Abstract:In this study, uracil has been introduced as the nucleating agent (NA) for bacterially synthesized poly[(3-hydroxybutyrate)-co-(3-hydroxyhexanoate)] (PHBHHx) copolymers with HHx content of 5, 10, 18 mol-%, and poly(3-hydroxybutyrate) (PHB) homopolymer for the first time. Its effect was compared with the conventional NA of PHB, that is, boron nitride (BN), and two other naturally occurring pyrimidine derivatives, i.e., thymine and cytosine. The effects of uracil on the crystallization kinetics, melting behavior, spherulite morphology, and crystalline structure of PHBHHx and PHB were investigated by differential scanning calorimetry (DSC), polarized optical microscopy (POM), and wide-angle X-ray diffraction (WAXD). Uracil and BN exhibit the comparable nucleation efficiency on the crystallization of PHB, whereas uracil shows much more effective nucleation ability than BN for PHBHHx copolymers. With incorporation of 1 wt.-% uracil, PHBHHx with 0-10 mol-% HHx units can finish crystallization upon cooling at 10 degrees C . min(-1). The crystallization half-times (t(1/2)) of all the PHB and PHBHHx samples decrease significantly with presence of uracil. The crystallization rate of polymers further enhances with increase in uracil concentration. With addition of 1 wt.-% uracil, the t1/2 value of PHBHHx with 10 mol-% HHx units melt-crystallizing at 80 degrees C decreases to approximate to 4.0% of the neat polymer, and the nucleation density increases by 3-4 orders of magnitude. The incorporation of uracil has no discernable effect on the crystalline structure of PHBHHx, as evidenced by WAXD results. It was proposed that the nucleation mechanism of the uracil/PHBHHx (or PHB) system might be the epitaxial nucleation.

Abstract:A new activity of alkaline protease from Bacillus subtilis for Michael addition reactions of imidazole, 4-nitro-1 H -imidazole and 2-methyl-4-nitro-1 H -imidazole with acrylates and acrylic acid was investigated. The reactions were carried out in pyridine at 50 °C for 72 h. Five N -substituted imidazole derivatives were obtained using acrylate esters, but not acrylic acid, in yields from 62% to 76%.

Abstract:Hydrolase-catalyzed Michael addition of imidazole derivatives to acrylic monomers in organic medium was described. A serial of N-substituted imidazole derivatives were successfully synthesized in moderate yields by the catalysis of hydrolases in organic medium. Nine commercially available hydrolases from different sources were screened and all of them were found to be able to catalyze this type of addition reaction. The reaction yields depended on the solvent properties and the solvents with higher logP value supported this enzyme-catalyzed reaction to give higher conversion. Influence of the structure of the Michael acceptor and donor on the enzymatic Michael addition was also investigated. The acceptor with shorter alcohol chain afforded a higher yield. A more rapid conversion was observed when the donor had an electron-withdrawing group. This hydrolase-catalyzed Michael addition reaction has widened the applicability of biocatalysts in organic and bioorganic synthesis.

Abstract:A general method for accessing 5-alkyl-5-aryl-gamma-lactams has been developed using readily available aryl bromides, nitroalkanes, and methyl acrylate as the starting materials. The palladium-catalyzed arylation of nitroalkanes gave the 1-aryl-substituted nitroalkanes, which underwent the DBU-mediated Michael addition with methyl acrylate at room temperature to afford the methyl 4-aryl-4-nitroalkanoates. The latter were then subjected to the nitro reduction using NaBH4-NiCl2 center dot 6H(2)O in MeOH at 0 degrees C to furnish, after treatment with aqueous K2CO3 at room temperature, the 5-alkyl-5-aryl-gamma-lactams in good to excellent overall yields. Abstract:The transesterification of α-d-glucose with divinylsuccinate, divinyladipate and divinylsebacate in pyridine at 55 °C for 3 days was catalyzed by an alkaline protease from Bacillus subtilis to give corresponding 6- O -vinyl glucose esters at 30%, 53% and 35% yield, respectively. The stereo-selectivity of the alkaline protease toward the α-anomer was affected by the acyl donor chain length. 6- O -Vinylsuccinyl-d-glucose was mixture of α- and β-anomers (α/β=44/56), the other two products were the pure α-d-glucose derivatives.

Abstract:This paper describes highly selective transesterification reactions, catalyzed by an alkaline protease from Bacillus subtilis in pyridine, of several mono- and di-saccharides with divinyl dicarboxylates ranging from 4 to 10 carbon atoms. A series of polymerizable vinyl fatty acid sugar esters were obtained with good selectivity and high yields. Most products had high proportions of the alpha anomer. The influences of the enzymes, solvents, temperature, and acyl donor chain length on the reaction were studied. Vinyl sugar esters offer a new family of functional water-soluble monomers for preparation of sugar-containing polymers. (C) 2004 Elsevier Ltd. All rights reserved.

Abstract:Utilizing the interactions between tryptophan, methyl viologen and cucurbit[8]uril, we found that the distance between the targeted peptides/protein and the reactive peptide was shortened, which facilitated the Michael addition reaction between cysteine and dehydroalanine. The highest acceleration was observed on cysteines with suitable pKa and spatial location to tryptophan, suggesting that our system can be used for regioselective cysteine modification.

Abstract:A highly enantioselective Michael addition of alpha-substituted cyanoacetates to vinyl ketones was accomplished in the presence of simple bifunctional thiourea/tertiary amine organocatalysts. A number of alpha-aryl or alkyl cyanoacetates have been successfully applied to give multifunctional compounds with an all-carbon-substituted quaternary stereocenter in excellent enantioselectivities (82-97 % ee) and yields (61-99 %). The optical pure adducts could be smoothly converted to variously structured beta(2,2)-amino acid esters. Moreover, an interesting reaction model involving multiple hydrogen-bonding interactions amongst the thiourea/tertiary amine catalyst and the reactants has been proposed based on the absolute configuration of the adduct and computational studies.

Abstract:The chemical selectivity between Markovnikov addition and acylation in the reaction of thiols and vinyl esters were developed. The addition reaction was efficiently catalyzed by CAL-B at 50°C in isopropyl ether, affording the Markovnikov adducts with good yields. Acylation was carried out under the catalysis of d-amino acylase in DMF at 50°C. A series of thioethers and thioesters were synthesized via the controllable enzymatic methods.

Abstract:Three acylases, including D-aminoacylase from Escherichia coli, acylase "Amano" from Aspergillus oryzae and immobilized penicillin G acylase from Escherichia coli have been found to possess novel activity to catalyze the Markovnikov addition reaction of N-heterocycles to vinyl esters. The aza-Markovnikov addition reactions of 4-nitroimidazle to vinyl acetate catalyzed by D-aminoacylase, acylase "Amano" and immobilized penicillin G acylase were up to 1260-fold, 720-fold and 320-fold faster than the respective non-enzymatic reaction. Some control experiments have been designed to demonstrate the catalytic specificity of acylases. Under the catalysis of these promiscuous acylases, a number of N-heterocycles, including some pentacyclic N-heterocycles, pyrimidines and purities, were successfully added to a series of vinyl esters in moderate to excellent yields to prepare N-heterocycle derivatives. The acylase-catalyzed Markovnikov addition reaction has provided a new strategy to perform the Markovnikov addition and expanded the application of biocatalysts.
Synthesis of Adenine Arabinoside from Uracil Arabinoside by Immobilized Cells

Abstract:Polyvinyl alcohol,carrageenan and polyacrylamide were used as mixed carriers to immobilize cells of Escherichia sp.AEM0812.The optimal immobilization conditions were as follows: 8% polyvinyl alcohol,1.0% carrageenan and 10% polyacrylamide were used as carriers,then the materials were treated in 4.0% potassium chlorideboric acid(pH 7.0) for 36 h with cell concentration of 30%.Under the above conditions,particles of immobilized cells were rather stable with the relative enzyme activity of 98.17%.The immobilized cells were used as catalyzer to synthesize adenine arabinoside from uracil arabinoside.The conversion rate of uracil arabinoside reached 68% at 54 ℃,pH 6.5 and ion concentration of buffer solution of 120 mmol/L.After 5 batches of repeated reaction,the average conversion rate was about 54.3%.

Abstract:Two disubstituted acetylenes bearing a vinyl group at one end (M1 and M2) were synthesized and polymerized by WCl6-Ph4Sn catalyst. The expected poly(disubstituted acetylenes) PDSAs (P1 and P2) were obtained in high yields. Both P1 and P2 have reactive vinyl groups on their side chains, thus they were used as precursors to be subsequently modified with a mercapto compound through the thiol-ene click reaction to produce the novel PDSAs (P1S and P2S) in good yield. The chemical structures of the polymers were carefully characterized by standard spectroscopic methods such as gel permeation chromatography (GPC), NMR, NMR and FTIR techniques, and satisfactory data were collected. The post-polymerization modification of P1 took a long reaction time (3 days) to convert P1 to P1S because the ene-functionality at the end of side chain of P1 links to a saturated alkyl segment. By using an activated ene-functionality (alpha,beta-unsaturated vinyl), the modification of P2 to P2S took only 1 day under mild conditions. Moreover, the activated end-ene group allowed the post-polymerization modification of P2 by Michael addition, which was confirmed using butylamine as the representative amine compound and the characterization data indicated the validity of the expected P2N. The thermal analysis results indicated that the modified polymers were highly stable thermally with a decomposition temperature over at least 240 degrees C, except for P2N, which showed lower stability due to unstable imine groups. Meanwhile, it was found that the modified polymers P1S and P2S were fluorescent and showed similar emission efficiency to their precursors P1 and P2. These results indicated that the thiol-ene click reaction and Michael addition reaction are accessible routes for post-polymerization modification to generate novel functional PDSAs.

Abstract:A novel strategy to perform Michael additions between 1,3-dicarbonyl compounds and α,β-unsaturated compounds was developed by the catalysis of hydrolase. We found that 11 hydrolase could catalyze the enzymatic Michael addition reaction to form the carbon–carbon bond. In 2-methyl-2-butanol d-aminoacylase showed high Michael addition activity. The influence of substrate and Michael acceptor structure on Michael addition was evaluated systematically. Some control experiments demonstrated that the active site of d-aminoacylase was responsible for the enzymatic Michael addition reaction. This novel Michael addition activity of hydrolase is of practical significance in expanding the application of enzymes and in the evolution of new biocatalysts.
