Protective effect of pyruvate on ischemia-reperfusion injury in rat small bowel

Abstract:Aim: To investigate the protective effect of pyruvate on ischemia-reperfusion injury in rat small bowel and its mechanism. Methods: Rat model with ischemia-reperfusion injury in small bowel was set up. Instilled liquid diet containing pyruvate or isoenergetic polyglucose into the lumen of the trial group or control group respectively. Histological studies of intestine were carried out and tissue levels of tumor necrosis factor α (TNF-α) and interleukin 6 (IL-6) were measured using enzyme-linked immunosorbent assay at different time. Results: The mean intestinal tissue injury grades administered with pyruvate were (2.17 ± 1.17), (2.17 ± 0.98), (2.33 ± 1.03) after 45 min ischemia and 30 min, 60 min reperfusion, which were significantly slighter than the control group(t = 55.00, 57.00, 57.00 respectively, P < 0.01). The mean levels of TNF-α in administered with pyruvate were (0.36 ± 0.06), (0.87 ± 0.06), (0.70 ± 0.17) μg/L after 45 min ischemia and 30 min, 60 min reperfusion, which were significantly lower than the control group (F = 313.58, 815.77, 105.84 respectively, P < 0.01). The mean levels of IL-6 were (122.88 ± 3.75), (213.37 ± 8.91), (154.53 ± 7.32) ng/L after 45 min ischemia and 30 min, 60 min reperfusion, which were significantly lower than the control group (F = 1587.18, 1232.96, 2447.93 respectively, P < 0.01). Conclusion: Pyruvate has protective effect on ischemia-reperfusion injury in rat small bowel and the protective effect may be associated with decreasing the levels of TNF-α and IL-6 during the intestinal tissue injury.
