Abstract:OBJECTIVE To investigate the effects of hydrophilic/hydrophobic additives on protein release from PLGA micro-spheres. METHODS The conventional PLGA microspheres and PLGA microspheres with one of additives, HP-β-CD, PEG 6000 or stearic acid, were all prepared by the double emulsion solvent evaporation method. Their protein contents of bovine serum albumin (BSA) were determined by micro-BCA method. RESULTS No significant effects of the additives on the particle sizes were observed. Additives affected the protein encapsulations apparently. Hydrophilic HP-β-CD obviously accelerated the release of BSA from the PLGA microspheres, Hydrophobic stearic acid significantly decreased the release of protein, and amphiphilic PEG 6000 delayed the protein release at some degree. Their SEM showed the corresponding changes. CONCLUSION The protein released from PLGA microspheres can be modified by adding into different hydrophilic/hydrophobic additives.
[Preparation of Alginate-Chitosan-poly (Lactic-Co-glycolic Acid) Composite Microsphere and Its Regulation of Protein Release].

Abstract:AIMTo elevate the encapsulation efficiency, decrease the burst release and improve the release of protein entrapped in poly (lactic-co-glycolic acid) (PLGA) microspheres. The bovine serum albumin (BSA) composite microspheres of alginate-chitosan-PLGA were prepared and the release characteristics of BSA from this composite microspheres were studied.METHODSThe much smaller calcium alginate microcapsules were first prepared by a modified emulsification method in an isopropyl alcohol-washed way and coated with chitosan, then the alginate-chitosan microcapsules were further entrapped in PLGA to form the composite microspheres. The protein concentration was determined using a BCA protein assay kit. The release profiles were changed with various formulation factors.RESULTSThe average diameter of the composite microcapsules was about 30 microm. Comparing with 60% to 70% of the conventional PLGA microspheres, the average encapsulation efficiency was more than 80%, and the burst releases in phosphate buffer solution of the composite microspheres decreased from 40% and 50% to 25% and further to 5% in saline solution.CONCLUSIONThe novel composite microspheres were prepared, the drug encapsulation efficiency increased and the burst release decreased. The desired release profiles could be obtained by regulating the ratios of PLG and PLA in the composite microspheres.

Abstract:To decrease the initial burst release of protein entrapped in PLGA microspheres, bovine serum albumin was entrapped into microspheres and a one-step modified method was evaluated. Low level of alginate was added into the internal aqueous phase with protein together; meanwhile calcium chloride and chitosan were put into the external, and all the other processes kept unvaried. After modification, the initial release was inhibited markedly, and the entrapment efficiency was increased slightly, while the particle size remained unaltered. An attracting result is that almost all the little pores at the surface of microspheres had been closed with the modification one under SEM.

Abstract:Microspheres as novel drug delivery carriers have been widely used in clinical study, and poly lactic-co-glycolic acid (PLGA) has been one of the most attractive polymeric candidates used to fabricate devices for drug delivery due to the biocompatible and biodegradable characteristics. In recent years, in order to figure out the defects during the drug release, polymeric combination and chemical modification has been extensively studied. And the design of double-walled microspheres also has been investigated. Moreover, encapsulation efficiency and the release profiles can be improved by different modification methods. And it is extensively used in drug deliver, gene therapy and imaging diagnosis, especially in the fields of tissue engineering.

Abstract:Sustained release drug delivery from microparticles is an excellent alternative for daily protein/peptide drug administration protocol. Poly(lactic acid) (PLA) and poly(lactic-co-glycolic acid) (PLGA) are the most commonly used polymer carriers in the development of protein/peptide microspheres. Basically there are three preparation methods for PLA/PLGA microspheres: the solvent extraction/evaporation based multiple emulsion (W/O/W emulsion) method, the phase separation method and the spray drying method. The stability of the protein/pipetide loaded, encapsulation efficiency, and the burst effect of the microspheres are key problems usually met in the preparation of microspheres. In this review the preparation techniques and progress in the development of protein/pipetide microspheres which aimed to stabilize protein/peptide structural integrity, keep the bioactivity of drugs, increase the encapsulation efficiency and improve the release profile were summarized and evaluated.

Abstract:A novel method was proposed to incorporate and modulate protein release from poly(lactide-co-glycolide) (PLGA) microspheres by a modified w/o/w emulsion solvent evaporation technique with poly(methacrylic acid) (PMAA)/insulin complex suspension as the inner aqueous phase instead of the neat protein solution. It was found that a reversible, water-insoluble complex could be formed between PMAA and insulin by electrostatic interactions. A great increase in insulin entrapment efficiency was observed as the PMAA/insulin complex was adopted to prepare PLGA microspheres. A large number of the complex particles adsorbed at the surface of the microspheres, resulting in the more rapid insulin release. The complexation and microencapsulation processes have little effect on insulin bioactivity, which was revealed by examination of the plasma glucose levels of the diabetic rats administrated with the microspheres.

Abstract:A modified w/o/w emulsion solvent evaporation technique was adopted to prepare human Chorionic Gonadotropin (hCG)-loaded polylactide (PLA) or poly(lactide-co-glycolide) (PLGA) microspheres. The effects of preparative parameters, such as stirring rate, polymer MW and concentration, and the composition of both the inner aqueous phase and oil phase etc., on hCG entrapment efficiency and microsphere characteristics were investigated. It was found that by adding 20% glycerol into the inner aqueous phase and 40% acetone into the oil phase, smooth microspheres approximately 1 microm in diameter could be produced with high hCG entrapment efficiency (>90%). In vitro release test showed a burst release of hCG from PLGA (75:25) microspheres, followed by sustained release of 55% hCG over 2 months. The initial hCG burst from PLGA microspheres increased with the glycerol concentration in the inner aqueous phase, but decreased to a low value (ca. 20%) with the addition of acetone into the oil phase, which could be attributed to the associated changes in surface morphology of the microspheres. In vivo experiments demonstrated that a single shot of hCG-loaded PLGA microspheres could produce a comparable antibody response with the inoculation of free hCG four times.

Abstract:AIM To establish a HPLC method with evaporative light scattering detector(ELSD)to determine the encapsulating efficiency and loading capacity of ginkgolide B(GB)in poly(ethyleneglycol)-poly(L-lactic-co-glycolic acid)(PEG-PLGA)nanoparticles.METHODS Chromatographic conditions were as follows:Shiseido Cg column(250 mm×4.6 mm,5μm);V(methanol):V(tetrahydrofuran):V(water)= 31:11:58 as mobile phase.ELSD parameters were set as follows:evaporation temperature was 105℃,carrier gas(N_2)flow rate was 1.2 L·min~(-1)and column temperature was 45℃.Low temperature ultracentrifugation and gel filtration methods were used to purify nanoparticles.RESULTS In the range of 50-500 mg·L~(-1),all plots were linear and the correlation coefficients were high than 0.999 0;the relative standard deviation(RSD)of the reproducibility was 4.28%;average recovery was in the range of 95%-105%.As the dosage of GB was 1 mg,the encapsulating efficiency(EE)and the loading capacity(LC)was75.33%and3.77%, respectively.CONCLUSION The method is simple,rapid,accurate and can be used for quality control of GB loaded nanoparticles.

Abstract:To encapsulate extractants by method of microencapsulation can solve the problems of phase dispersion, phase mixing, phase separation, the leakage of extractant, as well as complicated structure of equipment in the extraction process. In the present work polysulfone and polystyrene as polymer materials were used to encapsulate the extractants such as TBP, D2EHPA, TOA, Aliquat 336 etc by means of solvent evaporation. The influence of shell materials, dispersants, stirring speed and composition of the organic phase has been investigated. The experimental results showed that TBP and D2EHPA were successfully encapsulated with. polysulfone as the shell material while for the preparation of TOA and Aliquat 336 the shell material of polystyrene should be applied. The spherical shape and monodispersity of the microcapsules were related to the dispersion agent used in the O/W system. The mean diameter of the microcapsules decreased greatly with an increase of the stirring speed. Besides the stirring speed, the viscosity of polymer solution and the interfacial tension of working systems are the major factors that influence the size of the microcapsules. An optimal encapsulation efficiency of the microcapsules was reached when the content ratio of the shell material to the extractant in the polymer solution was optimized.
Preparation of protein-loaded sustained-release microspheres via 'solid-in-oil-in-hydrophilic oil-in-ethanol (S/O/hO/E)' emulsification

Abstract:While formulating proteins into solid particles prior to microsphere preparation is regarded as an effective way to stabilize such macromolecules, the protein particles may still contact the aqueous continuous phase and be re-dissolved. Dissolved proteins may not only leak into the aqueous continuous phase (resulting in reduced loading efficiency), but also contact water-oil (the hydrophobic polymer solution) interfaces, factors known to denature proteins. To avoid dissolution of solidified protein particles, we developed a microencapsulation procedure involving a hydrophilic "oil" (hO) continuous phase to which the hydrophobic solution of the controlled-release polymer was dispersed. The hydrophilic "oil" phase was a glycerol-based liquid mixed with ethylene glycol and polyvinyl alcohol solution to adjust viscosity and surface tension. This non-water hydrophilic continuous phase is immiscible with the hydrophobic polymer solution yet unable to dissolve pre-formulated protein particles. After the embryonic microspheres loaded with the protein particles were formed in this hydrophilic "oil" phase, the formulation was transferred into a cold ethanol bath where the microspheres were immediately hardened due to extracting the organic solution by ethanol. This method was examined by microencapsulating bovine serum albumin (BSA) and beta-galactosidase (beta-gal) into polylactide-co-glycolide (PLGA) microspheres for encapsulation efficiency, release kinetics and bioactivity preservation. As measured using size exclusion chromatography (SEC-HPLC), up to 90% added BSA was encapsulated in microspheres, and the release kinetics of the protein was adjusted by selecting surfactants used in microencapsulation emulsification. The assay of enzymatic activity of beta-galactosidase in hydrolysis of o-nitrophenyl-beta-d-galactopyranoside (ONPG) indicated that over 90% of the protein recovered from the microspheres was active.

Abstract:This paper presents a solvent evaporation method with silicone oil(PDMS) as the continuous phase for preparation of microcapsules.No surfactant was used in the microcapsule preparation process.Polyacrylonitrile(PAN),cellulose acetate(CA) and chitosan(CTS) microcapsules containing Aliquat 336(ALQ),tetraethylene glycol(TEG) and bovine serum albumin(BSA) have been prepared respectively.The prepared microcapsules are orbicular with very compact surfaces,and the mean diameters are around 10～100 μm.With increasing the viscosity of PDMS or decreasing the concentration of polymer solution,the mean diameters of microcapsules decrease.The loading ratio of microcapsules can be easily controlled by adding separation agents into PDMS.The loading ratio of ALQ/PAN,TEG/CA and BSA/CTS microcapsules can reach up to 0.43,0.38 and 0.08 g/g respectively.
Exploring the Effects of Process Parameters during W/O/W Emulsion Preparation and Supercritical Fluid Extraction on the Protein Encapsulation and Release Properties of PLGA Microspheres

Abstract:In this study, protein-loaded poly(lactic-co-glycolic acid) (PLGA) microspheres were prepared via supercritical fluid extraction of emulsion (SFEE) technology. To understand the correlation between process parameters and the main quality characteristics of PLGA microspheres, a comprehensive prior study on the influence of process variables on encapsulation efficiency (EE), initial drug burst release (IBR), morphology, surface property, and particle size distribution (PSD) was conducted within a wide process condition range of each unit process step, from the double-emulsion preparation step to the extraction step. Bovine serum albumin (BSA), a high-molecular weight-protein that is difficult to control the IBR and EE of PLGA microspheres with, was used as a model material. As double-emulsion manufacturing process parameters, the primary (W/O) and secondary emulsion (W/O/W) homogenization speed and secondary emulsification time were evaluated. In addition, the effect of the SFEE process parameters, including the pressure (70–160 bar), temperature (35–65 °C), stirring rate (50–1000 rpm), and flow rate of supercritical carbon dioxide, SC-CO2 (1–40 mL/min), on PLGA microsphere quality properties were also evaluated. An increase in the homogenization speed of the primary emulsion resulted in an increase in EE and a decrease in IBR. In contrast, increasing the secondary emulsification speed resulted in a decrease in EE and an increase in IBR along with a decrease in microsphere size. The insufficient secondary emulsification time resulted in excessive increases in particle size, and excessive durations resulted in decreased EE and increased IBR. Increasing the temperature and pressure of SFEE resulted in an overall increase in particle size, a decrease in EE, and an increase in IBR. It was observed that, at low stirring rates or SC-CO2 flow rates, there was an increase in particle size and SPAN value, while the EE decreased. Overall, when the EE of the prepared microspheres is low, a higher proportion of drugs is distributed on the external surface of the microspheres, resulting in a larger IBR. In conclusion, this study contributes to the scientific understanding of the influence of SFEE process variables on PLGA microspheres.

Preparation of uniform microcapsules with silicone oil as continuous phase in a micro-dispersion process.

Abstract:This paper presents an improved solvent evaporation method with silicone oil ( PDMS) as the continuous phase for preparation of microcapsules to make more polymer solvents available. A microchannel device was used to produce emulsions instead of mechanical stirring to prepare the mono-dispersed microcapsules. Under the conditions of lower evaporation temperature and shorter evaporation time, uniform polyacrylonitrile (PAN) microcapsules containing Aliquat 336 (ALQ) have been successfully prepared. N,N-dimethylformamide (DMF) with lower evaporation ability was applied as the polymer solvent. The prepared microcapsules have rough surfaces and homogeneously internal structures. By changing the two-phase flow rate, the mean size of microcapsules can be easily controlled. When more ALQ was added in polymer solution, the loading ratio of microcapsules increased. The mass transfer performance and stability were determined by extraction of Cr (VI) ions from its aqueous solution. The mass transfer rate was fast enough. After three times of repeated extraction and stripping, the microcapsules kept almost the same extraction ability, which indicated that the microcapsules have very good stability.

Biodegradable polymers with certain compositions and characteristics such as polylactide (PLA), poly(epsilon-caprolactone) (PCL) and their block copolymer were used to prepare the microspheres. The results indicate that, under the same processing conditions, the drug entrapment efficiency was similar (similar to 80%) for microspheres prepared with PLA and P(LA-b-CL) (78.7/21.3 by mole), but it was only 25.4% for PCL microspheres. The in vitro drug release rate decreased in the order of PCL, P(LA-b-CL) (78.7/21.3 by mole) and PLA microspheres. PCL microspheres showed more serious burst release during the first day (almost 80%) than P(LA-b-CL) (50%) and PLA microspheres (18%). The complete ibuprofen release duration from the last two kinds of microspheres exceeded I month. Characterization of the microspheres by differential scanning calorimetry (DSC), scanning electron microscopy (SEM) and polarized optical microscope (POM) revealed that ibuprofen was amorphous in PCL microspheres and partially crystalline in P(LA-h-CL) and PI-A microspheres. The different release behaviour of ibuprofen from the three kinds of microspheres could be attributed to the different crystallinity of the studied polymers and drug dispersion state in polymer matrices. All the above results suggest that the copolymer with a certain ratio of lactide to epsilon-caprolactone could have potential applications for long-term ibuprofen release.
Solvent hydrolysis rate determines critical quality attributes of PLGA microspheres prepared using non-volatile green solvent

Abstract:This study aimed to develop a solvent hydrolysis-based microencapsulation technique that could fabricate PLGA microspheres, while using dimethyl carbonate as a green dispersed solvent. Instead of existing physical solvent removal techniques, a strategy was derived to use a chemical reaction that could transform oil droplets into microspheres. An oil-in-water emulsion was first produced by emulsifying a PLGA/Nile red/progesterone/dimethyl carbonate dispersed phase in an aqueous phase. Adding a NaOH solution into the emulsion led to the decomposition of dimethyl carbonate that partitioned to the water phase. This chemical reaction allowed the continuous diffusion of dimethyl carbonate existing in emulsion droplets into the aqueous phase and its complete removal. The solvent hydrolysis rate was identified as the most important process parameter affecting the major quality attributes of PLGA microspheres. For instance, it was shown through a 3D analysis that Nile red was uniformly dispersed across the microsphere matrix at a fast solvent hydrolysis rate. In comparison, a slow solvent hydrolysis rate allowed the dye to disperse heterogeneously in the microsphere matrix. A drug crystallization phenomenon, being commonly observed in conventional emulsion-templated processes, was inhibited by increasing the hydrolysis rate of dimethyl carbonate. Furthermore, adjusting the solvent hydrolysis rate made it possible to improve drug encapsulation efficiency, to disperse drug homogeneously across microsphere matrix, and to reduce variations in size distribution. The green solvent hydrolysis-based microencapsulation technique could be a promising alternative to conventional microencapsulation methods using toxic halogenated organic solvents.
Optimization of the formulation of PLA microspheres containing insulin

Abstract:OBJECTIVE: To investigate the preparation method and optimization technique of polylactic acid (PLA) microspheres containing polypeptides and proteins. METHOD: Insulin was chosen as a model of protein drugs. The trapping efficiency, drug loading and content of insulin were used to evaluate the formulations of microspheres. Oil phases, emulsifiers, temperature of emulsification and temperature of solvent evaporation were chosen as variables for the optimization of PLA blank microspheres. Ratio of aqueous phase/oil phase, PLA concentration, stirring speed and stirring time were optimized. The concentration of emulsifier, PLA molecular weight and PLA concentration were optimized using uniform design and step regression. PLA molecular weight and PLA concentration were evaluated by orthogonal design. RESULTS: Light liquid paraffin and Span-80 was the optimum constitutents of PLA blank microspheres. The orthogonal design demonstrated that PLA-10k at a concentration of 250 mg/ml would produce desirable trapping efficiency, yield and drug loading for insulin-loaded microspheres. Three batches of the optimum formulation were produced. The result of SEM demonstrated that the microspheres had an intact outer surface with a smooth and non-porous surface before hydrolysis. After 15 days in release medium, small pores were seen while the matrices of the microspheres still remained. The mean diameter of the microspheres was 20 μm. CONCLUSION: An optimum formulation was obtained using several optimization steps. The experimental values of the response variables of the fomulation were closen to the predicted values by mathematical models. The methods of the optimization are useful for PLA microspheres containing insulin.

The different release behaviour of ibuprofen from the three kinds of microspheres could be attributed to the different crystallinity of the studied polymers and drug dispersion state in polymer matrices. All the above results suggest that the copolymer with a certain ratio of lactide to epsilon-caprolactone could have potential applications for long-term ibuprofen release.
Drug-nanoencapsulated PLGA Microspheres Prepared by Emulsion Electrospray with Controlled Release Behavior

Abstract:The development of modern therapeutics has raised the requirement for controlled drug delivery system which is able to efficiently encapsulate bioactive agents and achieve their release at a desired rate satisfying the need of the practical system. In this study, two kind of aqueous model drugs with different molecule weight, Congo red and albumin from bovine serum (BSA) were nano-encapsulated in poly (dl-lactic-co-glycolic acid) (PLGA) microspheres by emulsion electrospray. In the preparation process, the aqueous phase of drugs was added into the PLGA chloroform solution to form the emulsion solution. The emulsion was then electrosprayed to fabricate drug-nanoencapsulated PLGA microspheres. The morphology of the PLGA microspheres was affected by the volume ratio of aqueous drug phase and organic PLGA phase (Vw/Vo) and the molecule weight of model drugs. Confocal laser scanning microcopy showed the nanodroplets of drug phase were scattered in the PLGA microspheres homogenously with different distribution patterns related to Vw/Vo. With the increase of the volume ratio of aqueous drug phase, the number of nanodroplets increased forming continuous phase gradually that could accelerate drug release rate. Moreover, BSA showed a slower release rate from PLGA microspheres comparing to Congo red, which indicated the drug release rate could be affected by not only Vw/Vo but also the molecule weight of model drug. In brief, the PLGA microspheres prepared using emulsion electrospray provided an efficient and simple system to achieve controlled drug release at a desired rate satisfying the need of the practices.

Abstract:Porous microspheres composed of poly (l-lactic-co-glycolic acid) (PLGA), poly (l-lactic acid) (PLA), and poly (l-lactide–co-caprolactone) (PLCL) were fabricated. Based on the hydrophilic properties and porous microstructures, PLGA (100kDa Mw and 1.5% concentration) microspheres were selected and cross-linked with gelatin using two different chemical modification methods: ethanediamine/glutaraldehyde (EN/GA) and N-(3-dimethylaminopropyl)-N′-ethyl-carbodiimide/N-hydroxysuccinimide (EDC/NHS). The effect of chemical modification on the surface topography, porosity, wettability, and biocompatibility was evaluated. Porous microspheres modified with gelatin through ethanediamine/glutaraldehyde cross-linking improved the hydrophilic properties of PLGA and generated an open pore microstructure. A significant increase in the cell metabolic activity was recorded after 5 days in culture, suggesting that ethanediamine/glutaraldehyde may be a suitable cross-linking agent for cell culture applications.
