Role of L-arginine in Renal Injury of Diabetic Rats

Abstract:Objective: To investigate the role of L-arginine in renal injury of rats with diabetic mellitus (DM). Methods: 24 rats were divided into three groups: 1 normal controls (group A), 8 rats; 2 diabetic rats (group B), 8 rats; 3 diabetic rats +0.5% L-arginine at 4th weeks after initiating DM (group C), 8 rats, and they were observed for eight weeks. Results: From the 1st week, Ccr and UAE increased significantly in group B and C compared to those in group A (P< 0.01), but showed no difference between in group B and C. At the 1st and 2nd week, urinary NO 2 -/NO 3 - were increased in group B and C compared to that in group A (P<0.01), but decreased significantly in group B compared to group A and increased in group C by the 8th week. At the 8th week, PAS positive stained materials were increased significantly in glomerular mesangial matrix in group B. Diabetic animals showed significant increase in glomerular mesangial matrix by light microscopy and immunostaining for laminin in mesangium and tubular basement membrane. Administration of L-arginine to diabetic animals at two weeks relieved the morphologic alteration and laminin deposition. Conclusion: Nitric oxide initially increased and then gradually decreased in the development of diabetic nephropathy. Administration of L-arginine to diabetic animals increased the synthesis of NO and worsened renal injury in early DM. But administration of L-arginine in the development of diabetic nephropathy can extenuate mesangial matrix expansion and may delay the process of glomeruloscherosis.
