Abstract:Objective:To investigate the efficacy of autologous transplantation of bone-marrow mononuclear cells for the treatment of patients with severe lower limb ischemia.Method:5 patients (5 legs) with severe lower limb ischemia were treated by autologous transplantation of bone-marrow mononuclear cells from March to August, 2003. They were 4 males and 1 female. The mean age was 71.5 (range 51—81).Among them, 4 were diagnosed as diabetic foot with gangrene or ulcer,1 thromboangitis obliterans. After aspiration of 350 ml bone marrow blood from the ilium, the mononuclear cells were isolated and then injected into the skeletal muscles of the ischemic leg.Result:No complications were found in this group. All the ischemic legs were preserved. The limb-preserved rate were 100%. The skin turned warm in all legs(100%). TcPO2 of the ischemic legs was obviously elevated in all patients. Angiography showed a noticeable increase of visible collateral vessels in 2 limbs of 2 patients who had angiographic follow-up.Conclusion:Autologous transplantation of bone-marrow mononuclear cells could be a simple, safe, and effective method to treat patients with severe lower limb ischemia.
