Effects of amyloid-beta protein injected into Meynert nucleus on memory and the expression of choline acetyltransferase and somatostain in rat

Abstract:Objective: To investigate the effects of amyloid-beta protein injected into Meynert nucleus on memory ability and the expression of choline acetyltransferase (ChAT) and somatostain (SOM) immunoreactive positive neurons. Methods: The Alzheimer disease (AD) rat model was established by injecting amyloid-beta protein (Aβ1-40, 10μg) into right Meynert nucleus in rat. Results: Learning and memory disorder appeared in the experimental group at 1 week after Aβ infection, aggravated progressively at 4 week. ChAT and SOM immunoreactive positive neurons distributed abundantly in Meynert nucleus, hippocampus and cortex fields in the control group and sham-operation group. However, ChAT and SOM immunoreactive positive neurons were obviously reduced in the experimental group, and remarkablely decreased at 4 weeks. Conclusion: Aβ injected into Meynert nucleus could induce persistent learning and memory disorder, and the decreased ChAT and SOM neurons induced by Aβ may be one of important factors for memory disorder and AD formation.
