Abstract:Glow discharge plasma (GDP) was used to modify chitosan films to obtain desirable properties. The chitosan films were treated with GDP of 0-800 W for 1 min, respectively. The molecular structure of chitosan films changed under GDP treatment. Chitosan films showed an enhancement of crystallinity with the increasing power of GDP, and the GDP treatment provided them with higher thermal stability, manifested as the exothermic peaks shifted from 291.5 degrees C (0 W) to 294.1 degrees C (800 W). Scanning electron microscope (SEM) indicated that the surface of chitosan films got rougher with 0-600 W GDP treatment, accompanied by the increase of tensile strength (TS), and the TS did not increase with the 800 W GDP treatment. However, the water vapor permanent (WVP) became higher with GDP treatment. GDP treatment also exhibited influence on chromatic aberration of chitosan films. GDP is proved to be a promising green, pollution-free, rapid technology to modify chitosan films. (C) 2019 Elsevier B.V. All rights reserved.

Abstract:The present study illustrated modulation of protein aggregation by affecting disulfide/sulfhydryl exchange reactions by adding different concentrations of free thiol represented by reduced-glutathione (GSH) for modulating myofibrillar protein (MP) gel properties at 75 °C or 95 °C. Gel strength and rheological results showed the effects of GSH were dependent on the concentrations (5, 10, 20, 40, and 80 g/kg) and heating temperatures. SEM results showed that the addition of GSH improved the gel microstructure at 95 °C. AFM and DLS results indicated that protein aggregation was also inhibited. At 75 °C, the addition of GSH influenced both MP aggregation and gel properties. Low concentrations (5, 10 g/kg) of GSH promoted aggregation, whereas high concentrations (20, 40, and 80 g/kg) of GSH inhibited this. By analyzing the protein structure and cross-linking pattern changes of MP and MP/GSH composites, a pathway involving GSH influencing MP gel properties was determined.
Perturbation of ribosome subunit interaction by glutaraldehyde fixation

Abstract:Ultracentrifugal analysis of ribosomal purity is complicated by the rapid reequilibration of ribosomes with their subunits, and this is further enhanced by the effects of hydrostatic pressure. Fixation of the ribosome system prior to ultracentrifugal analysis supposedly freezes the reequilibration, and thus tends to obviate these difficulties. However, no redistribution of the ribosome-subunit population must be allowed to occur during fixation. Thus, it is necessary that fixation be extremely rapid compared to the ribosome-subunit reequilibration, in order to avoid errors in analysis. It was the purpose of this investigation to make a direct experimental comparison of the rates of these two processes, fixation and ribosome-subunit reequilibration, using the stopped-flow technique with a light-scattering detector, under a variety of buffer environment readjustments. The following findings resulted from this study: (1) Fixation in Tris buffers could not be followed by light scattering because of interaction of glutaraldehyde with Tris, leading to continuous production of high molecular weight contaminants. (2) Polymerization of glutaraldehyde leads to increased ultraviolet absorption, which must not be confused with scattering changes. (3) Undialyzed ribosome solutions prepared by dissolving stock suspensions stored at high levels of magnesium and univalent electrolyte into a known standard buffer produced solutions having free Mg2+ levels lower than those of original buffer, complicating kinetic observations and threatening ribosome stability. (4) Addition of malonic acid as a buffer for Mg2+ largely eliminated this problem. (5) The kinetics of glutaraldehyde were measured quantitatively by studying the perturbation of ribosome association and dissociation kinetics produced during shifts of Mg2+ levels. (6) At fixative levels not producing coagulation, fixation kinetics can be competitive with those of association-dissociation. (7) Mass action effects like those of dilution can be caused by fixation, and can give rise to excess subunits, which may be mistaken for loose couples in original ribosome preparations.

Abstract:Phycocyanin (PC), an algae-extracted colorant, has extensive applications for its water-solubility and fresh blue shade. When PC is added to acidified media, dispersions are prone to aggregate and decolorize into cloudy systems. For palliating this matter, chitosan with high, medium, and low molecular weights (HMC, MMC, and LMC) were adopted in PC dispersions, and their protective effects were compared based on physiochemical stabilities. The optimal mass ratio between chitosan and PC was identified as 1:5 based on preliminary evaluations and was supported by the higher ζ-potential (31.0–32.1 mV), lower turbidity (39.6–43.6 NTU), and polyacrylamide gel electrophoresis results. Through interfacial and antioxidant capacity analyses, LMC was found to display a higher affinity to PC, which was also confirmed by SEM images and the maximum increase in transition temperature of their complex (155.70 °C) in DSC measurements. The mechanism of electrostatic interaction reinforced by hydrophobic effects and hydrogen bonding was elucidated by FT-IR and Raman spectroscopy. Further comprehensive stability evaluations revealed that, without light exposure, LMC kept PC from internal secondary structure to external blueness luster to the maximum extent. While with light exposure, LMC was not so flexible as HMC, to protect chromophores from attack of free radicals.

Study on the Physiology and Degradation of Dye with Immobilized Algae

Abstract:Immobilization of chlorella (pyrenoidosa) with calcium alginate was carried out, Both algal growth and physiological activity increased after immobilization. Algal size and initial density have little influence on cell growth and physiological activity. Algal cell division inside the support was restricted without bubbling air containing 2% CO2. Individual algal cell increased and algal distribution inside the support was not homogenous, indicating that within the support the resistance of mass transfer of CO2 was the limiting factor for the growth and cell division of immobilized algae. After bubbling 2% CO2 algal growth, the cell division considerably increased and individual cell size restored normally. Study of the degradation of dye (direct brown NM) by immobilized algae was better than that of free algae. Bubbling air containing 2% CO2 in the culture solution was more favorable for increasing decolorization rate. Preliminary study of co-immobilization of algae plus bacterium illustrated that the addition of bacterium increased the anabolic activity of algae, thus increased the decolorization capacity if immobilized algae for dye.
Specific Bleaching of Phycobiliproteins from Cyanobacteria and Red Algae at High Temperature in Vivo

Abstract:Exposure of blue-green or red algal cells to temperatures exceeding 60–65°C for several minutes resulted in bleaching of all phycobilin absorption in the visible range, with virtually no alteration in chlorophyll or carotenoid absorption. Difference spectra of non-bleached vs bleached cells appeared identical to absorption spectra of purified phycobilisomes isolated from the same cell culture in high phosphate medium. All phycobilin chromophores were bleached at approximately the same rate during heating. There were no changes in apparent molecular weights or relative amounts of the phycobilisome apoproteins during chromophore bleaching. Phycobilisomes in cell extracts from Anacystis nidulans resisted bleaching when suspended in medium of high phosphate concentration, but were bleached at 60–65°C within a few minutes when placed in diluted medium. The results indicate that phycobilisomes in vivo are stabilized by a mechanism other than high osmotic and ionic strength. This represents a rapid and quantitative method to characterize the phycobiliprotein content of cyanobacteria and red algae in vivo.

Abstract:The effects of glycerol and high temperatures on structure and function of phycobilisomes (PBSs) in vivo were investigated in a chlL deletion mutant of the cyanobacterium Synechocystis sp. PCC 6803. When the mutant was grown under light-activated heterotrophic growth conditions, it contained intact and functional PBSs, but essentially no chlorophyll and photosystems. So the structural and functional changes of the mutant PBSs in vivo can be handily detected by measurement of low temperature (77 K) fluorescence emission spectra. High concentration glycerol induced an obvious disassembly of PBSs and the dissociation of phycocyanins in the rod substructures into their oligomers and monomers. PBSs also disassembled at high temperatures and allophycocyanins were more sensitive to heat stress than phycocyanins. Our results demonstrate that the chlL(-) mutant strain is an advantageous model for studying the mechanisms of assembly and disassembly of protein complexes in vivo.

Abstract:Objective To observe the sterilization efficacy of glutaraldehyde disinfector on medical appliances and catheters. Methods The carrier quantitative germicidal test and simulated field trial were used to carry out the experiment. Results 20 g/L glutaraldehyde in this machine was vaporized and the final concentration of which achieved 47 g/m3.Then under 42℃ and exposed to this glutaraldehyde vapor for 6.5h,Spores of Bacillus subtilis var.niger on the stainless steel carrier and the surface,denta-terminal and joint of hemostatic forceps that placed on different layers in the machine were totally killed.The spores of B.subtilis var.niger in medical-catheters with diameter≥φ4.0 mm and length ≤1 500 mm could also totally be killed while that in medical-catheters with diameter≤φ2.8mm could not be totally killed. Conclusion The glutaraldehyde disinfector can kill all the spores of B.subtilis var.niger on the surface of medical appliances except for those in the medical-catheters with smaller diameters.
Formation of chlorophyll-anionic polysaccharide complex coacervates to improve chlorophyll color stability: Thermodynamic and kinetic stability studies

Abstract:Chlorophyll (Chl) is the predominant pigment in green plants that can act as a food color and possesses various functional activities. However, its instability and rapid degradation on heating compromise the sensory qualities of its products. This study aimed to enhance the heat resistance of Chl by forming complex coacervates with two negatively charged polysaccharides, sodium alginate (SA) and K-carrageenan (KC). Dynamic light scattering and scanning electron microscopy analyses confirmed the formation of coacervates between Chl and the polysaccharides, whereas Fourier-transform infrared spectroscopy revealed that hydrogen bonding and electrostatic attraction were the primary forces behind complex formation. Electron spin resonance and thermodynamic studies further revealed that these complexes bolstered the thermal stability of Chl, with a maximum improvement of 70.38 % in t1/2 and a reduction of 50.72 % in the degradation rate constant. In addition, the antioxidant capacity of Chl was enhanced up to 35 %. Therefore, this study offers a novel approach to Chl preservation and suggests a viable alternative to artificial pigments in food products.
Physicochemical degradation of phycocyanin and means to improve its stability: A short review

Abstract:The cyanobacterium Arthrospira platensis, spirulina, is a source of pigments such as phycobiliprotein and phycocyanin. Phycocyanin is used in the food, cosmetics, and pharmaceutical industries because of its antioxidant, anti-inflammatory, and anticancer properties. The different steps involved in extraction and purification of this protein can alter the final properties. In this review, the stability of phycocyanin (pH, temperature, and light) is discussed, considering the physicochemical parameters of kinetic modeling. The optimal working pH range for phycocyanin is between 5.5 and 6.0 and it remains stable up to 45 °C; however, exposure to relatively high temperatures or acidic pH decreases its half-life and increases the degradation kinetic constant. Phycobiliproteins are sensitive to light; preservatives such as mono- and di-saccharides, citric acid, or sodium chloride appear to be effective stabilizing agents. Encapsulation within nano- or micro-structured materials such as nanofibers, microparticles, or nanoparticles, can also preserve or enhance its stability.
Changes of Phycocyanin in Spirulina Platensis after Ultraviolet Light Irradiation

Abstract:The aggravating eutrophication in waters all over the world in recent years results in over growth of algae,especially Cyanophyta,and leads to the decline of water quality.How to control the cyanobacterial growth has become an important research topic.Phycocyanin is a group of key proteins in light absorption and energy conversion.To control algae overgrowth by ultraviolet light irradiation,using Spirulina platensis as an example of cyanobacteria,the potential mechanism of phycocyanin activity change was investigates by studying phycocyanin quantity,purity and structure in the algae cells using three-dimensional fluorescence.UV irradiation can effectively control the cyanobacterial growth probably through damaging key bio-chemical molecules.After UV irradiation,the quantity increase of phycocyanin was restrained,maybe due to the damage of the cell structure which then retained the synthesizing of proteins and the adsorption of sun-lights by the cell.At the same time,the purity of phycocyanin in the living cell was decreased,and after 7 days culturing,the Ex620/Ex280 value of treated samples with 1 min and 30 s of UV irradiation was only 32% and 77% of that of untreated samples at 12.5 mW/cm2,was 52% and 73% at 30 mW/cm2,respectively.UV irradiation not only decreased the quantity of key proteins but also lowered the quality of formed proteins.The main reason for phycocyanin purity decreasing might contribute to the change of phycocynobilin structure,which was obvious in the 3-D fluorescence spectra.Large ring-structure,seldom existed in untreated samples,was common in the treated samples after 7 days culturing.The Ex620/Ex370 value of treated samples at12.5 mW/cm2·min-1 was only 0.68,much lower than that of the untreated samples.The change of phycocyanin concentration and structure was significantly impacted by the time and intensity of UV irradiation.Longer irradiation time was more important than higher UV intensity,indicating that unlike that of UV disinfection,UV dose is not the determining factor in algae growth control and there exists different pathways in UV algae control and UV bacterial control.

Abstract:This review article highlights the role of glutaraldehyde as a matrix activator/stabilizer in imparting higher operational and thermal stability to β-galactosidase (βG) for biotechnological applications. Glutaraldehyde has been used extensively as a crosslinking agent as well as for functionalization of matrices to immobilize β-galactosidase. Immobilized β-galactosidase systems (IβGS) obtained as a result of glutaraldehyde treatment has been employed to hydrolyze whey and milk lactose in batch reactors, continuous packed-bed and fluidized bed reactors under various operational conditions. Moreover, these IβGS have also been utilized for the production of galactooligosaccharides in food, dairy and fermentation industries. It was observed that glutaraldehyde provided remarkable stability to immobilize βG against various physical/chemical denaturants, thus enhancing thermal/operational stability and rendering it more suitable for repeated utilization in industrial scale operations.

Abstract:In this study, a simple method with improved properties for Glutamate Dehydrogenase immobilization is proposed by using bacterial cellulosic nanofiberas as a novel and inexpensive biomatrix. Acetobacter xylinum ATCC 10245 has been chosen and cultivated for synthesis of this nanofiber with the diameter of 30-70 nm. Thereafter, Glutamate Dehydrogenase was immobilized into the nanofiber pores in two steps: 1) enzyme adsorption and 2) crosslinking by glutaraldehyde. Optimum pH for the activity of immobilized enzyme and optimum temperature was reported in this work. Also the relative activity of immobilized enzyme was about 38 % in comparison to the enzyme activity of the first day after about two months. Moreover after reusing of the enzyme for 9 cycles, the immobilized enzyme retained over 83 % of its initial activity. Glutamate Dehydrogenase is immobilized on bacterial cellulosic nanofiber for the first time which can affect on facilitation of Glutamate production. Also can be a good method for having a stable and robust immobilized Glutamate Dehydrogenase that can withstand harsh operating conditions in processes which are common in these kind of reactions.

Effect of autoclaving on the mechanical properties and structure of the giant squid hydrolysate-polysaccharide gel

Abstract:Fish hydrolysates are an attractive option for preparing composite gel foods. However, not much is known regarding their gel properties. Here, we investigated the effect of the autoclaving treatment (121°C, 10 min) on the microstructure and properties of the giant squid hydrolysate-konjac glucomannan-κ-carrageenan-locust bean gum gel (GSH-P gel). The nuclear magnetic resonance proton spin-spin relaxation time (T2 ) measurements indicated that autoclaving led to stronger water-binding ability of the GSH-P gel. The rheological measurements indicated that autoclaving led to significantly higher viscoelastic modulus and gel strength. The results of confocal laser scanning microscopy and small-angle X-ray scattering indicated that the phase separation of polysaccharides and proteins/peptides was enhanced by autoclaving, and the polysaccharides swelled better, resulting in less, but ordered, network structures. Autoclaving had a similar, but insignificant, positive effect on the polysaccharide (P) gel. Thus, GSH seems to play an important role in the process of polysaccharide gelation. This study shows that autoclaving can alter the structure and improve the properties of the GSH-P gel.
Impact of Spirulina platensis biomass on the viability of Lactobacillus delbrueckii subsp. bulgaricus strain during the freeze-drying process

Abstract:In this work, we evaluated the protective capacity of Spirulina platensis biomass in preserving Lactobacillus delbrueckii subsp. bulgaricus WDCM 00102. The L. bulgaricus strain was freeze-dried in the presence of S. platensis biomass and the freeze-dried samples were then stored at 5 and 25°C for varying periods. Subsequently, the samples were rehydrated and bacterial plate counts were determined. The results indicate that a concentration of 12% S. platensis biomass was highly effective in preserving L. bulgaricus. Commercial products with higher S. platensis biomass content exhibited greater protective capacity. While S. platensis biomass is well-known for its prebiotic properties, its protective role has not been previously reported or thoroughly explored. This study demonstrates the protective capacity of S. platensis biomass in preserving L. bulgaricus, a strain particularly sensitive to preservation processes.

Abstract:The importance of studying the structural stability of proteins is determined by the structure-function relationship. Protein stability is influenced by many factors among which are freeze-thaw and thermal stresses. The effect of trehalose, betaine, sorbitol and 2-hydroxypropyl-β-cyclodextrin (HPCD) on the stability and aggregation of bovine liver glutamate dehydrogenase (GDH) upon heating at 50 °C or freeze-thawing was studied by dynamic light scattering, differential scanning calorimetry, analytical ultracentrifugation and circular dichroism spectroscopy. A freeze-thaw cycle resulted in the complete loss of the secondary and tertiary structure, and aggregation of GDH. All the cosolutes suppressed freeze-thaw- and heat-induced aggregation of GDH and increased the protein thermal stability. The effective concentrations of the cosolutes during freeze-thawing were lower than during heating. Sorbitol exhibited the highest anti-aggregation activity under freeze-thaw stress, whereas the most effective agents stabilizing the tertiary structure of GDH were HPCD and betaine. HPCD and trehalose were the most effective agents suppressing GDH thermal aggregation. All the chemical chaperones stabilized various soluble oligomeric forms of GDH against both types of stress. The data on GDH were compared with the effects of the same cosolutes on glycogen phosphorylase b during thermal and freeze-thaw-induced aggregation. This research can find further application in biotechnology and pharmaceutics.
Ultrasonic Cell Disruption Followed by Isoelectric Point Precipitation for Extraction of Phycobiliprotein from Spirulina platensis

Abstract:A procedure based on ultrasonic cell disruption followed by isoelectric point precipitation was considered for the extraction of phycobiliprotein from Spirulina platensis powder. The respective optimal conditions for ultrasonic cell disruption and isoelectric point precipitation were determined. Results showed that an optimal ultrasonic cell disruption was obtained through 630 W ultrasonic treatment for 20 min of 5% aqueous suspension of Spirulina platensis powder. The optimal medium was glacial acetic acid for the isoelectric point precipitation of phycobiliprotein, which was a simple and convenient operation to obtain phycobiliprotein at pH 4.0 and 4 ℃. Glacial acetic acid is easy to volatilize so that the extraction procedure is simplified. Finally, lyophilization was performed to obtain a crude protein powder. The content of phycocyanin was as high as 4.36% in the supernatant separated from ultrasonic treated 5% aqueous suspension of Spirulina platensis powder under optimized conditions, and the yield of crude protein powder from Spirulina platensis powder was 52.5%, in which, the contents of phycocyanin, allophycocyanin and phycoerythrin were 7.7%, 7.9% and 0.8%, respectively.

Stability of B-Phycoerythrin/Gel Complex

Abstract:The stabilities of B-phycoerythrin(B-PE)/sodium alginate and B-PE/gelatin complexes were investigated by means of the resonance light scattering(RLS),the visible absorption spectroscopy and the differential scanning calorimetry(DSC).The results show that the RLS intensities of the two complexes both increase with the concentrations of sodium alginate and gelatin,respectively,and that B-PE has a stronger interaction with gelatin than with sodium alginate because the light and thermal stability of B-PE/sodium alginate is higher than that of B-PE but lower than that of B-PE/gelatin.

Abstract:Phycobiliproteins (PBPs) are light collecting pigments of cyanobacteria that attract growing interest for several industrial applications. Each step of the extraction process is crucial for yield, concentration and quality of obtained pigments. In the current work, we present an optimization scheme of major limiting steps for PBPs extraction from Arthrospira platensis biomass. As first step, the effects of pretreatment, extraction time, and separation conditions on the recovery of PBPs were compared. Subsequently, the influence of pH and concentration of the extraction buffer as well as the addition of preservatives (Polyethylene glycol (PEG), Magnesium chloride (MgCl 2 ), and Calcium chloride (CaCl 2 )) was studied. In addition, the effect of the biomass type (dried vs wet) and its concentration in the extraction buffer was also investigated. Optimal extraction required the use of dry biomass at relatively low ratio (1:50, solvent:biomass), without previous treatment. The use of concentrated phosphate buffer (100 mM) at a neutral pH gave the highest PBPs recovery and concentration after 6 h of extraction followed with a separation at 6000 rpm during 15 min. Calcium chloride used at 1.5% improved by 30% both PBPs recovery and concentration in the crude extract. The optimized protocol allowed the recovery of 464.5 mg/g PBPs from spirulina biomass with concentration of 15.9 mg/ml. The crude PBPs obtained with this extraction method reduced the stable radical DPPH with a percentage scavenging activity of 86.45 ± 1.2%. This protocol could reduce both PBPs time and cost extraction and is easily scalable for industrial application.
