Regulation function of transforming growth factor-beta on the proliferation and differentiation of rat neural stem cells

Abstract:Aim: To observe the regulation mechanism of transforming growth factor-β acting on the proliferation and differentiation of neural stem cells in the anterior subventricular zone (SVZa) of neonatal rat brain (P0 day). Methods: mRNA expression of transforming growth factor-beta(TGF-β) in the neonatal rat brain was shown by in situ hybridization which labeled the cRNA probes with the digoxin. Results: The message TGF-β RNA was expressed extensively in the brain of neonatal rat brain (P0), and the pathway from anterior subventricular zone through rostral migratory stream(RMS) to olfactory bulb was the most obviously expression region. The strongest intensity was in the SVZa region, that is, the number of positive cells were the most and the density of the positive cells were the highest in the SVZa, while the weaker in RMS and olfactory bulb was increased. Positive immunoreactive products could be seen both in the cytoplasm and the nucleus of cells, and mainly deposited in the nucleus. Conclusion: TGF-β is highly expressed in the neural stem cells of rat SVZa. It suggests that TGF-β takes part in maintaining the proliferation status of neural stem cells, which results in a properly stable proliferation of the neural stem cells. Moreover, the high expression of TGF-β in the olfactory bulb infers that the it may mainly affect the differentiation of olfactory bulb cells.
