shift to the side of chaotic enhancement under anesthesia.
