Protective effect of inhibition of p38 MAPK on the damage to rat hippocampal neurons induced by epileptic seizure

Abstract:Aim: To examine the effect and mechanism of inhibition of p38 MAPK in reducing the impairment to the hippocampal neurons, which results from temporal lobe epileptic seizure induced by kainic acid(KA). Methods: Microinjection of SB203580(0, 0.01, 0.1, 1 μg, respectively), a p38 MAPK inhibitor, was administrated into the lateral ventricle of rats as pretreatment; 30 minutes later, KA was injected by the same approach; 7 days later, pyramidal cells and granular cells within the hippocampus were observed and measured by means of Nissl staining method. Results: Seven days later, the pyramidal cells showed gradual degeneration and a great loss, accompanied by granular cells proliferation, which distributed in a state of irradiation, in the hippocampal CA3 region due to status epilepticus induced by KA. And there was a similar change in the hippocampal CA1, but to a certain extent cell loss was found. However, the above-mentioned changes were obviously diminished by pretreatment with SB203580(≥0.1 μg). In the sham-operation group, no significant neuronal damage was detected. Conclusion: Inhibition of p38 MAPK may play a role in protecting the neurons within the hippocampus of rats against the impairmets resulting from whole-body spastic status epilepticus induced by KA.
