The establishment of the models of brain injury in rat brain slices and the novel method of quantitative analysis

Abstract:AIM To establish the models of different kinds of brain injuries and a simple, sensitive, rapid method of quantitative analysis. METHODS The rat cortical and hippocampal slices were prepared and the tissue activities were evaluated by the population spikes in the CA1 region. Brain slices were damaged by oxygen and glucose deprivation (OGD), glutamate, hydrogen peroxide (H 2O2) injury, and pretreated with ketamine, D-APs, propofol when subjected to these three damages respectively, then incubated in 2, 3, 5-triphenyltetrazolium chloride (TTC). The red formazan was extracted by solvent extraction and measured by ELISA reader. Lactic dehydrogenase (LDH) effluent rate in the incubated supernate were also measured. RESULTS The population spikes of the CA1 region in hippocampal slices could be recorded. Progressive prolongation of OGD injury resulted in decreased TTC staining in the cortical and hippocampal slices. There was a parallel increase in LDH effluent rate. The brain injury percentage had a positive correlation with the LDH effluent rate: cortical slices r = 0. 9609, P<0. 01; hippocampal slices r = 0. 8921, P<0. 05. Compared with the control groups, the TTC staining was decreased when slices were incubated in 1 mmol · L-1 glutamate and 2 mmol · L-1 H2O2. The decreases of TTC staining induced by 10 min of OGD injury, glutamate and H 2O2 injury were significantly attenuated when pretreated with ketamine 5 μmol · L-1 D-AP5 50 μmol · L-1 or propofol 5 μmol · L-1 respectively. CONCLUSION The models OGD, glutamate, H2O2 injuries in rat brain slices and the method of quantitative analysis as TTC staining, solvent extraction, and colorimetry with ELISA reader can be objectively, rapidly, and sensitively used in the studies of brain ischemia and drug evaluation.
