Study of Inhibitory Effect and Mechanism of Nitric Oxide on the Proliferation of Vascular Smooth Muscle Cells

Abstract:Objective: To evaluate the effect of a nitric oxide donor, diethylenetriamineNONOate (DETANO) on the proliferation of vascular smooth muscle cells and on a cell cycle regulator P27kipl. Methods: The cultured vascular smooth cells were treated with DETANO, DETA and negative control(DMSO). Cell number was determined every two days with hematocytometer. The level of P27 expression in synchronized cells, proliferative cells and DETANO treated cells was assessed by Western blot. Results: DETANO inhibited the proliferation of vascular smooth muscle cells in response to 10% fetal calf serum(FCS), and it was most significant at 10-3 mol ·L -1. However, DETA had no effect on cell proliferation. There was no toxic effect of DETA or DETANO on smooth muscle cells as assessed with trypan blue. P27 was detected in G0 phase, and decreased substantially after 24-hours' stimulation by 10% FCS. In contrast, this decrease was blunted in DETANO-treated cells. Conclusion: NO inhibits vascular smooth muscle cells proliferation in a dose-dependent manner. The mechanism underlying this effect is partly through cell cycle regulation and prevention of P27kipl down-regulation.
