Effects of basic fibroblast growth factor and epidermal growth factor on proliferation of bone marrow neural stem cells in rhesus monkey in vitro induction

Abstract:Aim: To investigate the proliferating conditions of bone marrow stromal cells (BMSCs) differentiated into neural stem cells (NSCs) in vitro, and compare the effects of basic fibroblast growth factor(bFGF) and epidermal growth factor(EGF) on the growth curve of neural stem cells, which provides evidences for the studies in neural scientific field. Methods: As experimental targets, BMSCs were acquired from the bone marrow of rhesus monkey. In experimental group, NSCs culture medium, bFGF and EGY were used to culture, induce and proliferate in vitro. NSCs culture medium was used to culture and proliferate in control group. Nestin and CD133 antigen were served as immunocytochemistry to identify the sign of NSCs. Results: Most cells of primary culture began to express Nestin and CD133 positive antigen on the sixth day, and the number of cells was increased in these two groups, however, the result in experimental group was obviously. The number of cells in bFGF & EGF group was increased from (96.30 ± 8.78) × 10 4 on the eighth day to(1407.90 ± 26.62) × 10 4 on the 18th day, and the beginning time of proliferation was ahead of two days compared with that in control group. A few of NSCs were differentiated into neuron and neuroglia after ten days. Conclusion: The application of bFGF combined with EGF can accelerate the proliferated time of NSCs and improve the effects of proliferation. Moreover, bFGF and EGF can induce the NSCs to differentiate into nerve cells.
