An experimental study on acellular dermal matrix prepared by a modified method

Abstract:Aim: To investigate the acellular xenodermis prepared by digestion with low concentration of trypsin and repeated freeze-thaw cycles, and to observe the results after transplantation under the dorsal skin of rats. Methods: The partial-thickness porcine skins were immersed into 0.5 g/L trypsin solution for digestion overnight at 4°C so as to remove the epidermis and most cells in the dermis. Then the dermis was treated at 4°C for cooling, - 70°C for freezing, 37°C for thawing respectively for three times to remove the remaining cells. The prepared acellular dermis was examined macroscopically with gross observation, histological assay, bacteria culture test and cytotoxicity test. After implantation on the fascia beneath the skin flap of the rat, the acelluar dermal matrix were examined by gross and histological observation at 2, 4, 6 and 8 weeks postoperation. Results: The surface of the prepared acellular pig dermis was smooth, pliable, elastic and ductile. Observed under light microscopy, the acellular dermal matrix was covered with very thin intact basement membrane, and well-organized collagen existed. There were not any cellular components, cytotoxicity and bacteria. The satisfactory healing without obvious inflammation of the acellular dermal matrix after transplantation under rat skin was observed. Conclusion: Trypsinization with low concentration and repeated freeze-thaw cycles is a simple and effective method for the preparation of acellular xenodermis with intact basement membrane and uninjured collagen.
