Study on cardiovascular disease mechanism:the regulative effect of extracellular signal-regulated kinase in bradykinin-induced prolfferation of vascular smooth muscle cells

Abstract:Aim: To study the role and possible regulating function of mitogen-activated protein kinase (MAPK) signal path in promoting the proliferation of the bradykinin (BK)-induced rat vascular smooth muscle cell (VSMC) from the angle of extracellular signal-regulated mitogen-activated protein kinase (ERKs). Methods: Two clean SD rate were selected, and VSMC was cultured with the separated carotid artery smooth muscle. In this experiment, the DNA metabolism and protein synthesis ratio was expressed by 3H-thymidine (3H-TdR) and VSMC 3H-leucine participation rates respectively, and was pretreated with BK inhibitor N-acetyl-L-cysteine (NAC) and ERKS inhibitor UO126. Their effect on the cell protein synthesis and cellular proliferation was observed. Results: The participation rate of VSMC 3H-TdR and 3H-leucine treated with BK (10-8 mmol/L for 30 minutes was significantly increased (increased 412.1% and 137.9%, P < 0.01). Partial function of heightened 3H-TdR could be inhibited by NAC and UO126 (the increased inhibitive rate was 27.70% and 28.60%, P < 0.05) and was obviously inhibited by NAC + UO126 (the increased inhibitive rate was 65.50%, P < 0.01). Participation function of 3H-leucine could he inhibited by NAC and UO126 (the increased inhibitive rate was 28.70% and 20.80%, P < 0.05), and was completely inhibited by NAC + UO126 (the increased inhibitive rate was 116.70%, P < 0.01). Conclusion: The activation of ERKs plays an important role in VSMC growth and proliferation induced by BK. And the proliferation effect of vascular smooth muscle cells can be influenced by the inhibition of ERKs signal path special characteristic inhibitor. So that, it can effect the occurrence of cardiovascular disease.
