Expression of Nestin in different stage of neural stem cells derived from rabbit bone marrow

Abstract:Aim: To investigate the expression of Nestin in different stage of rabbit bone marrow stroma cells (BMSCs) differentiation into nerve cells, for ascertaining the optimal transplantation time of BMSCs in repairing nerve injury. Methods: BMSCs were acquired from rabbit bone marrow and cured in nerve stem cell medium of CYTOKINE, and then divided into control group glial cell line-derived neurotrophic factor (GDNF, 10 ng/mL) plus retinoic acid (RA, 0.5 pg/mL) group and basic fibroblast factor (bFGF, 10 ng/mL) group. Fluorescent immunocytochemistry was employed to identify Nestin antibody of nerve stem cells. At each culture stage, the proportion of cells that express Nestin was measured by flow cytometry (FCM). Results: Under the same culture condition, some of the rabbit BMSCs were proliferated and differentiated into large round cells with rough and large cytoplasmic granules, which were found to express Nestin could be further differentiated into neuron-like cells. FCM examination showed that about 5.52% of purified BMSCs expressed Nestin; In the control group, the proportions of Nestin-positive cells were 1.56% and 0.47% respectively on the 8th and the 16th day after inducement. In the GDNF + RA group, the proportion was decreased from 1.84% on the 8th day to 0.31% at the 16th day; However, in the bFGF group, the proportion was increased to 6.18% on the 8th day, and then declined to 1.33% at the 16th day. Conclusion: Rabbit BMSCs could be differentiated into nerve cells under the above experimental condition. During the differentiation of BMSCs into neuron-like cells, the expression of Nestin has a decreased tendency, while bFGF can enhance the proportion of Nestin-expressed cells which can be used to enrich nerve stem cells in vitro.
