Progesterone regulates TGF-β1, β2 expression in MG-63 cells

Abstract:Aim. To characterize the effects of progesterone (P) on the expression of transforming growth factors-beta (TGF-β1, -β2) in MG-63 cells so as to prove the hypothetical mechanism of progesterone in treating postmenopausal osteoporosis. Methods. MG-63 cells were cultured by MEN culture medium without phenolsulfonphthalein. Alkaline phosphatase(ALP) activity and content of osteocalcin (BGP) were assayed in MG-63 cells cultures. ELISA were used to show the intervention of P on the expression of TGF-β 1, -β2 protein production in MG-63 cells cultures. Results. 12-24 h after treatment, progesterone at 1 × 10-9mol/L increased TGF-β1, β 2 protein production in cultures of MG-63 cells, and treatment with increasing dose of progesterone caused a dose-dependent increase in the expression of TGF-β1, β2 protein by MG-63 cell (t = 10.232 - 109.454, P < 0.001). Conclusion. The data suggest that a direct action of progesterone on osteoblasts may facilitate cellproliferation and differentiation, increase osteoblast function and bone matrix synthesis, and accelerate bone formation mediated by up-regulated TGF-β1, β2 production.
