Abstract:A new stigmasterol 3beta, 7alpha, 22-trihydoxystigmast-5-ene (1) and a new eremophilenolide 8alpha-methoxy-6beta-angeloyloxyeremophil-7(11)-en-8beta, 12-olide-14-oic acid (2) were isolated from Ligularia dolichobotrys Diels. Their structures were deduced on the basis of spectral data.

Abstract:Two new eremophilane sesquiterpene lactones, 1β-angeloyloxy-6β, 10β-dihydroxy-8β-methoxyeremophila-7(11)en-8α,12-olide and 1β-angeloyloxy-6β,10α-dihydroxy-8α-methoxyeremophila-7(11)en-8β,12-olide were isolated from the extract of the whole plant of Ligularia myriocephala Ling. Their structures and stereochemistry were elucidated by various spectroscopic methods including intensive 2D-NMR techniques (COSY, gHMQC, gHMBC and 1H-1H NOESY) and HR-ESIMS.

Abstract:Two new compounds, a stigmasterol (1) and an eremophilenolide (2), were isolated from Ligularia dolichobotrys (Diels) together with ten known sesquiterpenoids, two known triterpenes and five known sterols. Their structures were elucidated by spectroscopic methods (IR, MS, 1H, 13C and 2D NMR). In addition, bakkenolide A (3) exhibited effective antitumor activity to human leukemia cells (HL-60), human hepatoma cells (Bel-7402) and human ovarian neoplasm cells (HO-8910).

Abstract:Phytochemical investigation of the roots of LIGULARIA VEITCHIANA has resulted in the characterization of two new eremophilanolides together with beta-sitosterol, stigmasterol, taraxol, and liguhodgsonal whose structure was further confirmed by the (13)C-NMR techniques. The structures of the new constituents were elucidated by spectroscopic methods and shown to be 1beta, 10beta-epoxy-6beta(2'-hydroxymethylacryloyloxy)-8beta-hydroxy-eremophila-7(11)-en-12,8alpha-olide ( 1) and 1beta, 10beta-epoxy-6beta-(2'-hydroxymethylacryloyloxy)-8beta-ethoxy-eremophila-7(11)-en-12,8alpha-olide ( 2).

Abstract:A phytochemical study of the ethanolic extract of Ligularia przewalskii (Maxim.) Diels led to the isolation of two new terpenoids, (1βH,3βH)-epoxy-olean-13(18)-ene-3α,2-olide (1) and 8β-hydroxy-(10βH)-14β-methyl-6α-angeloyloxy eremophil-7(11)-en-8α,12-olide-15α-oic acid (2), along with 22 known compounds (3–24), of which compounds 3–13 were isolated from this plant for the first time. The structures of these compounds were established on the basis of spectroscopic methods. Compounds 1–2 were evaluated for their in vitro anti-proliferative activities against Hep-G2 and MCF-7 tumour cell lines. Compound 1 exhibited strong inhibitory activity against Hep-G2 cell growth, in contrast to moderate cytotoxic activity against MCF-7 cells. Although compound 2 showed strong inhibitory activity against MCF-7 cell growth, it appeared to have modest activity against Hep-G2 cells.

Abstract:Chemical investigation of the roots of Ligularia sagitta has resulted in the characterization of six eremophilenolides 60,80-dimethoxy-10beta-hydroxyeremophil-7(11)-en-12,8alpha-olide (1), 6beta-angeloyloxy-10beta-hydroxy-8beta-methoxyeremophil-7(11)-en-12,8alpha- olide (2), 6beta-(2'-methylbutanoyloxy)-10beta-hydroxy-8beta-methoxyeremophil-7(11)-en-12,8alpha-olide (3), 6beta-angeloyloxy-10beta-hydroxy-8alpha-methoxyeremophil-7(11)-en-12,8alpha-olide (4), 6beta-(2'-methylbutanoyloxy)-10beta-hydroxy-8alpha-methoxyeremophil-7(11)-en- 12,8beta-olide (5) and 8beta,10beta-dihydroxy-6beta-methoxyeremophil-7(11)-en-12,8alpha-olide (6), together with one monoterpene (3R,4R,6S)-3,6-dihydroxy-1-menthene (7), two triterpenes lupeol (8) and ursolic acid (9), and beta-sitosterol (10). The structures of five new constituents (1-5) were elucidated by spectroscopic methods including 2D-NMR experiments. The compounds 1, 5 and 7 showed antibacterial activity by being assayed against Staphylococcus aureus, Bacillus subtilis and Escherichia coli.

Their structures were established as 3beta-angeloyloxy-8beta H-eremophil-7(11)-ene-12,8alpha (14beta, 6alpha)-diolide, 3beta-angeloyloxy-8beta-hydroxyeremophil-7(11)-ene-12,8alpha(14beta,6alpha)-diolide,3beta-angeloyloxy-8beta-methoxyeremophil- 7(11)-ene-12,8alpha(14beta,6alpha)-diolide,3beta-angeloyloxy-8beta-ethoxyeremophil-7(11)-ene-12, 8alpha (14beta,6alpha)-diolide, 3beta-angeloyloxy-10beta- hydroxyeremophil-8(9),7(11)-diene-12,8(14beta,6alpha)-diolide, 3beta-angeloyloxy-8,12-expoy-12alpha-hydroxy-8beta-methoxyeremophil-7(11)-en-14beta,6alpha-olide and 3beta-angeloyloxyeremophilan-7,11-dien-14beta,6alpha-olide, by means of spectroscopic analyses. Moreover, application of a photooxygenation reaction on 7 resulted in the generation of 2 with an alpha,beta-unsaturated gamma-lactone moiety. Abstract:A new sesquiterpene, 7α-H-11-hydroxyeremophil-1(10)-en-2-one (1), together with 14 known sesquiterpenoids (2–15), were isolated from the extract of the whole plant of Ligularia fischeri. Among them, compounds 2–6 and 15 were isolated from Ligularia fischeri for the first time. The new compound was characterized by means of spectroscopic methods including 1D and 2D NMR and HR-ESI-MS, and the known ones were established by comparing their NMR data with those of the corresponding compounds in the literature.

Abstract:From the whole plants of Ligularia duciformis, four new sesquiterpenoids, 3 beta-acetoxy-6 beta-methoxyeremophila-7(11),9(10)-dien-12,8 beta-olide (1), 3 beta-acetoxy-8 alpha-hydroxy-6 beta-methoxyeremophila-7(11),9(10)-dien-12,8 beta-olide (2), 3 beta-acetoxy-10 beta-hydroxy-6 beta,8 beta-dimethoxyeremophil-7(11)-en-12,8 alpha-olide (3), and 3 beta-acetoxy-6 beta,8 beta,10 beta-trihydroxyeremophil-7(11)-en-12,8 alpha-olide (4) were isolated. Their structures were established by high-field NMR techniques ((1)H,(1)H-COSY, (13)C-APT, HMQC, HMBC, and NOESY) and HR-ESI-MS analysis, together with comparison of the spectroscopic data with those of structurally related compounds. In addition, the cytotoxicity of the new compounds against human hepatic cancer cells Bel-7402, human pneumonic cancer cells A-549, and human colonic cancer cells HCT-8 were evaluated, the new compounds showed no cytotoxicity against the three tumor cells (all IC(50) values > 200 mu M).

Abstract:Four new eremophilenolides, 1-4, have been isolated from the whole plant of Ligulariopsis shichuana, Their structures were elucidated by spectroscopic methods, including 2D NMR experiments. Structures of compounds 1 and 2 were unequivocally established by X-ray diffraction experiments. All of these sesquiterpenes have eremophilane skeletons with 7(11)-en-8(12) lactone units. Compounds I and 2 showed moderate antibacterial activities against E. coli and B. subtilis.
